Procedure

• Remove plasmids from registry – Transform• Miniprep, Digest, Ligate, Transform– Select for non RFP cells that grow on CAP

• 2 overnight cultures to prepare for a fluorescence reading

• Confirmation using PCR amplification of Biobrick sequequence inside the prefix and suffix.
