Avian Influenza (AI)

• Small RNA virus

• Surface spikes –hemagglutinin & neuraminidase proteins

• 16 hemagglutinin and 9 neuraminidase subtypes – Ex. H5N1, H9N2, H7N7

• Vary in severity of disease: – Low Pathogenicity (LP)– High Pathogenicity (HP)

Concepts

• Exposure: access to the virus strain • Adaptation: progressive genetic changes in a virus strain that result in increasing efficiencies of replication in a host species• Infectivity: ability of the virus strain to infect and replicate in a particular host species• Transmissibility: natural host-to-host spread• Pathogenicity: ability to produce lesions, disease and/or death in a host

Critical Factors• Infectivity for the host (quantity of virus replication directly associated with pathogenicity)

• Receptor binding sites (HA) + host cell receptors

• AI viruses bind to α2,3-galactose linkage receptors on cells• Swine and human influenzavirus A bind to α2,6-galactose linkage receptors• Other receptors contribute to host species and cell type specificity?

• Cleavage of HA: HA0 → HA1 + HA2 (Trypsin-like vs. Furin proteases) needed for infectivity• Replication machinery (polymerase complex - PB1, PB2 & PBA)

Host Adaptation & Transmission of Influenza A Viruses

• Most Common: Intraspecies, e.g. human-to-human, pig-to-pig, chicken-to-chicken, etc.

• Occasional: Interspecies & intraclass, e.g. pig-to-human, wild-mallard-to-domestic turkey, etc.

• Recently, but rarely: Interspecies & interclass, e.g. bird-to-human, bird-to-pig, etc.

• Conclusion: Influenza viruses express host adaptation to various levels

• AI viruses are composed of diversely different HA & NA subtypes and pathotypes

• Most AI viruses are LP

• AI viruses are host adapted, but they can cross between species and re-adapt

• Some LP H5 and H7 AI virus have mutated to HP

• There have been 24 epizootics of HPAI in the past 50 years

• The Asian H5N1 HPAI virus has spread into northern and western Asia, and eastern Europe, with evidence of involvement of migratory birds

U.S. Poultry Industry Structure- Advantages Modern integrated agricultural system

Birds owned by agribusiness (integrator) Major facilities owned by agribusiness

Feed mill, hatchery, processing plant Growers dedicated to specific processing plants

Contracted to single company Small number of people tend large numbers of birds Single-age, all-in all-out production scheme Avg 60 mile radius of feedmill/plant

Page 29: Today’s Speakers: Dr. David Swayne - virologiahoy.orgvirologiahoy.org/Dealing_with_the_challenge.pdf · Today’s Speakers: Dr. David Swayne Director of the Southeast Poultry Research

U.S. Poultry Industry Structure- Advantages

Closed system NOT traded in livestock markets Little cross-over between complexes/companies

AgriBUSINESS Morbidity, mortality, daily gain, feed

conversion, condemnations etc all closelymonitored

Measured in hundreds of a cent ($0.0001) Problems identified and responded to quickly

Page 30: Today’s Speakers: Dr. David Swayne - virologiahoy.orgvirologiahoy.org/Dealing_with_the_challenge.pdf · Today’s Speakers: Dr. David Swayne Director of the Southeast Poultry Research

U.S. Poultry Industry Structure- Advantages Effective prevention programs in place

Strong veterinary/health oversight Company veterinarians trained in poultry medicine

Enclosed housing Bird-proof, no access to outdoors (wildlife)

Extensive biosecurity programs Limited access to poultry houses Dedicated clothing, boots etc Limited sharing of equipment Contact with psittacines/other fowl prohibited

Page 31: Today’s Speakers: Dr. David Swayne - virologiahoy.orgvirologiahoy.org/Dealing_with_the_challenge.pdf · Today’s Speakers: Dr. David Swayne Director of the Southeast Poultry Research

U.S. Poultry Industry Structure- Advantages Extensive and modern diagnostic network

Company laboratories State veterinary diagnostic laboratories Federal veterinary diagnostic laboratory

Extensive surveillance programs in place USDA-APHIS NPIP program

Breeders and market birds State programs

 Active and passive surveillance

Page 32: Today’s Speakers: Dr. David Swayne - virologiahoy.orgvirologiahoy.org/Dealing_with_the_challenge.pdf · Today’s Speakers: Dr. David Swayne Director of the Southeast Poultry Research

U.S. Poultry Industry Structure- Disadvantages Just in time production scheme

Large numbers of birds processed daily Large number of birds in one flock Large number of flocks on a premise Often grown in concentrated poultry

production areas Delmarva, Shenandoah Valley, Arkansas

Euthanasia and disposal of large flocks can bechallenging

Page 33: Today’s Speakers: Dr. David Swayne - virologiahoy.orgvirologiahoy.org/Dealing_with_the_challenge.pdf · Today’s Speakers: Dr. David Swayne Director of the Southeast Poultry Research

Industry Response Plans HPAI Response Plans

 Already developed in poultry producingstates

Industry association/State cooperative effort Tested by recent LPAI outbreaks in several

states VA, NC, MD, DE, TX, PA, CA

 Continuously reviewed and improved

Page 34: Today’s Speakers: Dr. David Swayne - virologiahoy.orgvirologiahoy.org/Dealing_with_the_challenge.pdf · Today’s Speakers: Dr. David Swayne Director of the Southeast Poultry Research

Industry Response Plans Contain provisions for:

Immediate quarantine of affected and at-risk premises

Rapid depopulation and disposal ofinfected birds

Effective cleaning and disinfection ofpremise prior to re-stocking

Heightened surveillance of surrounding at-risk flocks

Page 35: Today’s Speakers: Dr. David Swayne - virologiahoy.orgvirologiahoy.org/Dealing_with_the_challenge.pdf · Today’s Speakers: Dr. David Swayne Director of the Southeast Poultry Research

Summary U.S. poultry industry has several

inherent advantages due to its modernintegrated agricultural productionsystem Inherent firewalls in the system

Factors affecting cost of production areclosely monitored so a disease outbreakwill not be missed

Page 36: Today’s Speakers: Dr. David Swayne - virologiahoy.orgvirologiahoy.org/Dealing_with_the_challenge.pdf · Today’s Speakers: Dr. David Swayne Director of the Southeast Poultry Research

Summary Extensive prevention, surveillance, and

response systems are already in placeand have been tested

Introduction and significant spread ofAsian HPAI in the U.S. commercialpoultry production system is unlikely tohappen
