Considerations specific to the Waters Source Pressure Test Unit

Solvent leakage hazard

The source pressure test unit is used with Waters mass spectrometers fitted with a ZSpray source. The tests detailed in this document are designed to ensure that no more than 10% of the eluent entering the ZSpray™ source can enter the laboratory atmosphere. Waters recommends you perform a hazard analysis, assuming a maximum leak into the laboratory atmosphere of 10% LC eluate.

Flammable solvents hazard

Never let the nitrogen supply pressure fall below 400 kPa (4 bar, 58 psi) during an analysis in which flammable solvents are used. Laser radiation hazard.

Warning: To confirm the integrity of the source exhaust system, renew the source O-rings at intervals not exceeding one year.

Warning: To prevent the ignition of accumulated solvent vapors inside the source, maintain a continuous flow of nitrogen through the source whenever significant amounts of flammable solvents are used during the instrument’s operation.

High temperature hazard

The solvent delivery system (LC pump or syringe pump) can become hot during operation. Power off the delivery system when not in use.

Hazards associated with removing an instrument from service

Warning: To avoid burn injuries, avoid touching the source enclosure with your hand when operating or servicing the instrument.

Warning: • To avoid electric shock, do not remove the mass spectrometer’s

protective panels. The components they cover are not user-serviceable.

• To avoid nonlethal electric shock when the instrument is in Operate mode, avoid touching the areas marked with the high voltage warning symbol. To touch those areas, first put the instrument in Standby mode.

Warning: To avoid personal contamination with biohazardous or toxic materials, wear chemical-resistant gloves during all phases of instrument decontamination.

Warning: To avoid puncture injuries, handle syringes, fused silica lines, and borosilicate tips with care.

When you remove the instrument from use to repair or dispose of it, you must decontaminate all of its vacuum areas. These are the areas in which you can expect to encounter the highest levels of contamination:

• Source interior• Waste tubing• Exhaust system• Rotary pump oil (where applicable)

The need to decontaminate other vacuum areas of the instrument depends on the kinds of samples the instrument analyzed and their levels of concentration. Do not dispose of the instrument or return it to Waters for repair until the authority responsible for approving its removal from the premises specifies the extent of decontamination required and the level of residual contamination permissible. That authority must also prescribe the method of decontamination to be used and the appropriate protection for personnel undertaking the decontamination process.You must handle items such as syringes, fused silica lines, and borosilicate tips used to carry sample into the source area in accordance with laboratory procedures for contaminated vessels and sharps. To avoid contamination by carcinogenic, toxic, or biohazardous substances, you must wear chemical-resistant gloves when handling or disposing of used oil.

The source pressure test unit must be used whenever an instrument’s source enclosure O-rings are replaced (this is typically performed as part of the annual preventative maintenance). The Waters Source Pressure Test Unit is for research use only.See also: The appropriate Waters mass spectrometer and source option operator’s guides.

Calibrating To calibrate LC systems, follow acceptable calibration methods using at least five standards to generate a standard curve. The concentration range for standards should include the entire range of QC samples, typical specimens, and atypical specimens.When calibrating mass spectrometers, consult the calibration section of the operator’s guide for the instrument you are calibrating. In cases where an overview and maintenance guide, not operator’s guide, accompanies the instrument, consult the instrument’s online Help system for calibration instructions.

Quality-controlRoutinely run three QC samples that represent subnormal, normal, and above-normal levels of a compound. Ensure that QC sample results fall within an acceptable range, and evaluate precision from day to day and run to run. Data collected when QC samples are out of range might not be valid. Do not report these data until you are certain that the instrument performs satisfactorily.

ISM classification

ISM Classification: ISM Group 1 Class AThis classification has been assigned in accordance with CISPR 11 Industrial Scientific and Medical (ISM) instruments requirements. Group 1 products apply to intentionally generated and/or used conductively coupled radio-frequency energy that is necessary for the internal functioning of the equipment. Class A products are suitable for use in commercial, (that is, nonresidential) locations and can be directly connected to a low voltage, power-supply network.

1 Using the Waters Source Pressure Test Unit

This chapter tells you when to use the source pressure test unit, how to fit it to the mass spectrometer and how to perform the pressure test.

When to use the source pressure test unit

The source pressure test unit is used with Waters mass spectrometers fitted with a ZSpray source.To ensure that your instrument complies with EN61010-1 regulation, you must use the source pressure test unit to verify the operation of the source when either or both these conditions apply:

• You change any of the source enclosure O-rings.• You doubt the integrity of the source and source exhaust.

Note: The pressure test is not applicable to NanoFlow™ sources.

The source pressure test unit

The source pressure test unit comprises a pressure test meter (with integral pressure relief valve), a source exhaust valve, and parts used to install the device on the mass spectrometer.The pressure test meter connects into the instrument’s desolvation gas line via a 4-mm tee-piece. When pressure testing is not being performed, the pressure test meter is removed and a blanking plug fitted in its place.

Fitting the source pressure test unit to the mass spectrometer

GeneralThe pressure test meter is supplied with a length of 4-mm flexible tube. To perform the pressure test, you must connect the tube into the instrument’s desolvation gas line via a 4-mm tee-piece. After fitting, this tee-piece is left in position during normal instrument operation (that is, when source pressure testing is not being performed).See also: “Fitting the tee-piece to the desolvation gas line” on page 1-3.

Warning: The exhaust system of a ZSpray source is a safety device and MUST be operational.

You must connect the source exhaust valve into the source exhaust line (not the rotary vacuum pump exhaust line) before performing the source pressure test. After performing the source pressure test, you must remove the source exhaust valve from the source exhaust line.See also: “Preparing the source exhaust line for fitting the source exhaust valve” on page 1-5, and “Returning the instrument to normal operation” on page 1-12.

• Chemical-resistant, powder-free gloves• Sharp knife

To fit the tee-piece to the desolvation gas line:

1. In the MassLynx™ Tune window, click Press for Standby, and confirm that the instrument status indicator shows red.

Warning: To avoid electric shock, ensure that the mass spectrometer is in Standby mode when commencing this procedure.

2. Wait 3 minutes.Rationale: Waiting 3 minutes allows the desolvation gas flow to cool the source.

3. Click to stop the desolvation gas flow.

4. Power-off the solvent delivery system (LC pump or syringe pump).

5. Open the instrument’s access door.

6. Disconnect the solvent delivery line from the probe.

7. Disconnect and remove the 4-mm flexible tubing between the instrument’s desolvation gas connection and the probe adjustment flange.

8. Use the sharp knife to cut the tubing to fit the tee-piece.

9. Connect the two cut ends of the tubing to two of the tee-piece ports.Alternative: Connect the tee-piece into new 4-mm flexible tubing of appropriate length.

10. Fit the 4-mm blanking plug to the open tee-piece port.Note: You will connect the pressure test meter to this port when preparing the instrument to perform the pressure test.See also: “Preparing the instrument for the pressure test” on page 1-7.

11. Connect the flexible tubing, with tee-piece, between the instrument’s desolvation gas connection and the probe adjustment flange. See the figure “Gas connections when performing the source pressure test” on page 1-3.

Warning: The source can be hot. To avoid burn injuries, take great care while working with the instrument’s access door open.

Warning: The solvent delivery connections can be contaminated with biohazardous and toxic materials. Always wear chemical-resistant, powder-free gloves while performing this procedure.

Caution: To avoid gas leaks, cut the flexible tubing squarely (that is, perpendicular to its horizontal axis).

12. Close the instrument’s access door.

Preparing the source exhaust line for fitting the source exhaust valve

Required materials

• Chemical-resistant, powder-free gloves• Sharp knife

To prepare the source exhaust line for fitting the source exhaust valve:

1. Use the sharp knife to cut the tubing connected between the nitrogen exhaust waste bottle and the laboratory exhaust port at a point approximately 50 mm (2 inches) from the bottle. See the figure “Nitrogen exhaust waste bottle” on page 1-6.

Warning: The nitrogen exhaust waste line can be contaminated with biohazardous and toxic materials. Always wear chemical-resistant, powder-free gloves while performing this procedure.

Caution: • To avoid damaging the instrument, you must remove the source

exhaust valve after performing the source pressure test and before resuming normal instrument operation.See also: “Returning the instrument to normal operation” on page 1-12.

• To avoid gas leaks, cut the flexible tubing squarely (that is, perpendicular to its horizontal axis).

3. Connect the short length of tubing to one of the source exhaust valve’s ports.

4. Store the source exhaust valve and attached short length of tubing with the pressure test meter.

5. Connect the cut end of the tubing (leading to the laboratory exhaust port) to the nitrogen exhaust waste bottle’s outlet connection.

To prepare the instrument for the pressure test:

3. Click to stop the desolvation gas flow.

4. Turn off the solvent delivery system (LC pump or syringe pump).

5. Open the instrument’s access door.

6. Disconnect the solvent delivery line from the probe.

7. Remove the blanking plug from the tee-piece.

Warning: To avoid electric shock, ensure that the mass spectrometer is in Standby mode when commencing this procedure.

Warning: The source can be hot. To avoid burn injuries, take great care while working with the instrument’s access door open.

8. Connect the pressure test meter tube to the tee-piece.

9. Disconnect the tubing leading to the laboratory exhaust port from the nitrogen exhaust waste bottle’s outlet connection.

10. Connect the short length of tubing fitted to the source exhaust valve to the nitrogen exhaust waste bottle’s outlet connection.

11. Connect the free end of the tubing leading to the laboratory exhaust port to the free connection on the source exhaust valve (so that the source exhaust valve is now connected in the line between the nitrogen exhaust waste bottle and the laboratory exhaust port). See the figure “Gas connections when performing the source pressure test” on page 1-3.

12. Ensure that the source exhaust valve is open.

13. Ensure that the instrument is setup for use, that it is in the correct configuration, and that the probe is fitted.See also: The appropriate instrument operator’s guide.

Performing the pressure test

To perform the pressure test:

1. On the MassLynx Tune window Source page, set Desolvation to 0 L/hr.

2. Set Cone to the appropriate value for the source/probe type to be tested, and, if appropriate, adjust the other conditions as shown in the following table.

3. Switch on the pressure test meter by pressing and simultaneously.

4. Set the pressure test meter to read pressure in mbar.See also: The pressure test meter manufacturer’s documentation.

5. On the MassLynx Tune window, click to start the gas flow.

6. Wait a few seconds for the gas flow to stabilize.

7. Record the reading displayed on the pressure test meter (P1).

8. Close the source exhaust valve.

9. Wait a few seconds for the gas pressure to stabilize.

10. Record the reading displayed on the pressure test meter (P2).

11. Open the source exhaust valve.

12. Switch off the pressure test meter by pressing .

13. On the MassLynx Tune window, click to stop the gas flow.

14. Compare the values of P1 and P2.The source is operating correctly (that is, it passes the pressure test) if P2 > 10 × P1, and P2 > 100 mbar.

15. If the source fails the pressure test, perform the following procedure.

If the source passes the pressure test, prepare the instrument for normal operation. See also: “Returning the instrument to normal operation” on page 1-12.

If the source fails the pressure testIf the source fails the pressure test, you must check both the source and the probe and then repeat the test.

To check the source and probe:

1. Ensure that the source and probe are fitted correctly.

2. Ensure that all the source and probe O-ring seals are in place.See also: The appropriate instrument operator’s guide.

3. Ensure that the source exhaust line between the instrument and the source exhaust waste valve is correctly fitted.

Warning: If the source fails the pressure test, the integrity of its waste system can be impaired; do not reconnect or start the solvent delivery system until the source passes the pressure test.

Caution: You must remove the source exhaust valve after performing the pressure test and before resuming normal instrument operation.

To repeat the pressure test:

1. Repeat step 1 to step 13 from “Performing the pressure test” on page 1-9.

2. If the source fails the pressure test, do not reconnect or start the solvent delivery system; contact Waters.See also: “Contacting Waters” on page 1-13.

If the source passes the pressure test, prepare the instrument for normal operation. See the next section.

Returning the instrument to normal operation

Required materialsChemical-resistant, powder-free gloves

To return the instrument to normal operation:

1. Disconnect the tubing leading to the laboratory exhaust port from the source exhaust valve’s outlet connection.

2. Disconnect the short length of tubing fitted to the source exhaust valve from the nitrogen exhaust waste bottle’s outlet connection.

3. Connect the free end of the tubing leading to the laboratory exhaust port to the nitrogen exhaust waste bottle’s outlet connection (so that the waste bottle’s outlet connects directly to the laboratory exhaust port).

Warning: If the source fails the pressure test, the integrity of its waste system can be impaired; do not reconnect or start the solvent delivery system; contact Waters.

Caution: You must remove the source exhaust valve after performing the source pressure test and before resuming normal instrument operation.

Warning: The solvent delivery and nitrogen exhaust waste line connections can be contaminated with biohazardous and toxic materials. Always wear chemical-resistant, powder-free gloves while performing this procedure.

4. Disconnect the pressure test meter tube from the tee-piece.

5. Fit the blanking plug to the tee-piece; ensure that it is pressed in firmly.

6. Connect the solvent delivery line to the probe.

7. On the MassLynx Tune window, click to start the gas flow.

8. Click Press for Operate, and confirm that the instrument status indicator shows green.

9. Start the solvent delivery system.

Warning symbols alert you to the risk of death, injury, or seriously adverse physiological reactions associated with an instrument’s use or misuse. Heed all warnings when you install, repair, and operate Waters instruments. Waters assumes no liability for the failure of those who install, repair, or operate its instruments to comply with any safety precaution.

Task-specific hazard warningsThe following warning symbols alert you to risks that can arise when you operate or maintain an instrument or instrument component. Such risks include burn injuries, electric shocks, ultraviolet radiation exposures, and others. Burst warning

This warning applies to Waters instruments fitted with nonmetallic tubing.

Mass spectrometer flammable solvents warning

This warning applies to instruments operated with flammable solvents.

Warning: (Risk of personal exposure to laser radiation.)

Warning: (Risk of exposure to biological agents that can pose a serious health threat.)

Warning: Pressurized nonmetallic, or polymer, tubing can burst. Observe these precautions when working around such tubing:• Wear eye protection.• Extinguish all nearby flames.• Do not use tubing that is, or has been, stressed or kinked.• Do not expose nonmetallic tubing to incompatible compounds like

tetrahydrofuran (THF) and nitric or sulfuric acids.• Be aware that some compounds, like methylene chloride and

dimethyl sulfoxide, can cause nonmetallic tubing to swell, which significantly reduces the pressure at which the tubing can rupture.

Warning: Where significant quantities of flammable solvents are involved, a continuous flow of nitrogen into the ion source is required to prevent possible ignition in that enclosed space. Ensure that the nitrogen supply pressure never falls below 400 kPa (4 bar, 58 psi) during an analysis in which flammable solvents are used. Also ensure a gas-fail connection is connected to the HPLC system so that the LC solvent flow stops if the nitrogen supply fails.

Mass spectrometer shock hazard

This warning applies to all Waters mass spectrometers.

This warning applies to certain instruments when they are in Operate mode.

Biohazard warning

This warning applies to Waters instruments that can be used to process material that might contain biohazards: substances that contain biological agents capable of producing harmful effects in humans.

Warning: To avoid electric shock, do not remove the mass spectrometer’s protective panels. The components they cover are not user-serviceable.

Warning: High voltages can be present at certain external surfaces of the mass spectrometer when the instrument is in Operate mode. To avoid non-lethal electric shock, make sure the instrument is in Standby mode before touching areas marked with this high voltage warning symbol.

Warning: Waters's instruments and software can be used to analyze or process potentially infectious human-sourced products, inactivated microorganisms, and other biological materials. To avoid infection with these agents, assume that all biological fluids are infectious, observe good laboratory practices and, consult your organization’s biohazard safety representative regarding their proper use and handling. Specific precautions appear in the latest edition of the US National Institutes of Health (NIH) publication, Biosafety in Microbiological and Biomedical Laboratories (BMBL).
