Anatomia e distribuição topoquímica de
componentes em materiais lignocelulósicos

Outline
- Why to study anatomy and topochemical distribution of
components in lignocellulosic materials?
- Wood and non-wood anatomy
- Cell wall structure: SEM and cell wall models
- Polysaccharide and lignin localization in wood cell walls
- Lignified cells do not permit enzyme infiltration
- Lignocellulosic materials with reduced lignin content are easier to
be hydrolyzed
1. Enzymatic hydrolysis of polysaccharides is supposed to
be a key step in cellulosic ethanol production
Why to study anatomy of lignocelullosic materials
and component topochemistry?
Is cellulose
free an
“clean” as
shown in this
figure?
2. Understanding cell wall structure is useful for any type
of application of lignocellulosic materials
Lignified cells are
arranged in a
complex matrix!
It is necessary to
study the anatomy of
lignocellulosic
materials Softwood
Madeira e gramíneas são similares em termos
anatômicos?
Onde está a cellulose e a hemicelulose que se
pretende utilizar (ou digerir com enzimas)?
A lignina ocorre em todos os tipos de células? A
concentração varia de acordo com a região
celular?
O que deveríamos poder responder........

Hemicellulose localization in cell walls
• Immunohistochemistry
– Primary antibody: monoclonal antibody to (1-3,1-4)-ß-glucan (MOUSE IgG)
– Secondary antibody: Alexa Fluor 514
Fluorophore
Example o immunofluorescence localization of substitute xylans in fiber (F)
and vessel (V) cell walls of mature xylen from apen wood. LM11 denotes
the antibody used to bind to substituted xylan structures. The bar size
corresponds to 10 m. Note that the cell corners and middle lamella were
poorly labeled whereas secondary walls were strongly labeled with the LM
11 antibody indicating the regions where substitute xylan predominates in
the mature xylem (Reproduced from Kim et al. 2012)
