Lampiran 1. Analisis konsentrasi asam lemak (AOCS official method Ce 1-62 1990) Sebanyak ± 25 mg sampel ditimbang secara tepat, kemudian ditambahkan
standar internal (C17 : 0) sebanyak 0.5 mg. Ke dalam campuran ditambahkan 1.5
ml NaOH metanolik 0.5 N, kemudian tabung diisi dengan gas N2, ditutup rapat,
dikocok dan dipanaskan dalam penangas air suhu 100 0C selama 5 menit. Setelah
tabung didinginkan, ke dalam campuran ditambahkan 2 ml BF3-metanol (14% b/v),
tabung diisi dengan gas N2 ditutup dan tabung divorteks selama 30 detik. Ke dalam
tabung segera ditambahkan larutan NaCl jenuh sebanyak 5 ml, diisi dengan gas N2
ditutup dan dikocok. Lapisan heksana dipisahkan dan dipindahkan ke dalam vial.
Fase metanol/air diekstrak kembali dengan 1 ml heksana dan hasil ekstraksi
digabung serta dilakukan penyaringan dengan Na2SO4 anhidrid. Sampel methil
ester asam lemak dikonsentratkan dengan hembusan gas N2 hingga volume ± 1 ml.
Sampel siap diinjeksikanke alat kromatografi gas. Kondisi pemisahan asam lemak
adalah suhu injektor 250 0C, suhu detektor 260 0C, suhu kolom awal 140 0C yang
dipertahankan selama 6menit, peningkatan suhu kolom kemudian adalah 3 0C per
menit sehingga suhu akhir 230 0C dan dipertahankan selama 20 menit. Gas
pembawa digunakan helium dengan tekanan 1 kg/cm2, sedangkan tekanan gas
Hidrogen dan udara masing-masing adalah 0.5 kg/cm2. Identifikasi dilakukan
dengan menggunakan standar methil ester asam lemak dan kuantitasi dilakukan
dengan perbandingan standar internal.

Sampel organ diambil dan diawetkan
Sampel dipotong dengan ketebalan ± 3mm Dan difiksasi BNF 10%
Organ kembali dipotong dengan ukuran panjang ± 4mm,lebar 3mm dan tebal 3mm
Pengambilan air dari jaringan (dehidrasi): Organ-organ didehidrasi dengan alkohol konsentrasi bertingkat mulai dar 70%, 80%,
90%, 95% absolut 1 dan absolut 2. Masing-masing konsentrasi membutuhkan waktu ± 2 menit
Tahap penjernihan : Organ dimasukan ke dalam xylol selama 30 menit
Penanaman dalam parafin (embedding dan blok) : Organ-organ tersebut ditanam ke dalam parafin cair dalam lemari pemanas selama
2 jam dan kemudian organ-organ dimasukan ke dalam blok pencetak
Blok parafin dibekukan kemudian disimpan dalam lemari es
Blok dipotong dengan menggunakan mikrotom dengan ketebalan 5 µm (agar hasil potongan tidak mengkerut maka potongan diletakan diatas permukaan air bersuhu
45 oC), hasil potongan kemudian diangkat dengan gelas objek.
Gelas objek tersebut disimpan dan dikeringkan ke dalam inkubator dengan suhu 60 oC selama satu malam

Pewarnaan hematoksilin, dilakukan dengan memasukan sediaan ke dalam zat
warna hematoksilin selama 2 – 3 menit
Sebelum dimasukkan ke dalam pewarnaan eosin harus dibilas terlebih dahulu dengan air mengalir, demikian pula setelah pewarnaan eosin sediaan kembali
dibilas dengan air
Sediaan dimasukkan ke dalam alkohol konsentrasi bertingkat
dan alkohol absolut 1 dan 2
Penjernihan : proses ini dilakukan dengan memasukan sediaan ke dalam xylol 1 dan
xylol 2 selama beberapa menit
Lalu dikeringkan di udara terbuka agar sediaan terhindar dari pengaruh lingkungan
Kemudian ditutup dengan cover glass yang direkatkan dengan zat perekat entelen

Pemeriksaan mikroskopik dilakukan dengan pembesaran 400 x
