INTRODUCTION
The immune response is very important in
our body, is regulated by lymphocytes, in
this case, monocytes, macrophages.
In this presentation we're gonna talk about
the immune response by monocyte FcyR
receptor. For this you need to know some
importants concepts .
INTRODUCTION PHAGOCYTOSIS: It is a
mechanism through which
some cells (neutrophils and macrophages) aroundwith
a plasma membrane antigen and introduced into the cell.
•LyGDI
Binding Proteins
INTRODUCTION
SHIP PROTEIN: Inositol phosphatases are
enzymes that influence multiple signaling
pathways. are also enzymes that
regulate phagocytosis.
FCɣR: Receptor mediator; binds ligands
and is linked with the ability inm
unofagococitica effector cells such
as macrophages and neutrophils.
INTRODUCTION
OBJETIVE
Knowing the effect of protein LyGDI
linked to the phosphatase SHIP in the
regulation of phagocytic receptor
response FCyR.

REACTIVOS:
Anticuerpos: FCɣR

ACTINA
SHIP
LyGDI
FUNCION: BLOQUEO DE ACTIVIDAD.
METODOLOGÍA
INMUNOPRECIPITACIÓN
Técnica que utiliza anticuerpos específicos
para una proteína para quitar esas proteínas
de la solución.
METODOLOGÍAWESTERN BLOT
Técnica usada para detectar proteínas
especificas en una muestra
determinada, para separar proteínas
(peso molecular y estructura).
PÉPTIDOS PULL-DOWN
Mezcla completa de proteínas.
El péptido pull-down es una prueba
imparcial para identificar lectores de
proteínas que se unen a péptidos
específicos modificado por una mezcla
de proteínas complejas.
METODOLOGÍA
ENSAYO COMPETENCIA
Método en el que se utilizan dos
células, en la cual una es
estimulada, pero la otra no recibe
estimulo ambas en presencia de un
mismo receptor.
METODOLOGÍA
CITOMETRÍA DE FLUJO
Técnica que proporciona resultados que
son aplicables en un diagnostico de
forma rápida.
Gran especificidad para detectar una
población celular, en una muestra de
varias poblaciones celulares.
Ofrece información simultanea de cada
célula analizada.
METODOLOGÍA
TRANSFECCIONES
Introducción de material genético
externo en células eucariotas mediante
vectores víricos.
Se realiza abriendo poros en la
membrana plasmática mediante
electroporacion.
METODOLOGÍA
MICROSCOPIA DE FLUORESCENCIA
Microscopio de fluorescencia es una
variación del microscopio de luz
ultravioleta.
Se usa para detectar sustancias con
autofluorescencia o sustancias
marcadas con fluocromos.
METODOLOGÍA
DIGE ELECTROFORESIS
Separación simultánea de máximo 3 muestras.
Controla perfil proteómico entre las células.
METODOLOGÍA
ENSAYO FAGOCITOSIS
IgG con PKH26 Se marcaron GR de
oveja, fueron analizadas por microscopia de
fluorescencia. El índice fagocítico se definió
como numero total de GR ingeridos por los
fagocitos.

DISCUSSION
AUTOR OPINION YES/NO
Damen JE et al. SHIP and SHIP-2 are important
regulatory phosphatases that
play distinct roles despite
similar catalytic activity, as
suggested
by in vivo studies.
YES
Allen LA et al. To identify as-yet undiscovered
proteins that uniquely associate
with SHIP or SHIP-2 we have
made use of DIGE analysis.
DIGE
is traditionally used to quantify
relative abundances and
monitor
differences in expression
profiles of proteins in a given
sample.

Koch A, Mancini A et al Both SHIP and SHIP-2 are
recruited to phagocytic cups
upon
FccR clustering and are known
to influence actin dynamics in-
directly through SHIP’s catalytic
activity.

CONCLUSIONS
Understand these interactions are
important to know about autoimmune
diseases.
The immune system is highly
regulated, this may cause deficiencies
and bad effects as self-destruction of
healthy tissue.
CONCLUSIONS
It is important that the human mind different
types of response against infections and
other, necessarily fails because there are
others.
They should conduct studies
to determine and eliminate the receptors
that cause autoimmune
diseases, developing methods for
total eradication.
