Methods: This study combined an in vitro digestion, a lactase treatment and the Caco-2 cell model to examine quercetin and quercetin glucoside uptake from shallot and apple homogenates. Results: The in vitro digestion alone significantly decreased quercetin aglycone recovery from the shallot digestate (p < 0.05), but had no significant effect on quercetin-3-glucoside recovery (p > 0.05). Digestion increased the Caco-2 cell uptake of shallot quercetin-4'-glucoside by 2-fold when compared to the non-digested shallot. Despite the loss of quercetin from the digested shallot, the bioavailability of quercetin aglycone to the Caco-2 cells was the same in both the digested and non- digested shallot. Treatment with lactase increased quercetin recovery from the shallot digestate nearly 10-fold and decreased quercetin-4'-glucoside recovery by more than 100-fold (p < 0.05), but had no effect on quercetin recovery from apple digestates. Lactase treatment also increased shallot quercetin bioavailability to the Caco-2 cells approximately 14-fold, and decreased shallot quercetin- 4'-glucoside bioavailability 23-fold (p < 0.05). These Caco-2 cells had lactase activity similar to that expressed by a lactose intolerant human. Conclusions: The increase in quercetin uptake following treatment with lactase suggests that dietary supplementation with lactase may increase quercetin bioavailability in lactose intolerant humans. Combining the digestion, the lactase treatment and the Caco-2 cell culture model may provide a reliable in vitro model for examining flavonoid glucoside bioavailability from foods. A diet high in fruits and vegetables has been correlated with a reduced risk for both cancer and heart disease [1,2]. AbstractBackground: Quercetin and quercetin glycosides are widely consumed flavonoids found in manyfruits and vegetables. These compounds have a wide range of potential health benefits, andunderstanding the bioavailability of flavonoids from foods is becoming increasingly important.

Methods: This study combined an in vitro digestion, a lactase treatment and the Caco-2 cell modelto examine quercetin and quercetin glucoside uptake from shallot and apple homogenates.

Results: The in vitro digestion alone significantly decreased quercetin aglycone recovery from theshallot digestate (p < 0.05), but had no significant effect on quercetin-3-glucoside recovery (p >0.05). Digestion increased the Caco-2 cell uptake of shallot quercetin-4'-glucoside by 2-fold whencompared to the non-digested shallot. Despite the loss of quercetin from the digested shallot, thebioavailability of quercetin aglycone to the Caco-2 cells was the same in both the digested and non-digested shallot. Treatment with lactase increased quercetin recovery from the shallot digestatenearly 10-fold and decreased quercetin-4'-glucoside recovery by more than 100-fold (p < 0.05), buthad no effect on quercetin recovery from apple digestates. Lactase treatment also increased shallotquercetin bioavailability to the Caco-2 cells approximately 14-fold, and decreased shallot quercetin-4'-glucoside bioavailability 23-fold (p < 0.05). These Caco-2 cells had lactase activity similar to thatexpressed by a lactose intolerant human.

Conclusions: The increase in quercetin uptake following treatment with lactase suggests thatdietary supplementation with lactase may increase quercetin bioavailability in lactose intoleranthumans. Combining the digestion, the lactase treatment and the Caco-2 cell culture model mayprovide a reliable in vitro model for examining flavonoid glucoside bioavailability from foods.

BackgroundCardiovascular disease and cancer are the two most com-mon causes of death in the United States and most indus-trialized nations. A diet high in fruits and vegetables hasbeen correlated with a reduced risk for both cancer andheart disease [1,2]. It is thought that the phytochemicals

found in fruits and vegetables may be responsible in partfor these health benefits [3]. Phytochemicals from fruitsand vegetables may inhibit cell proliferation, protectagainst oxidative stress, influence cell-signaling pathways,and reduce inflammation. Because these compoundsappear to have such beneficial effects, interest has been

raised in examining the bioavailability of thesecompounds.

A phytochemical of particular interest is quercetin, astrong antioxidant that is widely consumed in many fruitsand vegetables. Quercetin has potential protective effectsagainst both cancer and heart disease. Briefly, quercetinhas been found to down regulate expression of mutantp53 in breast cancer cells, arrest human leukemic T-cellsin G1, inhibit tyrosine kinase, and inhibit heat shock pro-teins [4]. Quercetin has been shown to decrease lipid per-oxidation, inhibit cell proliferation, induce apoptosis,and inhibit platelet aggregation [5-8]. Because quercetinexhibits such a wide array of positive health effects, it isespecially important to understand quercetin bioavaila-bility from whole foods. Two widely consumed, goodfood sources of quercetin are apples and onions [9-13]. Inmost foods, quercetin does not exist in the aglycone form,but is, instead, conjugated. The type of sugar moiety towhich quercetin is bound affects quercetin bioavailability.For example, quercetin in the apple is bound mainly togalactosides, rhamnosides, and arabinosides, and thesequercetin conjugates are not well absorbed by the smallintestine. The onion contains mainly quercetin gluco-sides, which are well absorbed by the small intestine [14].

More work is needed to understand the bioavailability ofquercetin and other flavonoids from foods. The Caco-2cell culture model is a well-established in vitro technique,extensively used to study intestinal cell absorption ofcompounds such as pharmaceuticals and nutrients; it isan excellent in vitro tool to study bioavailability of spe-cific compounds. We have previously used the Caco-2 cellculture model to examine the uptake of quercetin fromapple and onion extracts [15]. Using this model, we foundthat absorbed quercetin from onion extracts was signifi-cantly greater than from apple extracts, as expected [15].Others have used the Caco-2 cell culture model to evalu-ate cell transport and/or accumulation of pure phyto-chemicals such as quercetin, quercetin glucosides,chrysin, flavone, epicatechin, proanthocyanidin, and car-otenoids [16-23].

To further understand quercetin bioavailability, it isimportant to examine the effects of digestion on foodsprior to intestinal uptake. An in vitro digestion has beenpaired successfully with the Caco-2 cell culture model tostudy iron and carotenoid bioavailability [23,24]. Use ofthe in vitro digestion with the Caco-2 cell culture modelcould be quite useful in more specifically analyzing quer-cetin bioavailability from foods. At this time there is littleinformation available describing the effects of digestionon flavonoids from foods. Vallejo et al. [25] found thatover 80% of total flavonoids were lost during an in vitrodigestion of broccoli. In a study of ileostomy patients,

Walle et al. estimated that the intestine might absorb 65–81% of major forms of dietary flavonoids after enzymatichydrolysis [26].

A good in vitro model would aid in evaluating bioavaila-bility of phytochemicals from foods by offering a simplemethod to screen for factors that may affect intestinalabsorption of quercetin and quercetin glucosides, such asthe food matrix, food processing, digestion, and interac-tions with other foods. Human and animal models can beexpensive and time consuming, while a cell culture modelallows for rapid, inexpensive screenings. The Caco-2model has the potential to be a good model to measurequercetin absorption, however there are some drawbacks.Caco-2 cells have been shown to express significantly lesslactase phlorizin hydrolase (LPH) than the averagehuman small intestine [27]. Since this enzyme is mostlikely responsible for the first step in the metabolism ofquercetin glucosides [28], this deficiency would clearlylimit the ability of the Caco-2 cells to metabolize andabsorb quercetin from quercetin glucosides.

Caco-2 cells used in our lab have expressed greater LPHactivity (3 mU/mg protein) than other Caco-2 cells (0.3mU/mg protein) [15], resulting in lactase activity similarto that expressed by enterocytes from a lactose intoleranthuman (2–10 mU/mg protein)[27]. The compound for-skolin induced lactase phlorizin hydrolase activity four-fold in Caco-2 cells [29]. In weanling rats, lactose con-sumption increased lactase activity in the jejunum bythree-fold [30]. Thus, we hypothesized that treating Caco-2 cells with either forskolin or lactase may raise lactaseexpression to rates comparable to humans, making theCaco-2 cell model a more valid model for screening quer-cetin glucoside bioavailability from food. If lactase activ-ity cannot be induced in Caco-2 cells, treating foodsamples with lactase following the digestion procedureand prior to cell bioavailability assays may give morecomparable results to humans.

The objectives of this study were (1) to develop an opti-mized in vitro digestion method for examining quercetinand quercetin glucoside recovery from digestates usingonions and apples; (2) to examine the effect of lactase onshallot digestates; and (3) to examine Caco-2 cellularuptake of quercetin and quercetin glucosides fromdigested and lactase treated shallot.

MethodsChemicals and materialsShallots and onions (Northern Yellow) were obtainedfrom a local grocery store. Apples (Red Delicious andCortland varieties) were obtained from the CornellOrchards (Cornell University, Ithaca, NY). Porcine pep-sin, bile extract, pancreatin, lactase (beta-galactosidase,

from Kluyveromyces lactis, activity of 3000 units/mL), quer-cetin, and quercetin-4'-glucoside were purchased fromSigma Chemical Company (St. Louis, MO). Quercetin-3-glucoside was purchased from Indofine Chemical Com-pany, Inc (Hillsborough, NJ). Caco-2 cells were obtainedfrom the American Type Culture Collection (Rockville,MD) and were cultured in Dulbecco's Modified EagleMedium (DMEM; Gibco Life Technologies, Grand Island,NY) supplemented with 5% fetal bovine serum (GibcoLife Technologies, NY), 10 mM HEPES, 50 units/mL pen-icillin, 50 µg/mL streptomycin, and 100 µg/mL gen-tamicin, and were maintained at 37°C in 5% CO2.

In vitro digestionTwo hundred grams of each food sample were chopped,blended for 5 min with 200 mL saline (140 mM NaCl, 5mM KCl) using a Waring blender, and then homogenizedusing a Virtis 45 homogenizer. The total homogenateswere aliquotted in 15 mL centrifuge tubes and stored at -20°C until use.

For the digestion treatment, 2 g aliquots of the food sam-ple were placed in a centrifuge tube with an equal amountof saline. The pH was decreased to 2.0 by drop-wise addi-tion of 1M HCl, and porcine pepsin was added to a finalconcentration of 1.3 mg/mL. The digestate was incubatedin a shaking water bath at 37°C for 30 minutes. The pH ofthe digestate was then increased to 5.8 with the drop-wiseaddition of 1M NaHCO3. Porcine bile extract and pancre-atin were added to a final concentration of 1.1 and 0.175mg/mL, respectively. The pH was increased to 6.5 by drop-wise addition of 1M NaHCO3, and the samples were incu-bated for 1 hour in a water bath at 37°C. Following diges-tion the pH was decreased to 2 by addition of HCl and thedigestates were stored at -80°C for further analysis.

To examine and optimize the effects of digestion time andpH on the recovery of compounds from the digestate, theabove parameters were varied. To examine the effects ofpepsin digestion time, the pepsin digestates were incu-bated for 0, 30, 60 or 90 minutes and then incubated withthe intestinal digestion enzymes for 60 minutes. To exam-ine the effects of intestinal digestion, the samples wereincubated with pepsin for 30 minutes, then incubatedwith pancreatin and bile for 0, 30, 60, or 90 minutes.

The effects of 100 µM ascorbic acid and a nitrogen envi-ronment on quercetin and quercetin glucoside recoveryfrom onion and apple digestates were examined. Follow-ing homogenization and prior to digestion, the food sam-ples were mixed 1:1 with saline containing 200 µMascorbic acid, leaving a final sample concentration of 100µM ascorbic acid. During the digestion proceduredescribed above, the samples were flushed constantlywith nitrogen.

The effect of pH of either 6.5 or 7.0 during intestinal diges-tion on quercetin and quercetin-3-glucoside recovery wascompared. The effect of the storage pH was examined bycomparing digested samples having either a final pH of2.0 or 6.5. The effect of storage pH on 20 µM pure querce-tin and 20 µM quercetin-3-glucoside was examined bycomparing recoveries from samples stored at pH = 2.0,3.5, 5.0 or 7.0. All samples were stored overnight at -80°C. Prior to HPLC analysis samples were thawed andextracted 4 times with acidified ethyl acetate (pH 2.0),evaporated to dryness and reconstituted in 2 mL acidifiedmethanol (pH 2.0).

Lactase digestionDoses of lactase (0.5 units to 3000 units per gram sample)were applied to 1 gram shallot extract and incubated for15 minutes at 37°C. The final pH was brought to 2.0 andthe samples were stored at -80°C. The time kinetics wereexamined by incubating 1 gram shallot extract with 100units of lactase for 0, 15, 30, 60, 90, 120, 240, and 720minutes.

The effect of both lactase and digestion were examined bydigesting the samples with pepsin and pancreatin asdescribed above, then incubating the samples with 100units of lactase for 30 minutes at 37°C. Samples werestored overnight at -80°C.

Prior to HPLC analysis, all digestate samples were thawedand extracted 4 times with acidified ethyl acetate (pH 2.0),evaporated to dryness and reconstituted in 2 mLmethanol.

Uptake of quercetin-4'-glucoside and quercetin from shallot digestates by Caco-2 cellsCaco-2 cells were seeded at a density of 5 × 105 cells perwell in a collagen coated 6-well, flat bottom plate andincubated at 37°C in a 5% CO2 environment. Caco-2 cellswere used between passages #10–25, and the cells reachedconfluence approximately 5 days post seeding. Culturemedia was changed three times a week. On day 14 postseeding, the DMEM was removed and the cells were rinsedthree times with phosphate buffered saline (PBS).

Shallot homogenates were digested as previouslydescribed and were placed directly on the 14 day oldCaco-2 cells, or the samples were diluted 1:2 or 1:4 inHBSS (Hank's Balanced Salt Solution). Cells were alsoincubated with non-digested shallot homogenates forcomparison. For each treatment, two wells of cells wereused for each sample, and each treatment was repeated intriplicate.

To examine the effect of lactase on quercetin uptake fromshallots, shallots were digested then incubated with

lactase (50, 100, 300, and 1000 units/g shallot) for 20minutes at 37°C. The digested shallot homogenates andthe digested plus lactase treated shallot homogenates werediluted 1:2 in HBSS and placed on the cells. In all experi-ments, Caco-2 cells were incubated with treatment for 30minutes at 37°C in 5% CO2. The shallot treatment andHBSS was removed and the cells were rinsed three timeswith 20% methanol in PBS. Cells were scraped in acidifiedmethanol (pH = 2.0) and the wells were rinsed three timeswith methanol. The scraped cells were sonicated for 15minutes, centrifuged at 1600 g for 5 minutes, and themethanol supernatant was collected. The cells were rinsedthree more times with methanol, the supernatants werecollected and the methanol extracts were evaporated todryness under nitrogen and reconstituted in 400 µl acidi-fied methanol for HPLC analysis.

Induction of lactase activity in Caco-2 cellsCaco-2 cells were seeded at a density of 5 × 105 cells perwell in a 6-well flat bottom plate. The cells were culturedin DMEM spiked with different doses of either lactose (10,50, 100, 500, and 1000 µM), or forskolin (1, 10, 50, 100,and 200 µM). Media was changed every two days, andcells were harvested and lactase activity was measured at14 days post-seeding. Lactase activity of Caco-2 cells wasmeasured using a method adapted from Dahlqvist [31].Cells were trypsinized, collected, centrifuged and resus-pended in homogenization buffer (50 mM sodium phos-phate; 1 mM EDTA; 10 mM dithiothreitol; proteaseinhibitor cocktail, Sigma Chemical Co., St. Louis, MO).Cells were homogenized 5 times for 30 seconds with 1minute of cooling between bursts using a benchtophomogenizer. Homogenates were treated with 56 mM lac-tose and incubated at 37°C for 60 minutes. Glucose oxi-dase, peroxidase, and o-dianisidine were applied to thecell homogenates and the final colored products weremeasured at 420 nm using a spectrophotometer [31]. Theresults were compared to a glucose standard curve todetermine the amount of glucose released by lactase in theCaco-2 cell monolayer. Protein was determined fromcrude cell homogenates colorimetrically using the Lowrymethod with comparisons to a bovine serum albuminstandard curve. Results are expressed as milliunits/mg ofprotein, and one unit is defined as the lactase activity thathydrolyzes 1 µmole of lactose per minute at 37°C.

HPLC analysisQuercetin and quercetin-3-glucoside content of untreatedhomogenates, digestates, and Caco-2 cell extracts weredetermined using an RP-HPLC procedure with a Supel-cosil LC-18-DB column (150 mm × 4.6 mm, and 3 µmpore size). Waters 515 HPLC pumps (Waters Corp., Mil-ford, MA) and a Waters 2487 dual wavelength absorbancedetector (Waters Corp., Milford, MA) set at 370 nm wereused for all HPLC analysis. Quercetin, quercetin-3-gluco-

side, and quercetin-4'-glucoside were used as standards.For the analysis of quercetin, quercetin-3-glucoside, andquercetin-4'-glucoside in the apple peel extracts, shallotextracts, and digestate extracts, the solvent system usedwas (A) acidified water (pH 2.0; triflouroacetic acid) and(B) acetonitrile. The gradient method was the following:0.0 min, flow rate = 1.4, (A) 90% and (B) 10%; 53 min,flow rate = 1.5, (A) 80% and (B) 20%; 58 min, flow rate =1.7, (A) 65% and (B) 35%; 64 min, flow rate = 1.4, (A)90% and (B) 10%. Twenty µL injections were made foreach sample. Quercetin, quercetin-3-glucoside, and quer-cetin-4'-glucoside concentrations in the apple peelextracts, shallot extracts, and in the digestates were extrap-olated from the pure quercetin and quercetin-3-glucosidestandard curves.

Statistical analysisAll data were reported as means ± SD for three replicatesof each treatment. An analysis of variance (ANOVA) wasused to compare results between treatment groups, andpairwise multiple comparisons were performed usingFisher's LSD with an individual error rate of 0.05. The sta-tistical analysis was completed using Minitab Release 12software (State College, PA).

ResultsIn vitro gastrointestinal digestionThe total pepsin digestion time and pancreatin/bile diges-tion time had little to no effect on recovery of both quer-cetin and quercetin-3-glucoside from apple and onionhomogenates when treated for up to 60 minutes (data notshown). After 90 minutes of pepsin digestion and 90 min-utes of pancreatic digestion, quercetin and quercetin 3-glucoside in both the apple and the onion decreasedslightly. Based on these results we chose to use a 30-minute pepsin digestion and a 60-minute pancreatin/biledigestion. In past studies, quercetin from onion and quer-cetin from quercetin-4-glucoside supplements reachedthe plasma in less than an hour following consumptionby human volunteers [32]. Therefore, long in vitro diges-tion times were not necessary to mimic human digestionof quercetin compounds from apples and onions.

The presence of ascorbic acid and nitrogen had no effecton quercetin or quercetin-3-glucoside recoveries from thedigestates (data not shown). Quercetin and quercetin-3-glucoside recoveries from digested samples treated withascorbic acid, nitrogen or both ascorbic acid and nitrogenwere not different from the recoveries from the untreateddigested samples.

The factor that had the greatest effect on recovery was pH(Figure 1A). Quercetin is less stable at higher pH, there-fore the effect of pH during intestinal digestion was exam-ined. Recoveries of quercetin and quercetin-3-glucoside

after intestinal digestion at pH 7.0, when compared to pH6.5, were not significantly different (p > 0.05). However,following overnight storage at -80°C, the samples storedat pH 2.0 had significantly greater quercetin and querce-tin-3-glucoside recoveries than samples stored at pH 6.5and 7.0 (p < 0.05). Pure quercetin and quercetin-3-gluco-side were also more stable at lower storage pH followingdigestion (Figure 1B). At pH 2.0, the recoveries for purequercetin and quercetin-3-glucoside were 74.8% and86.2% when compared to the control. At the highest pH(7.0), recoveries for quercetin and quercetin-3-glucosidewere 46.5% and 13.9%, respectively.

Based on these results, optimal digestion conditions weredecided to be: pepsin digestion at pH 2.0 for 30 minutes,pancreatin/bile digestion at pH 6.5 for 60 minutes, and afinal storage at pH 2.0. Ascorbic acid and nitrogen treat-ments were not continued. Using these conditions, theeffect of digestion on quercetin and quercetin-3-glucosiderecoveries from apples, onions, and pure quercetin andquercetin-3-glucoside was examined (Figure 2). Follow-ing digestion, recoveries of pure quercetin-3-glucoside,apple quercetin-3-glucoside, and onion quercetin-3-glu-coside were similar to recoveries from non-digested sam-ples (p > 0.05). digested apple, onion, and pure compound were 87.7,89.5, and 86.4%, respectively. Quercetin recovery was sig-nificantly reduced in digested pure quercetin and digestedonion samples, when compared to non-digested samples(p < 0.05). Quercetin recovery was lower than quercetin-3-glucoside recovery and tended to vary more, dependingon the food matrix: 52.5% and 74.3%, from the onionand pure compound, respectively. There was no signifi-cant difference in quercetin recovery between to the non-digested and digested apple homogenates. There was onlytrace or no quercetin in non-digested apple homogenates,so the appearance of any quercetin following digestionresulted in a net increase.

Uptake of quercetin-4'-glucoside and quercetin from shallot digestates by Caco-2 cellsQuercetin-4'-glucoside and quercetin were absorbed byCaco-2 cells following treatment with both digested shal-lot and non-digested shallot homogenates (Figure 3).Quercetin-3-glucoside was not detected in any sample.Quercetin-4'-glucoside uptake by the Caco-2 cellsincreased by approximately 2-fold following digestion (p< 0.05). Caco-2 cells treated with shallot homogenateabsorbed approximately 2.9 ± 0.65 nmol of quercetin-4-glucoside, and Caco-2 cells treated with digested shallotabsorbed 5.4 ± 0.04 nmol. non-digested homogenate (Figure 3B insert), howeverquercetin uptake from the digested samples was similar tothe non-digested samples (p > 0.05). Caco-2 cellsabsorbed 2.8 ± 0.4 nmol and 2.7 ± 0.2 nmol quercetinfrom the non-digested and digested shallot homogenates,respectively. Absorption of both quercetin-4-glucosideand quercetin from digested shallot followed a doseresponse. The Caco-2 cells absorbed quercetin 4'-gluco-side and quercetin incrementally less from the digestedsamples that were diluted 1:2 or 1:4 in HBSS.

Induction of lactase activity in Caco-2 cellsAddition of lactose and forskolin, a specific inducer oflactase [29], to Caco-2 cells did not significantly increaselactase activity of Caco-2 cells. The lactase activity of allcells ranged from 2–4 mU/mg protein in all treatments.

Lactase DigestionTreatment with 100 units lactase/g sample had a signifi-cant effect on both quercetin and quercetin-3-glucosiderecoveries from the shallot (p < 0.05; Figure 4). Quercetin

recovery from shallot digestates increased 5.5 fold, from47.5 ± 7.6 µg/g sample in the untreated digestate to 262.2± 17.6 µg/g sample in the lactase treated sample. Thelactase plus digestion treatment resulted in a non-signifi-cant decrease in quercetin recovery compared to thelactase only treated samples. Quercetin-3-glucoside fromshallot digestates also increased approximately 5 fold,from 17.3 ± 1.7 µg/g sample to 80.0 ± 10.3 µg/g samplefollowing the lactase treatment. The effect of lactase on theapple samples was not as great. Quercetin-3-glucosiderecovery decreased slightly, while changes in quercetinlevels were not significant (p > 0.05).

Because treating shallots with lactase increased quercetinrecovery so greatly without significantly decreasing quer-

cetin-3-glucoside recovery, the effect of lactase on querce-tin-4'-glucoside recovery was also examined. Morequercetin-4'-glucoside is found in shallots when com-pared to quercetin-3-glucoside. As the dose of lactaseincreased, quercetin-3-glucoside recovery increased from18.2 ± 3.7 up to 175.5 ± 48.1 µg/g shallot at the 1000 unitdose and then decreased to 60.2 ± 2.0 µg/g at the 3000unit dose (Figure 5A). As the dose of lactase increased,quercetin recovery increased and quercetin-4'-glucosidedecreased (Figure 5A). The increase in quercetin was quitedramatic. Recovery of quercetin from untreated shallotsamples was 93.7 ± 2.2 µg quercetin per gram sample, andat the highest lactase dose, recovery of quercetin fromshallot samples was 958.8 ± 76.1 µg quercetin per gramshallot. 518.2 ± 10.7 µg/g shallot from the untreated sample to 3.2± 0.8 µg/g shallot at the highest treatment dose. A similartrend was seen for all compounds in the kinetic experi-ment. As the incubation time increased, recoveries ofquercetin increased and quercetin-4'-glucoside decreased(Figure 5B). Quercetin-3-glucoside increased through twohours, and then decreased following four and eight hoursof incubation with lactase. In both the dose response andkinetic experiments, increases in quercetin recoveries weregreater than decreases in quercetin-3-glucoside or querce-tin-4'-glucoside recoveries.

Caco-2 cell uptake of quercetin and quercetin glucosides following lactase treatmentThe addition of lactase following the pepsin and pancrea-tin/bile digestion significantly increased the amount ofquercetin absorbed by the Caco-2 cells with a significantdecrease in the amount of quercetin-4'-glucosideabsorbed by the Caco-2 cells (p < 0.05, Figure 6). Querce-tin uptake increased from 0.98 ± 0.67 nmol from thedigested sample up to 14.1 ± 1.6 nmol from the digestedplus 1000 units lactase treated sample. Quercetin-4'-glu-coside uptake by Caco-2 cells decreased as the dose oflactase increased, however the increase in quercetin wasmore dramatic than the decrease in quercetin-4'-gluco-side. Quercetin-3-glucoside uptake was not detected.

DiscussionIn vitro digestionPreviously, our laboratory determined that our Caco-2cells had the potential to be used as a model to study quer-cetin bioavailability from onions and apples [15]. In thepresent study, we modified an in vitro digestion procedureand combined it with the Caco-2 cell model to give amore comprehensive examination of quercetin and quer-cetin glucoside bioavailability in Caco-2 cells.

The digestion procedure modified in these experimentsresulted in quercetin-3-glucoside recoveries that rangedfrom 78.9 to 89.5% and quercetin recoveries that ranged

from 47.3 to 74.3%. In both cases, the lowest digestaterecoveries were from the shallot. Interestingly, quercetinrecovery from both the onion and shallot was considera-bly lower than from the pure compound. This differenceis not yet explained, but could potentially be due tointeractions with other compounds found in the onionand shallot.

In all cases, quercetin recovery from digestates was lowerthan quercetin-3-glucoside. The glucoside moiety maylend stability to quercetin-3-glucoside during digestion,and could contribute to its greater bioavailability in vivo aswell. Quercetin aglycone may be more susceptible to

oxidation or other degradation during exposure to boththe digestive enzymes and the variations in pH in thestomach and intestine. Ascorbic acid and nitrogen, bothadded to help decrease oxidation, had no effect on quer-cetin or quercetin-3-glucoside recoveries from digestedonions.

The factor that had the greatest effect on both quercetinand quercetin-3-glucoside stability in the digestate waspH. Recoveries of digested pure compounds and digestedcompounds from the onion homogenates were signifi-cantly less if stored at a pH of 6.5 or 7.0 than if stored atpH 2.0 at -80°C (Figure 1). Vallejo et. al [25] used an invitro digestion method to measure the effect of digestionon a variety of compounds from broccoli. Following thepepsin and pancreatic digestion, they recovered only 16%of total flavonoids, and the main flavonoids found in thebroccoli were quercetin and kaempferol glycosides. Therecovery of quercetin and quercetin-3-glucosides, flavo-noids common to the foods in our study, was muchhigher than 16% from the digested shallot, onion andapple. It has been estimated that quercetin glucoside bio-availability may be as high as 80% in humans and querce-tin bioavailability may range from 35–53% [26,33]. Ourresults would appear to be more reasonable estimates, ifindeed, quercetin and quercetin glucoside bioavailabilitylies within the approximated ranges found by Walle et. al[26,33].

Effect of digestion on quercetin and quercetin glucoside uptake by Caco-2 cellsDigestion of the shallot resulted in decreased recoveries ofboth quercetin and quercetin-3-glucoside, therefore it wasexpected that digestion might decrease the bioavailabilityof these compounds as well. Following digestion, querce-tin aglycone in the shallot was decreased by approxi-mately 50%; however, quercetin bioavailability wasunchanged following digestion compared to the non-digested samples. This means that the digestion proceduremust degrade quercetin, and simultaneously enhances thebioavailability of quercetin, bringing the cellular uptakeback to comparable levels with the non-digested samples.Quercetin-4'-glucoside uptake by the Caco-2 cells fromthe shallot increased by approximately 2-fold followingthe in vitro digestion. We hypothesize that the digestionprocedure may have released more compounds from thefood matrix leaving them more available for uptake by theCaco-2 cells. The digestion procedure may also haveimproved solubility of the compounds, increasing theirabsorption by the Caco-2 cells.

Quercetin-3-glucoside was not detected in the Caco-2cells following treatment with shallot. Quercetin-3-gluco-side is a minor compound in the shallot, and it is believedthat the levels were below our detection limit. In the past,

we found that Caco-2 cells did absorb trace amounts ofquercetin-3-glucoside from shallot extracts [15]. In theprevious experiments, shallots were first extracted withethanol and ethyl acetate, and finally reconstituted andconcentrated in methanol. This procedure produced moreconcentrated shallot extracts for cell treatment than withour current procedure. In the current study, shallothomogenates and digestates were too dilute to detectsmall changes in initial concentrations or in cellularuptake of quercetin-3-glucoside, which was below thedetection limit.

Strong evidence suggests that quercetin glucosides aremore bioavailable in humans than the quercetin agly-cone, however it has not yet been determined why this isthe case. Based on the digestion data, quercetin glucosideis more stable following the in vitro digestion conditionsthan quercetin and is therefore more likely to reach theintestine intact. Quercetin-4'-glucoside bioavailability inCaco-2 cells was increased nearly 2-fold following diges-tion and quercetin absorption was not changed (Figure3). It has been hypothesized that absorbed intact querce-tin glucosides are then quickly hydrolyzed by cytosolic β-glucosidase to quercetin aglycone [34]. It has also beenhypothesized that the major pathway for quercetinglucoside absorption begins with hydrolysis by LPH.Deglycosylation of quercetin glucosides at the brush bor-der membrane positions the resulting aglycone in a primeposition for diffusion across the brush border. The degly-cosylation of the quercetin glucoside would result in ahigher concentration of aglycone at the apical enterocytemembrane and potentially increase the rate of absorption[35].

Effect of lactase on quercetin and quercetin-4'-glucoside uptake by Caco-2 cellsThe potential pathways for quercetin glucoside and quer-cetin metabolism and absorption can be seen in Figure 7.Quercetin aglycone passively diffuses across the apicalmembrane and is then glucuronidated. Evidence stronglysuggests that quercetin glucosides are first hydrolyzed bythe lactase site of lactase phlorizin hydrolase prior to dif-fusion across the apical membrane [36]. Quercetin gluco-sides may also be transported into the cell by the sodium-dependent glucose transporter1 (SGLT1) and thenhydrolyzed by the cytosolic beta-glucosidase. Quercetin-3-glucoside is not a good substrate for cytosolic beta-glu-cosidase [37]. Since research has shown that bothquercetin-3-glucoside and quercetin-4'-glucoside are sim-ilarly bioavailable in humans [14,38], this could be anindication that hydrolysis by LPH and the subsequentpassive diffusion of quercetin into the cell is the mainpathway for quercetin glucoside absorption across thebrush border. Following hydrolysis and incorporationinto the cells, quercetin aglycone is then glucuronidated.

Quercetin glucosides and possibly quercetin glucuronidesare then transported back into the lumen by multidrugresistance protein 2 (MRP2). Conjugated quercetinmetabolites also eventually reach circulation, but thetransporter involved in transporting them across the baso-lateral side is still unknown.

Since lactase is an important enzyme in the metabolismand subsequent absorption of quercetin glucosides,lactose intolerant individuals may have a reduced capabil-ity to hydrolyze quercetin glucoside for further absorptionacross the small intestinal wall. Many lactose intolerantpeople use commercial lactase to break down lactose. Notonly might this enzyme help improve digestibility of lac-tose, but it may also increase bioavailability of quercetin

from foods. Initial lactase treatments were applied both toshallot and apple homogenates and digestates. Lactasehad little to no effect on apple samples. Apples containquercetin bound mainly to galactosides, rhamnosides,and xylosides, conjugates that would not be readilyhydrolyzed by lactase. However, lactase treatment hadgreat effects on shallot and onion digestates. The shallotand onion are high in quercetin glucosides, mainly quer-cetin-4'-glucoside, quercetin-3-glucoside, and quercetin-3,4'-diglucoside, compounds readily hydrolyzed bylactase. Treatment with lactase in the range of 15 units/mgsample up to 1000 units/mg sample, significantlyincreased both quercetin and quercetin-3-glucoside recov-ery in shallot homogenates and digestates. The increase inquercetin-3-glucoside is most likely a result of

deglycosylation of quercetin-3,4'-diglucoside. Rhodes etal. [39] found that over time quercetin-3,4'-diglucoside inchopped onion will autolyze to monoglucosides, andwithin 24 hours the diglucoside will completely disap-pear. This may also explain the increase in quercetin-3-glucoside over time. Quercetin-4'-glucoside decreased fol-lowing treatment with lactase as expected. Results fromthis work suggest that quercetin-4'-glucoside is utilized bylactase prior to quercetin-3-glucoside. Interestingly, theincrease in total quercetin was greater than the decrease inquercetin-4'-glucoside. Digestion with lactase may releasequercetin from the food matrix as well, making it moreavailable for absorption.

Following hydrolysis of quercetin glycosides, it has beenhypothesized that quercetin is quickly glucuronidated,

and quercetin glucuronides are then found circulating inthe plasma [40]. In the current studies, these Caco-2 cellsshowed no signs of glucuronidating quercetin followingquercetin absorption, but do express some LPH activity aswas evident by the increased quercetin uptake from shal-lots and from the lactase activity assays [15]. Thus, theseCaco-2 cells have the potential to be a good model ofquercetin absorption, but not of further metabolism.

A good model of quercetin glucoside bioavailabilityshould incorporate lactase activity. The Caco-2 cells usedfor these experiments expressed lactase activity similar tothat of a lactose intolerant human (2–10 mU/mg pro-tein). These cells had approximate lactase activity ofbetween 2–4 mU/mg protein, consistent with what wepreviously reported [15]. Lactose tolerant humans tend to

have intestinal lactase activity that ranges from 20–80mU/mg protein [41]. Forskolin and lactose did not inducelactase activity in our Caco-2 cells. The lactase activity ofour cells was nearly 10 times higher than previouslyreported values for Caco-2 cells of 0.3 mU/mg protein oflactase activity [27], and it is quite possible that the lactaseenzyme in our Caco-2 cells is already expressed to the full-est extent.

Since lactase activity could not be increased in the Caco-2cells, we combined a lactase treatment with digestion toprovide an intestinal uptake model that is more compara-ble to a lactose tolerant human, or a lactose intoleranthuman ingesting a lactase digestive aid to help improvelactose digestion. Not only did lactase increase theamount of quercetin in digested shallot homogenates, butit also increased the amount of quercetin taken up by theCaco-2 cells from the digested shallot extracts. This sug-gests that a lactase containing digestive aid may increaseabsorption of quercetin from onions in lactose intoleranthumans. Combining the Caco-2 cell model with an invitro digestion, simulating stomach and small intestinaldigestion, and a lactase digestion may provide a more use-ful model to examine and screen for bioavailability of fla-vonoid glucosides from common foods (Figure 8).

ConclusionsFollowing an in vitro stomach and small intestinal diges-tion, the recovery of quercetin and quercetin-3-glucosideis optimized at a storage pH at or below 3.5. Storage at pHabove 3.5 results in loss of most of the compounds. Thein vitro digestion increased the uptake of shallot querce-tin-4'-glucoside by the Caco-2 cells. Quercetin uptake bythe Caco-2 cells was similar between digested and non-digested samples, despite the fact that approximately 50%of shallot quercetin is lost during digestion. Treatingshallot digestates with lactase increased the recovery ofquercetin aglycone 10-fold and decreased the recovery ofquercetin-4'-glucoside. Lactase treatment increased thebioavailability of quercetin aglycone 14-fold anddecreased the bioavailability of quercetin-4'-glucoside tothe Caco-2 cells. Combining pepsin, pancreatin/bile, andlactase digestions with the Caco-2 cell culture monolayermay results in a useful model for studying flavonoid bio-availability from foods.

Many advances have been made in understanding flavo-noid bioavailability, however many questions still remainunanswered. Food processing, interactions with othercompounds as well as with other foods are all factors thatmay affect bioavailability of flavonoids. A simple andinexpensive screening model would be beneficial as a firststep in examining many of these factors. The Caco-2 cellmodel has the potential to be a good model for analyzingintestinal uptake of quercetin, quercetin glucoside and

other flavonoids. While the Caco-2 cell model will neverbe an exact replica of a human small intestine, it couldprovide a valuable tool for initial screenings of large quan-tities of samples relatively quickly and inexpensively. Inthis study, it was found that a simple digestive aid such aslactase might increase quercetin bioavailability. This maybe of importance to lactose intolerant people. Suchsignificant trends may be examined in more detail usinghuman or animal models.
