    Experiment 1 quick description andguaiacol safety

    observing the effects of differentconditions such as pH,temperature concentration etc;on the rate of catabolicbreakdown of peroxide by theenzyme peroxidase from turnips.

    RH2 + H2O2 -----------> R+ 2 H2O

    reducing agent = guaiacol

    guaiacol

    guaiacol

    Analysis: guaiacol is a colourless solustate, but changes colour when it is ocan measured using a spectrophoto

    Safety:eyes; guaiacol is an irritant and will cand pain. [wear goggles]skin; causes irritation to skin. [ensure scompletely down, wear gloves and shoes]ingestion; Guaiacol is harmful if ingesdrink, make sure to not touch your mhandling guaiacol and wash hands iinhalation; can be dangerous if inhause a fume cupboard]

    H2O2 safety

    Much like guaiacolHydrogen peroxide isan irritant, soprotective eyegear,handgear andclothing must be

    warn, and care mustbe taken not to ingestor spill duringhandling.

    1 small beakerA test tube rack

    bottle of distilled water

    label 3 test tubes 1 2 and 3 and and fill them with the reagtable in the handout.

    transfer some solution from tube 1 into a cuvette anspectrophotometer. ( we use a reagent blank as, although

    look clear it may be slightly tinted and will therefore have a to clear water, i.e. it prevents zero error of the spectro

    mix tubes 2 and 3 to make the sample mixture and quickly acuvette this will be t = 0 then record the absorption at 500 nm

    until 2 minutes is reached.
