SHIMADZU FOR VITAMIN C

1. Please sign in the logbook placed near the instrument.

2. Switch the computer

3. Switch on the uflc,no . 3 (makesure to switch on from the detector until

the controller.

4. Double click on Lc solution.

5. Double click anylisis 1 and click ok

6. Turn the knob on uflc a quarter and press purge.The purging process

will take 10 minutes.

7. Once the purging finished,turn the knob back and press pump.

8. After 15 minutes,on coulechem cells,click nextrunedit method

PERFORMING ANALYSIS1.1 PERFORMING SINGLE ANALYSIS STANDARD / SAMPLE

1. Click < single run >button

2. Key in sample ID, method file, data files ,vial ,volume injection

3. Click ON START(single start)

4. Wait until anylisis completed (all peaks detected)

1.2 PERFORMING MULTIPLE ANALYSIS

1. Click the batch processing icon in the (Lc Data) assistant bar

2. Go to on the menu bar,select new batch file

3. Click the wizad icon in the Lc Data,

4. Insert the simple ID and data file name> tick auto increment>

5. Go to file>

6. Click batch start in the Lc data assistant bar.

HOW TO MAKE CALIBRATION CURVE

1.1 STANDARD

1. Click the Lc data analysis icon in the postrun assistant bar

2. Click standard,then click wizard (program).Choose the peak

3. Click the (integration off to on) button < ok >

4. Click next and then select retention time

5. Click quantitative method ,calibration level #6. Then click nextrename vitamin c

7. Put the standard concentration(depend on the concentration)

8. Highlight vitamin c and the concentration,then

9. Go to file,save data file < yes to all>

10. Apply to method,insert file name

11. Click the calibration icon in the postrun assistant bar, 12. Then drag and drop the data file selected in data explorer

13. Peak integrate all

1.2 ANALYSE SAMPLE

1. Go to file 2. Click sample,then go to file < choose the

calibration method>< open>
