Monoclonal Antibody Against Nsp2 Proteinof the Highly Pathogenic Porcine Reproductive
and Respiratory Syndrome Virus
Antigen Used for Immunization
Recombinant Nsp2 produced in Escherichia coli.
Method of Immunization
Six to eight-week-old BALB/c mice were immunized threetimes at 2-week intervals with 100mg of the pET28a-Nsp2protein.
Parental Cell Line Used for Fusion
Splenocytes from the BALB/c mice immunized with Nsp2protein were fused with mouse myeloma cells (SP2/0) ac-cording to conventional hybridoma technology.
Selection and Cloning Procedure
Hybridoma cells were grown in HAT selection medium. Po-sitive clones were subcloned by limiting dilution and screenedby ELISA analysis.
Heavy and Light Chains of Immunoglobulin
The MAb belonged to the IgG1subtype, and the light chainwas k type.
Specificity
The specificity was determined by Western blot and IFA an-alyses. The results clearly showed that this MAb could rec-ognize and bind to the Nsp2 specifically.
