สมมตฐานของงานวจย

โรคโลหตจางเกดข นในผทมภาวะขาดธาตเหลก สตรต งครรภและผปวยโรคไตวายเร อรงระยะสดทาย รกษาไดโดยการใหรบประทานยาธาตเหลก (รปแบบสารเคม) แบบชนดเมดหรอฉดสารละลายเขารางกาย หากปรมาณทใหมากเกนไปกสามารถกอใหเกดโทษไดเนองจากธาตเหลกมคณสมบตเปนสารโปรออกซแดนทเรงการสรางอนมลอสระทเปนพษตอสารชวโมเลกลในเซลลและรางกาย เหลกในขาวสนเหลกเกดข นในรปสารประกอบเชงซอน/จบตรงกบสารพฤกษาเคมทถกรางกายนาไปใชไดงาย และไมมความเปนพษเมออยโดยลาพง

Sub-chronic toxicity test in rats: OECD Guidelines

Rat divided into 6 groups

1) DI 1 ml/day =>90 days

2) Iron rice (low Fe dose) => 90 days

3) Iron rice (medium Fe dose) => 90 days

4) Iron rice (high Fe dose) => 90 days

5) DI 1 ml/day: Satellite => 90 + 28 days

6) Iron rice (high Fe dose): Satellite => 90 + 28 days

Blood collection - Hematological markers: CBC

- Biochemical markers: FBS, BUN,

CRE, AST, ALT, ALP, TP, Alb, TBIL,TC

Wistar rats were assessed for sub-chronic oral toxicity during GSO treatment following the OECD Guidelines 40 (1998). The animals were randomly divided into 4 groups (5 male and 5 female each) and fed with deionized water (DI) as a control and iron rice (low, medium and high dose) for 90 days. A recovery (satellite) group is a non-dosing group that follows the main dosing phase of a study and extends the study for an additional 28 days. The satellite groups are included in nonclinical toxicity studies to understand whether toxicities observed at the end of the dosing phase are partially or completely reversible and inform the persistence or reversibility of effects. Blood was collected for analysis of hematological parameters using Coulter automated blood cell analyzer according to the manufacturer’s instruction, and biochemical markers using Randox automated analyzer according to the manufacturer’s instruction. At the end of the study, vital organs were collected for biochemical analysis and histopathology examination.
