Science. Out of these, the KOD polymerases and kits demand a researcher’s unremitting attention. The KOD DNA polymerase was isolated from the hyperthermophilic archae-um Thermococcus kodakaraensis KOD1, and possesses superior fidelity and processivity that enables more accurate and efficient DNA amplification as compared to the amplifica-tion that can be achieved with conventional PCR enzymes including the Pfu and Taq DNA polymerases.

The KOD Plus (Cat. Nos. KOD-201 & KOD-401) series was developed based on the KOD DNA polymerase and exhibits high PCR fidelity. On the other hand, whilst the KOD FX (Cat. Nos. KFX-101 & KFX-201) series was also developed based on the KOD DNA polymerase, it shows a much greater PCR success-rate (based on efficiency and elongation capabilities) than that showed by the Taq-based PCR enzymes.

KOD FX is also effective for amplification from crude specimens (e.g. mouse tail lysates, plant lysates and organisms bearing cell walls (e.g. yeast, fungus, gram-positive bacteria). KOD Plus Neo and KOD FX Neo, the advanced versions of KOD Plus and KOD FX respectively, also contain an elongation en-hancer that enables long range and fast PCRs.

Toyobo offers a range of real-time PCR Master Mixes. The Realtime PCR Master Mix series (Cat. Nos. QPK-101 & QPK-201) and the THUNDERBIRD® series (Cat. Nos. QPS-101 &

QPS-201) are Taq DNA polymerase-based 2X master mixes for re-al-time PCR, which contain all the necessary components, except the primers and probes. The THUNDERBIRD® series shows re-duced primer-dimer formation and the ROX dye is provided sepa-rately to enable compatibility with various kinds of real-time PCR cyclers. The RNA-DirectTM series (Cat Nos.QRT-101 & QRT-201) is a 2X Master Mix for 1-step real-time PCR. This series is based on a thermostable DNA polymerase derived from Thermus ther-mophilus (Tth) HB8. The Tth DNA polymerase exhibits reverse transcriptase activity in the presence of Mn2+ ions. This system enables setting up a 1-step real-time PCR including the reverse transcription and qPCR steps.

The KOD SYBR® qPCR Mix (Cat. No. QKD-201) is a highly effi-cient 2X SYBR® Green Master Mix for real-time PCR that is based on the 3’-5’ exonuclease deficient KOD DNA Polymerase. This master mix contains all the components required for a qPCR ex-cept the ROX reference dye and primers (the 50X ROX reference dye is supplied separately with this kit). amplified using any polymerase that generates dA overhangs at the 3’ ends such as the Taq DNA polymerase, Blend Taq® (Cat. No. BTQ-101), Blend Taq® Plus (Cat. No. BTQ-201) or KOD Dash (Cat. No. LDP-101). TArget CloneTM Plus can be used for the TA cloning of the PCR products with blunt-ends or the ones amplified using the KOD Plus or KOD FX series. The KOD Plus Mutagenesis Kit (Cat. No. SMK-101) is an inverse-PCR based site-directed mutagenesis kit that was developed on the basis of the high-fidelity KOD Plus DNA Polymerase. This kit enables not only the introduction of point mutations, but also the introduction of large insertions and deletions. The PCR fidelity of KOD Plus is 80-folds great-er than that of Taq DNA polymerase, therefore, the rate of introducing unexpected 2nd-site mutations can be reduced. The PCR reactions can be performed using standard PCR primers and do not require phos-phorylated primers because this protocol contains a step for the ‘Phos-phorylation/Kination’ofPCRproducts.
