11-1 Location

l The place (bench, table or stand) where the incubator will be set up MUST be steady,

firm and level. Make sure the surface that incubator will be placed by using adjustable

feet as needed. To move the instrument, two or more persons are needed.

l Please keep 25cm or more of clearance in the back and both side of the incubator.

l Avoid placing an incubator where cold air drifts directly to the incubator such as air

conditioning or air vent. It may cause condensation in the chamber.

l All the selves, brackets, supports and surface of the chamber are thoroughly cleaned

and sterilized with ethanol or autoclaves to make sure there is no contaminant on all

these accessories. The laboratory itself introduces common source of contamination,

incubator is designed to promote growth, through and constant cleaning are

recommended.

l Choose the place in the laboratory where there is no drastic change in temperature is

expected to avoid condensation in the chamber. Preferably, incubator should be

placed in a clean room environment such as biological clean room.

11-2 Power

This incubator is designed to be operated with 110V 60Hz(US)/230V 50Hz (EU). Insert

the supply cable in a plug with correct 110V 60Hz(US)/230V 50Hz (EU). The power plug

connection must be independent, and provided with adequate earth connection.

28

Water in the humidity water pan should be distilled or pure water and supplied in the pan to

1~2 cm from the bottom of the pan. Humidity water, because it is stagnant, can be

common source of contamination within the incubator. So we strongly recommend that the

humidity water should be replaced weekly or more often, and the water pan should be

autoclaved or wipe down with a 70% of isopropyl solution or 70% ethanol.

It is also recommended to warm up the humidity water to about 37C before it is pored into

the pan for faster evaporation regain.

15. Humidity

When incubator is powered off more than 30 minutes, make sure to remove the water from the chamber. When incubator is powered back up, make sure there is NO humidity in the chamber. Humidity can harm CO2 and O2 sensor when incubator is powered off.

Gas port(s) is located on the back of the incubator as shown in Figure 16. Connect plastic

hose(s) (supplied with the incubator) and firmly tie them up with cable ties. The other ends

of the hose(s) needs to be connected to gas regulator of gas supply tank(s).

*APM series only

16-1 Gas Pressure

The optimal gas pressure is 0.03~0.05 Mpa (0.3~0.5 bar / 4.35~7.25 psi)

16. Gas Connections

Figure 16

APC series (CO2 incubator) can only be connected with CO2 gas, and APM series (Multi-Gas type) can be connected with N2 or O2 gas of the user ’s choice. In case O2 gas is connected to the incubator, make sure to be cautious of the usageof fire as high concentration of O2 gas increases the combustibility of flammable substances. Excess gas pressure may cause the malfunction of the incubator. Be sure to select the correct gas type to your application of the incubator. ? See page 24 for gas selection for N2 and O2. Humidity and partial pressure of CO2 may cause the unstable control of high concentration O2 gas at the approximate range of 18~23%.

19-1 CO2 Check prior to supplying CO2 gas to Incubator (CO2 Zero) Temperature and Humidity must be adequately prepared prior to supplying CO2 gas to

incubator. CO2 concentration must be 0.0% (ambient air).

19-2 CO2 Check CO2 gas concentration must be checked frequently by CO2 analyzer for optimized use. If

the displayed CO2 concentration differs from the concentration measured with calibrated

CO2 analyzer, modify the concentration on the display of the incubator to the concentration

measured with the analyzer.

Before the Gas in the incubator is sampled out for concentration check, set the incubator to

“Sampling Mode”. The Sampling Mode prevent CO2, N2 or O2 gas from coming into the

chamber from the gas tanks in order to measure more precise concentration in the chamber.

? See the section 22 for Sampling Mode.

? See the section 23 for Adjustment and Calibration of Gas concentration.

For more accurate measurement of CO2 and O2 concentration, there should be none of

such gas supplied during the measurement. The sampling mode allows the user to shut

down the supply of gas for 3 minutes for reason above.

Sampling Mode : <UP> key pressed for 5 seconds

During the Sampling Mode, Temperature display shows SP and the current temperature by

turns for 3 minutes. Concentration check must be done during this period. After 3

minutes, the Sampling Mode automatically cancels itself and goes back to normal function.

19. CO2 Concentration Check

When you samples out some amount of Gas in the incubator, it sometimes activates false ADD WATER alarm. This is provoked by the gas sample port is capped with the Gas analyzer, and the analyzer pumps some gas out of the incubator and this causes negative pressure in the chamber. The false ADD WATER alarm will be deactivated when the pressure in the chamber regains positive pressure when gas sample port is open after sampling gas.

(see page 24, Section 19-1-3 for CO2 Concentration setting), and it needs to be

adjusted, change the value on F-1 to actual CO2 concentration by using <UP> and

<DOWN> cursors. It will allow the incubator to automatically adjust the value to

the measured value.

21-6-2 CO2 ZERO Manual Calibration F-2

This function allows the incubator to carry out manual ZERO Calibration. As a

default, F-2 shows ON on the display. Change this to OFF with <UP> and

<DOWN> cursors and wait for 10 second until the incubator starts manual

calibration.

stabilized. And check the CO2 concentration again for double check.

As described in the section 22-5, the O2 sensor used on APM operates on galvanic battery

and it has life span. It needs to be adjusted the voltage that effect the O2 concentration

control. In case the battery is extremely run out, it must be replaced.

There are two ways to calibrate the O2 sensor on APM series as described below.

22-1 Automatic O2 Calibration Automatic calibration works when the incubator is powered on and O2 setting is ON (see

Page 29 for section 18-2-2: O2 ON/OFF setting). The automatic calibration performs no

later than 12 hours after the incubator is first turned on. And it will perform once in every

week.

During the calibration, the O2 display shows the actual concentration of O2 and CAL by turn.

22-2 Manual O2 Calibration Manual O2 calibration should be carried out when O2 sensor is replaced, or the incubator

has been not used for long period of time.

22-2-1 Manual Calibration

Go to ST5 by pressing SET, <UP> and <DOWN> keys at the same time, go to E-4.

You will see OFF on the display. Change it to ON by press <UP> cursor and leave it

for a few seconds. The incubator starts manual calibration on O2 sensor.

22-3 O2 Sensor failure When the battery in O2 sensor wears out, O2 display shows the actual concentration and

“---“ (bars) by turn, which indicates O2 sensor failure.

The O2 sensor still controls the concentration even if the sensor failure is shown, but it is

recommended to replace the sensor with new one at earliest possible.

After replacing the sensor, make sure to do the Manual Calibration of the sensor.

? Please refer to the section 23-2-1 for Manual O2 sensor calibration.

Is the inlet supplied with electric power?

• Check the breaker in your room or outside of

the building.

Is the breaker pulled out from the body?

• Put the breaker in place and power on again.

Warning! There are two fuses restorable: One is in the box on the back,

and a second on the back next the main switch. Check both.

Is the power switch on?

• Check the switch and press ON of it.

Breaker of the device

is turned off even

though power is on.

Is any liquid inside the device?

• Pull the power plug out immediately in the

case of water splashing or pouring on the body,

Contact Astec or any other distributors as soon as

possible. If you keep using it, it may result in fire.

Is the incubator placed in a rusty place?

• Rust may stick to the power line to make it

short circuit. Pull out the power plug immediately

and contact Astec or distributors. It may cause fire

if you keep using it.

Breaker on the side of

building goes down.

Is the same power line divided into many devices?

• Move each device to single lines. It may

cause fire if you continue using without taking

measure.

Is the incubator connected into outlet with other machine that needs

much power?

• Connect the line of incubator to the single

power inlet.

The temperature

inside the chamber

cannot be stabilized.

Is the fan inside the chamber moving properly?

• Check if the fan is not touched to the duct or

the sidewall of the chamber.

Is the glass window closed well?

• Close the window properly.

Inside temperature

cannot reach the set

temperature.

Is the set temperature low?

• Confirm the set temperature and set it to be

[room temp + 5℃] if it is low.

Is the room temperature too high?

• The set temperature can be effective only it is

not lower than [room temp + 5℃ ]. If the room

temperature is too high, adjust it lower.

CO2 concentration

cannot be stabilized.

Is the fan inside the chamber moving properly?

• Check if the fan is not touched to the duct or

the sidewall of the chamber.

Are shelves positioned where they should be?

• Disposition of the shelves causes bad circuit

of the air inside and CO2 concentration cannot be

stabilized. Put shelves in position. See p.10.

Is the humidifying water enough?

• Pour water into a tray immediately if there is

not enough water.

CO2 concentration

does not become the

set value.

Is the set CO2 concentration value low?

• Confirm the set value. See p. 13.

Is the rest of gas enough?

• Exchange with a new gas cylinder

immediately if there is no gas in the cylinder.

Is the regulator of CO2 gas cylinder adjusted to be 0.03 to 0.05 MPa?

• Adjust the second pressure of the regulator if

the pressure is too low. See p. 10.

Is the exit valve of the regulator full open?

• Open the exit valve fully on use.

Is the CO2 gas line tube connected to the provided CO2 gas supply?

• Confirm the tube connected to CO2 gas

supply on use.

Page 44: Astec - Penguin AQ (APC/APM) Incubator User Manual - Supplied by RI UK and Ireland

44

Is the gas line tube bended or squashed?

• Repair the bended or squashed parts on use.

Contact our distributor or us if the repair seems to

be too hard or the tube is torn.

CO2 alarm is beeping.

Is the gas line tube bended or squashed?

Repair the bended or squashed parts on use. Contact our distributor

or us if the repair seems to be too hard or the tube is torn.

ADD WATER alarm is

beeping.

The water level of the water jacket is low. Supply water from the

water supply port at the front of the body. Refer the “Supplying

water” on p. 11.

ADD WATER alarm

does not stop during

supplying water.

Is the water supplied Super pure water or Ion exchange water?

• The water level us detected by electrical

conduction. The sensor cannot response the water

with high resist value (14MΩ) such as super pure

water or ion exchange water. Supply antirust

solution attached before the water full because it

decreases resist value.

General information This device adjusts high-pressure gas to low pressure to release the gas with regular pressured

quantity.

There is a high-pressure gas inside the regulator. Use the pressure-resistant device and inspect

it regularly to keep normal condition.

Preparation 1. The rust in regulator will cause troubles (gas leak, closure of gas path,

etc). When installing to gas cylinder, clean the cap of the cylinder

completely. On new plumbing, remove all of the rust, water, and lotion

first.

2. When connecting regulator and gas cylinder, check that the gasket is

inserted in position. If there is no gasket or the gasket is damaged, use

preparatory one.

3. On installing regulator to gas cylinder, use right sized spanner and

tighten firmly. Check gas leak after the installation.

Operation 1. Turn the pressure-adjusting handle to “CLOSE” or “SHUT” (counterclockwise) before gas

supply. Shut the valve on the exhaust to stop gas flow.

Do not turn handle to “OPEN” by force when there is no gas in the chamber or the cylinder is

not connected.

2. Open the valve of gas cylinder slowly and keep it to small amount until the gas fills with the

chamber. After the gas full, open the valve fully.

Avoid standing right in front of the pressure meter when opening or closing

valve.

3. The regulator with high lateral pressure meter indicates the pressure at intake.

4. On the use of exhaustive pressure changeable regulator, the second pressure will be

increased if the from pressure adjusting handle or screw turned to the direction of “OPEN”.

The range of adjustable pressure is 60 to 70% of the maximum value of constant and lateral

meter.

5. Open the valve of the exhaust to release gas.

♦ Air dust filters are NOT autoclavable.

♦ Sterilization with ethanol or method is not recommended since such methods do not

remove dusts trapped in the filter.

♦ Incubators must be kept clean regularly even when air dust filter is in place.

♦ Do not touch air dust filters with bare fingers/hands. Aseptic gloves should be used.

♦ Air dust filters must be handled with care since it is somewhat fragile.

♦ Air dust filters must be stored protected from high temperature, humidity and dust.

♦ Do not immerse air dust filter in any kind of liquid.

♦ Air dust filters are disposable products, not reusable.

[How to install Air dust filter]

1. Hook on the right side of the Air dust filters to the right steel

clip as shown in Figure 1.

2. Gently push up the filter until the other side of the filter clips

on the left steel clip as shown in Figure 1.

3. Make sure the filter is firmly placed and centered.

[How to remove Air dust filter]

Unclip the left side of the filter by gently pushing up the left steel

clip.

Air dust filters should be replaced at least once in every year. However, every laboratory must define and optimize the replacement interval for its own risk management according to the environment of laboratories.
