An inoculating device having an inoculating loop, an improved streaking portion and a picker. The device is constructed of high impact polystyrene material which is injection molded to form a long thin member. One end of the device comprises a loop. The other end of the device forms a head having a plurality of distinct streaking surfaces. In the preferred embodiment, these distinct streaking surfaces have a convexity and come together to form a common point or apex, for example as in a curved pyramidal shape.

There is here provided a convenient bag used at the time when microorganisms which adhere to a specimen containing a solid material are extra- cted with a saline solution and are then ex- amined. In this bag, a filter sheet is stuck to plastic film sidewalls so as to extend from the opening inlet to the bottom portion of the sidewalls, so that the bag is divided into two chambers. A specimen and a saline solution are poured into one of the two chambers, and the resulting extract is taken out by a pipet from another chamber.

The disclosed method measures the chemiluminescence of macrophage derived from an immune-deficient mouse after stimulation with the sample thymic extract, phagocytic stimulator, and luminescent detec- ting solution. A kit suitable for practicing the method is also disclosed.
