Manual Purpose:

The scope of this manual deals primarily with the operational aspects of the

PrepLC/System 500 Liquid Chromatograph. To this end extensive use of both

verbal instructions and visual aids have been co-ordinated together on the follow-

ing pages t o present a clear and comprehensive scheme for carrying out prepara-

tive separations on the PrepLC/System 500.This detailed scheme covers such

operational aspects as starting the system, equilibrating the system column, load-

ing the sample material onto the column, collecting the eluting sample fractions,

and in the final phase of the separation process, shuting down the system for

future start-ups.

In a more limited way this manual also addresses itself to a few other aspects of

the PrepLC/System 500 that should augment the chromatographer's overall

success in this new technology of high-speed, high purity-yield preparative chro-

matography. To this aim a concise guide is included to help determine those pro-

blematical parameters necessary to achieve a given separation. Various tech-

niques are indicated therein that may prove applicable in obtaining the best re -

sults in the most efficient manner for the given separation problem. In other sec-

tions information is provided for diagnosing and correcting those minor opera-

tional malfunctions that may result from accumulated contamination or from

normal system wear after continued use. Moreover, a maintenance section i s de-

voted to detailing the routine replacement and cleaning of those expendable

items that have a limited duty life within the system. In the Appendix section

further information is presented concerning system parts, internal layout, and

schematics of the PrepLC/System500.
