Achievements in Research, Teaching and Extension
RESEARCH I) Honey bee-breeding and Bee-pathology :
Developed infra-red-photography technique which is accepted as a tool for conducting long term behaviouristic studies in honey-bee colonies internationally. The technique was used to study the natural defense behaviour of Apis mellifera-carnica against an ectoparasite mite, Varrora jacobsoni. These studies helped in solving previous suspicion and opening new vistas of bee breeding. a) First time gave photographic evidences regarding defensive abilities of Apis mellifera-
carnica against a serious pest Varrora jacobasoni. This damage was earlier presumed to be due to ants and other arthopod predators. Infra-red-video film was prepared showing defense reaction of A. melliferacarnica against Varrora jacobsoni.
b) Studies proved partial genetic control of Varroa defense traits by A. melliferacarnica,
because of colonies compositions of having different patrilines.
c) Did pioneering research by starting Bee breeding for Varroa resistance using excellently performing workers with respect to Varroa defense traits as drone layers. The semen of drones obtained from laying workers was used to instrumentally-inseminate the queens reared from Apis mellifera carnica being tested for high honey production and gentleness for several generations. The performance of the off springs was found
8
superior than control in responding to Varrora defense reaction. This revealed partial superiority of off springs against control carnica lines.
d) Repeatabilities of Varroa defense traits like uncapping, removing and grooming
behaviour were reported for the time.
e) Previously undescribed behaviour of queen opening Varroa parasitized cells was reported for the first time.
II) Queen Rearing and Artificial Insemination
i) In India for the first time standardized instrumental insemination technique in honey
bee queens. Attempted bee-breeding for higher honey production using exotic semen. The crossbreds were found to be more productive than existing stock.
ii) Investigations on factors influencing queen larva acceptance grafted for mass queen rearing in Apis mellifera L. revealed maximum acceptance (77.7 to 88.3%) of grafted larvae, 6-12 in number, was achieved by grafting 12.24 h old larvae in artificial queen cups. Most preferred queen cups were those prepared from wax of old combs. Best quality (heaviest) queen were produced during the month March-May and this period has been designated as a breeding period under mid-hill conditions of Himachal Pradesh.
III) Bee-Botany Wild flora namely Rubus ellipticus; Berberis lycium, Vitex negundo and Elaegnus umbellata were investigated for nectar secretion, nectar sugar production and nectar sugar concentration. Twenty-four hours sugar values were found to be 9.6, 0.72, 0.074 and 0.69 mg/flower for Rubus, berberis, Vitex and Elaegnus respectively. IV) Horticultural Entomology
Tested and recommended new miscible oils against San Jose Scale of apple in mid-hills and valley areas of Himachal Pradesh. Two new miscible oils Servo orchard spray oil (IOC) and Agro orchard oil (Homeland Specialty Oils Ltd.) at 2 and 3 per cent concentration were tested at three locations representing mid-hills and valley areas and efficacy were compared with recommended HPTSO-E at all sites. The spray was given at green tip stage. Both the oils were found equally effective at both the concentrations. The study suggested the Servo orchard spray oil, Agro spray orchard oil and HPTSO-E could be recommended at 2 per cent when sprayed at green tip stage for control of scale, but in the mid-hills and valley areas the left over population multiplies and required application of insecticides during season.
Field trials were conducted to test the efficiency of some insecticides against nymphal and adult stages of olive lace bug,Eteoneus stgilatus Drake and Poor. Fenvalerate 0.01%, endosulfan 0.05%, quinalphos 0.05%, oxydementon-methyl 0.025%, phosphamidon 0.03% and monocrotophos 0.04% were found effective in reducing the nymphal and adult populations of olive bug.
V) Comparison of jet black Carnica and golden ligustica for sexual maturity
9
Most of 5-13 days old drones of both carnica and ligustica remained non –evertive when, subjected to trapping of the abdomen or thorax pressing. All carnica dominated drones in the age group of 18and 22 days were prone to complete eversion while that of ligustica remained non-evertive at the age of 18days. The carnica dominated drones (18 and 22 days) yielded 1 to 2ul semen/drone while ligustica dominated gave 0.5 to 1.2 ul semen. Some of 13 days old drones of carnica dominance were found to expel negligible amount of semen without mucous. VI) Studies on Inter-specific queen rearing There was non acceptance of grafted larvae from either of species (A. cerana and A. mellifera) in the colony of another species. VIII) Selective breeding of honey bee, Apis mellifera L The findings of studies conducted on “Selection of honeybee A. mellifera L. for tolerance against T. clareae (Delfinado and Baker)” For selecting colonies, tolerant against T. clareae, data have been collected on variation in infestation level in different A. mellifera colonies and damage caused to the mites by bees. Three colonies (T3, T6 and T16) out of a stock of nineteen colonies exhibited maximum mite damage to the tune of 52.75, 49.08 and 47.65 per cent, respectively, during period of experiment. Overall degree of infestation was observed to be minimum in above colonies (0.96, 0.25 and 0.49%). Maximum mite damage during the year in above colonies proved to be the relevant natural defense mechanism against T. clareae which possibly helped in minimizing the overall infestation. The defense mechanism was considered as a tolerance trait of above colonies against T. clareae, hence sorted out for tolerance traits. Results revealed importance of mite damage as useful selection criteria like studies on Varroa jacobsoni in sorting out T. clareae tolerant lines. The mite count in the hive debris was an indicator of colony mite population being positively correlated to the brood area during different seasons (r=0.74). The mite population was maximum during summer and declined during low brood activity period i.e. monsoon and winters. Overall degree of infestation of bee colonies was observed to be maximum during June (4.76%) and no infestation was observed during January and February. Worker bees were found to be infested at a very low rate ranging between 0-0.63 per cent. Hygienic behaviour tests are important for selecting honeybee colonies tolerant to T. clareae. Testing of the three selected colonies for hygienic behaviour revealed quick removal (within 24 hrs.) of pin killed brood to the extent of 99.00, 97.50 and 95.00 per cent, respectively. The susceptible colony (T18S) which had least mite damage and maximum infestation during period under study responded for the removal of pin killed brood to the extent of 88.50 per cent in 24 hours. Similarly the tolerant colonies removed freeze killed brood to the extent of 100, 99.62, 99.37 per cent in 24 hours being significantly higher than susceptible one (95.75%). The selected colonies also responded positively in uncapping and removing the artificially mite infested brood to the extent of 24.36 per cent (1 live mite/cell) and 37.80 per cent (2 live mites/cell) as compared to removal response of dead mites and uncapping of cells given simple cuts. Significantly more uncapping and removal of artificially mite infested (1 and 2 live mites)
10
brood as compared to removal of dead mites and brood cells having simple cuts prove that A. mellifera can detect the presence of T. clareae in the cells possibly due to certain signals or perceiving odour. Uncapping and removal of pin killed, freeze killed and artificially mite infested brood by the selected colonies further confirm the selection for T. clareae tolerance by considering damaged mites as useful tool in future breeding programmes. Even tests of hygienic behaviour can be considered for sorting out T. clareae tolerance lines as established in case of Varroa jacobsoni. Significantly more leg damage (29.59%) was found as compared to head (4.47%) and shield (1.88%) during period under investigations. Sulphur treated colonies were found to be infested at a very low rate (0-2.15%). In view of above all, it is concluded that the quantity and quality of damaged naturally fallen mites indicate specific defensive behaviour of A. mellifera. Present studies indicated that A. mellifera is capable of defending itself from T. clareae by possibly uncapping and removing mite infested cells or damaging free moving mites in honeybee colonies. Above all traits can be exploited for selecting A. mellifera lines comparatively tolerant to T. Clareae and in long run, minimizing the use of chemicals in hive environment. IX) Studies on bumble bee (Bombus haemorroidalis) domicilation and related aspects The research work on Domestication of Bumble bee was initiated for the first time in the country during 2000-2001 in the search of an alternate pollinator for increasing crop productivity. Natural nests were located in the field of bitter gourd and French bean. The population of bumble bee was monitored regularly from September, 2001 onwards daily throughout the day between 0525 to 1825 hrs on incoming and outgoing foragers at the nest entrance. Considerable decline in activity was recorded during October-November and no activity was seen during winter months. Queen started appearing during month of November, 2002. Which was proved to be the time of queen appearance in the nature as no information was available on any of the aspect studied. Natural nests were dugged up to know about the nest architecture of the species and nest was found under the soil attached to tree roots. The nest was found to be made up of dried leaves of maize and rotten wood. Dead bumble bees were found trapped in the woven mass with the dried material including soil and stone pieces. Interestingly the nest was found to be heavily parasitized with caseworm (Lepidoptera). The nest was located at a depth of 17 cm and its diameter was 18cm. The length of tunnel from the active hole was 72.5 cm. The opening was at the height of 27.5 cm with opening diameter of 10 cm. The nest was placed at the pit measuring 22.5 cm in length and 17.5 cm in breadth. Queens were found heavily infested by Kuzinia laevis. In an attempt to artificially rear the bumble bee aSweden type domicile consisting of a pair of boxes, one of which was closed and other covered with glass was manufactured and tested. The boxes were placed next to each other and were connected with a narrow tube extending from small opening bored through both the walls. Bumble bees were fed 50% honey solution and were kept at a temperature of 33-340C and were fed corbicular pollen. After 29 days of confinement the queen started making fine cells in the small chamber. At first the wax flakes were deposited on the floor of the box and later scrapped to construct the cells. An egg was laid near the cell wall. The queens started making wax mounds and depositing eggs in the cells. This was the first attempt to rear Bombus haemorroidalis in India. Successful in rearing bumble bee colonies to its full strength during 2005. Reared another unknownbumble bee species existing under our conditions during 2006. Nine colonies of bumble bees were reared. For the first time in the country the bumble bees are being tried and utilized in polyhouses
11
for vegetable crops. Studied the mating behaviour of bumble bees in cages. In the studies conducted on the domicilation of bumble bee (Bombus sp.) and their resource partitioning with honey bees, the main flora sustaining the queen population was identified under Nauni conditions. The queen can be directly trapped from Salvia moorcroftiana, Rosmarius officinalis, Lavendula sp. and wild plants like Caryopteris bicolor, Scutelleria linearis. Studies on the rearing of bumble bee revealed that colonies can be initiated after trapping queens from these plants during morning and evening hours. Domestication of bumble bee was successfully done under controlled conditions maintaining the temperature of 25-300C and 60-65% relative humidity and by feeding them sucrose solution (50%) and corbicular pollen. Fifteen colonies were successfully reared with the range of population 80-100 individuals for 7-9 months. The best nesting cage was wooden box and for feeding bottle lid was easily accepted. Nest architecture of the artificial reared colonies were studied in two chambered wooden box and hoarding cages. All the nests contained number of cell layer and brood cells in the centre, where as number of honey pots were mostly arranged in cluster near the nest periphery. Foraging activity of three natural nests were recorded and found peaked in the morning hours. Bumble bee start foraging activity from dawn to dusk period. Therefore bumble bee has long working hours. Time spent outside by the marked bees ranged from 10.83-21.33 min./bumble bee. The study revealed that the forager dose not exhibit different foraging trip after leaving the nest during different day hours. Nest architecture of B. haemorrhoidalis nests were studied for the first time in Nauni, Solan District and all nests were found to be built underground inside the abandoned site of rodents interwoven with grass, moss, wooden pieces, leaves beneath which a thatched structure (16.8 cm in width and 7.5 cm in height) covering layers of brood cells, honey pots and worker brood was observed. Natural nests were digged out after ceasing the nest activity. All these nests were built underground inside the abandoned site of rodents burrows and found attached to the root of adjoining tree or shrub. Nests were found to be interwoven with grass, moss, wooden pieces and leaves beneath which brood cells lies. The study revealed that bumble bee prefers darker and corner side of the ground and make involucrum to protect the nest from outside environment. Comparative foraging activity of bumble bee and honey bee on the field grown cucumber, capsicum was recorded similar, but in bitter gourd bumble bee was found higher in number than honey bees. On pumpkin only bumble bee was observed to visiting the bloom shows its complete dominance. In sweet cherry bumble bees were playing an important role to pollinate the crop, where no honey bee was found visiting the bloom in Kumarsain, Shimla district. A new species B. assamensis Bingham was reported for the first time visiting apple bloom at an altitude of 2725 m above mean sea level in Narkanda, Shimla district. One artificially reared colony was kept inside the polyhouse at vegetable field on cucumber crop. Colony was kept without providing any additional food inside the rearing box. Bumble bee found better acclimatized with controlled environment. Therefore it can act as an alternate source of pollinator inside polyhouse where the honey bee activity is limited. Because of the importance of these bees as crop pollination and the recent outbreak of Varroa to honey bees industry effort should be made to profitably manage their population through manipulations so that these bees may adjunctively work with the hive bees for enhancing the crop production. A new species of bumble bee, Bombus assamensis Bingham was identified for the first time foraging apple bloom at an altitude of 2725 m above mean sea level. Developed colony was successfully used for cucumber pollination grown under polyhouse. X) Genetic improvements of Apis mellifera L. by inseminating with carniolan semen for cold hardiness.
12
Average performance of the colonies headed by carnica queens were almost same with respect to strength under packed and unpacked conditions. The colonies headed by queens having carnica blood can thrive well under winter conditions and they may have characters pertaining to the cold hardiness. Average performance of honey bee colonies headed queens of yellow blood and kept under unpacked conditions remained same with respect to strength, and storage of nectar as compared to colonies kept under kept under packed conditions and headed by same blood. Studies revealed the superiority of honey bee colonies headed by carnica blood queens in comparison to the colonies headed by the ligustica blood even if kept without winter packing. b) PATENTS APPLIED/OBTAINED OR IMPLEMENTABLE TECHNOLOGY
GENERATED Did pioneering research in the field of Bombiculture (Rearing of bumble bees. The research was initiated in 2000-2001. Successful in rearing bumble bee colonies to its full strength during 2005. Reared nine colonies of bumble bees bumble bee species during 2006. Fifteen colonies were reared during 2007- 2008. A new species of bumble bee, Bombus assamensis Bingham was identified for the first time foraging apple bloom at an altitude of 2725 m above mean sea level. Developed colony was successfully used for cucumber pollination grown under polyhouse . IMPLEMENTABLE TECHNOLOGY GENERATED: I) Mass queen rearing technique : • The technology was developed for mass queen rearing of honey queens under our conditions. • The bee-keepers can use the technique for multiplying their stock. • The queen rearing technique can be adopted as an independent profession by bee-keepers and can
supply any number of queens to the other bee-keepers. • The standardization of instrumental insemination technique for the first time in India solves the
mating problem of the queens. The bee-keepers can send the queens of their stock for artificial insemination which is as good as natural mating.
II) Infra-red video Technique
Infra-red Technique was developed to have more natural observations on bee behaviour in relation to various aspects. Till the development of this technique most of the behaviour studies in honeybees were done by using visible light. This technique was used rather than visible light because bees do not respond to infra-red light and do not feel disturbed during long term observations. Conducting experiments particularly on social behaviour under such influences attracts errors and may not be natural. Most of the studies till the advent of this method were confined to observe bees in observation hives. In observation hives also the behaviour may be altered by the influence of light. Moreover, it is not practically feasible to sit around the clock in front of observation hive to study particular behaviour of different castes.
Infra-red-camera is fixed on one side of the hive. The miniature camera : (4x4x3) consisted of
a black CCD-chip that had a resolution of 512 pixels (horizontal) and 582 : (vertical). The resolution was adequate, if observation area on the bee hive does not exceed 10x10 cm. The general sensitivity
13
of visible light was rated at 0.2 lux. However, we used mainly its extended sensitivity into infra-red region with the help of infra-red LED's. The optical lens had a focal length of only 4.48 mm, so that close ups (upto 2x2 cm) can be obtained.
The newly emerged bees can be caught or emerged in the incubator. They are marked with different colour combinations of number plates. The marked bees were placed in one frame cage. The cage containing these marked bees was placed in a nucleus with four frames including one adjacent broad frame. Twenty four hours before the test the queen is shifted to the cage containing marked bees. This assured stabilization and better harmonious conditions in the cage during long term observations. The unit was provided with enough provisions during experimentation. The camera was connected to controller, a monitor and a video-recorder assembled in a separate chamber for recording observations. The activities of bees for example what is really and naturally happening inside the hive can be seen on the monitor. The observations can be video-recorded continuously.
In our studies the technique yielded significant importance particularly while studying the defense behaviour of A. mellifera carnica against Varroa. The selection of individual workers for a particular trait and making use of their reproduction in particular, increased the importance of method. Such devices are very useful in carrying breeding studies in a complicated organization like honey bee. The same method can be modified for use in other honey bee species like Apis cerana or other social insects for naturally studying their behaviour under most natural conditions. Besides scientific importance, it can be a useful teaching aid for the student engaged in biological sciences. Children are attracted most, when they watch the amazing world of bees on a monitor installed in a room and they are working in a colony outside. The world of bees can be watched by the interested people in the drawing rooms on the monitor like aquarium. The technique is an important aid of carrying extension activities particularly in our country where people do not know much about bee world. The colony set up can be explained to small children and bees importance can be understood by them easily without fear of stinging. (III) Bumble bee rearing technology
A) Initiated bumble bee rearing research in India and developed technology to rear bumble bee colonies under captivity and maintained under laboratory conditions for nine months.Laboratory reared colonies were successfully used for pollinating cucumber, variety Punjab Naveen grown under poly house resulted in increased fruit set (76.97%), healthy fruits (83.62%), fruit length (20.75 cm), fruit diameter (11.01 cm), fruit weight (413.62g), seed number (422) and seed weight (31.50g) with reduction in crooked fruits (16.18%). Over control (no pollination) induced bumble bee pollination increased fruit set, healthy fruits, fruit length, fruit diameter, fruit weight, seed number and seed weight by 27.64, 22.21, 20.78, 23.80, 36.48, 21.52 and 12.54 percent respectively. The cost benefit ratio was found to be 1:2.02 and an increase of 22% in the total returns from bumble bee pollinated crop. These pioneering studies revealed the importance of inducing bumble bees in increased quality and quantity in cucumber grown under polyhouses.
