Introduction
Vidas and Vidas Up run on the same Vidas equipment. They are both ELFA (Enzyme Linked Fluorescent Assay).
Both provide Negative and Positive results
Both are a screen for enriched samples
Negative = Not Detected, test is complete, result is reported as Not Detected
Positive = Presumptive positive (also called suspect), has to be confirmed

VIDAS
Traditional ELISA and ELFA bind with antigens recognised by antibodies. These antigens are on the cell wall of the target bacteria, in our case our targets are Salmonella and Listeria. These antigens have the disadvantage of some level of cross reaction with non-target bacteria, such as Citrobacter and Hafnia for Salmonella.
VIDAS UP
Recombinant Bacteriophage Protein (RBP) use the receptor sites for bacteriophages (virus that invades bacteria) on the cell membrane to selectively capture bacteria. They have superior stability, specificity, and affinity to carbohydrate binding proteins such as antibodies and lectins over the traditional cell wall/membrane antigens used in ELISA and ELFA. To exploit the use of RBPs, VIDAS UP uses the phage protein screens to identify binding partners that recognize the host bacterium, i.e. the target Salmonella or Listeria.

Any sample quantity larger than listed in the table, so no 375 g samples, largest validated sample size is 125 g
Any food category not listed
Verification will be required for:
Any sample matrix not listed in the validated food category, until it has been verified, e.g. Misc – can test black pepper, but any other spice (ginger, chilli, garlic, etc.) must be verified first
We have access to the Mérieux NutriSciences corporate network verification database so we can check this first to see if verification may be required

Some Chocolate types – not all variations have been validated or verified
Verification will be required for:

What you need to know Turn around time is next day, not 24 hours. Incubation time alone
is 18-30 hours depending on the test and sample
To ensure next day results, sample receipt time at the lab is critical, especially for Listeria
Lab needs 1-2 hours from receipt to complete sample registration and setup
Incubation time is 22-26 hours for Listeria test environmental samples and 26-30 hours raw material/product samples
Incubation time for Salmonella is 16-26 hours (18-24 hours for most samples). The time depends on both the sample matrix and the sample size.
Note that some dairy products (e.g. raw milk cheese) also require a 6-8 hour second enrichment making a total incubation time of 24-32 hours

What you need to know (cont.) VIDAS UP analysis time is 1.5 hours
Data checking and issue/release of COA is 0.5 to 1 hour
Therefore, total time is 27-31 hours for environmental samples and 31-35 hours for raw material/product samples for Listeria
If you as a client want next day COA by 5pm, sample will have to be in sample registration preferably by 10 am, and absolutely no later than11 am. If received later, sample will have to be set up following day to comply with validated incubation times
Lab will need to be aware that VIDAS UP sample is due to be delivered so that suitable staff are rostered to process sample

Take Home Message: All of our labs are NATA accredited for Listeria and Salmonella by
VIDAS UP
Check with us that your sample matrix can be tested by VIDAS UP without having to first verify or validate
Remember the delivery/receipt time requirements at the LAB when sending samples for testing
Remember to advise the Lab to make sure that the Lab is aware of incoming samples. This will ensure they meet setup deadline to achieve next day result reporting to you
