Methylation andMemory FormationContributed byDavid L. Molese,
Ph.D.The methylation o DNA and histones is a dynamicprocess
catalyzed by a amily o enzymes calledDNA methyltranserases (DNMTs).
In vertebrates,60-90% o cytosine-phosphate-guanine (CpG) DNAsites
are methylated (1), indicating the importance othis mechanism to
the cell. DNA methylation playsa major role in the regulation o
gene expression bysilencing genes through conversion o cytosine
to5-methylcytosine (2, 3). This conversion intereres withRNA
polymerase initiation (4).
The methylation o DNA and histone proteins hasbeen implicated in
learning and memory, addiction,and depression. For example, in the
hippocampus, DNA methylation is dynamically regulatedduring
contextual ear conditioning in rats and mice, where blocking DNA
methyltranserases(DNMTs) inhibits memory ormation (5).
Additionally, DNA methylation plays a role in synapticplasticity.
In feld electrophysiology experiments, blocking DNMTs with
Zebularine or 5-aza-2-deoxycytidine impairs induction o long-term
potentiation at Schaer collateral synapses (6).
