ALF A) Can Reduce Peri-operative Transfusion Requirements in Patients Undergoing Major Orthopedic Surgery. R.W. Johnson Pharmaceutical Research Institute; M92-011; Phase III drug trial Site PI The major goals of the study were to determine if baseline hemoglobin concentration correlated with RBC transfusion, and if peri-operative administration of erythropoietin before orthopedic surgery could reduce allogeneic RBC transfusion. My role as the MGH site PI was to fulfill requirements for IRB approval and reporting, consent research subjects, evaluate adverse events, and supervise the study coordinator.

1993-1994 A Pilot Phase Ib Study to Evaluate the Safety and Tolerance of Hemoglobin-Based Oxygen Carrier-201 (HBOC-201) in Patients Undergoing Open Repair of a Unilateral Long Bone Fracture, Hip Replacement or Knee Replacement. 2000-2001 A Prospective, Randomized, Concurrent Trial of Leukoreduction for all Hospitalized Patients. Baxter HealthCare Corporation; investigator-initiated individual research project Co-PI The object of the study was to determine if there were differences in significant clinical outcomes (death, length of stay, infection rate) among patients who received leukoreduced blood components compared to patients receiving conventional, non-leukoreduced blood components. I participated in all phases of the study including design, execution, data

1998-2004 An Open-label, Randomized, Parallel Group Study to Confirm the Safety and Efficacy of Procrit (EPOIETIN ALF A) Administered Perioperatively vs. Standard of Care in Blood Conservation in Subjects Undergoing Major Elective Spinal Surgery R.W. Johnson Pharmaceutical Research Institute; PR97-19-002; Phase IV drug trial Site PI The study was carried out to determine if the incidence of thrombotic complications was increased m spme surgery patients, who do not routinely receive prophylactic anticoagulation, receiving peri-operative erythropoietin. My role as the MGH site PI was to fulfill requirements for IRB approval and reporting, consent research subjects, evaluate adverse events, and supervise the study coordinator. In addition to MGH being the top enrolling site, I was also on the writing committee.

2005-2006 Noninvasive Transdermal Delivery of Topical Anesthetics Using Ultrasound Pretreatment of Skin for Donors in a Blood Center Sontra Medical Corporation; 2004-TA-05; investigator-initiated individual research project Site PI The objective of this study was to evaluate the safety and efficacy of ultrasonically­facilitated delivery of an approved topical anesthetic. As the PI, I was responsible for the

2005-2006 Measurement of Soluble CD44 in Autologous Stem Cell Grafts. MGH Department of Pathology Vickery Grant; investigator-initiated research study Co-PI The goal of the study was to determine if there was a correlation between expression of CD44 in peripheral blood stem cell graft material in patients with hematologic malignancies and clinical outcomes following autologous transplantation. I was responsible for the IRB approvals and reporting, and participated directly in the study design and data analysis, as well as writing the paper.

2007-2014 Red Cell Storage Duration Study (RECESS) National Heart, Lung and Blood Institute Site PI; Vice-Chair, Protocol Leadership Committee This multi-center study, which is being performed by the NHLBI-supported Transfusion Medicine/Hemostasis Clinical Trials Network, will compare clinical outcomes in cardiac surgery patients randomized to receive RBC stored for :::; 10 d or :=::: 20 d. The results were published in 2015 (Ref. 35) and 2017 (Ref. 46).

2009-2014 Impact of Duration of Storage of Blood on Physiologic Variables in Cardiac Surgical Subjects Enrolled in the RECESS Trial (RECAP) National Heart, Lung and Blood Institute; 1 R01 HL101382-0l Co-PI

Tissue oxygen saturation and microcirculatory flow will be assessed using non-invasive techniques among patients enrolled in the parent RECESS study at 4 sites including MGH to compare these physiologic measures among cardiac surgery patients receiving RBC stored for < 10 d or > 20 d. The results were published in 2017 (Ref. 45).

2012-2015 Tissue Oxygenation by Transfusion in Anemia with Lactic Acidosis National Heart, Lung and Blood Institute; 1 R21 HL109518-01A1 Investigator This funding is for a prospective, randomized clinical trial comparing clinical outcomes and lactate levels in children in Uganda with severe malarial anemia who will be transfused with conventional banked RBC which have been stored for .:::; 10 d vs. units which have been stored for::::::. 25 d up to the allowed maximum ( 42 d). The protocol follows the design of a pilot study conducted in 2011-12 which demonstrated that lactic acidosis was corrected by RBC transfusion. The results were published in 2015 and 2016 (Refs. 39, 40, 42).
