TitleInvestigation of the Enzymatic Effect of Materials on Hydrogen Peroxide Solution___________________________________________________________________________

Objective To investigate the enzymatic effect of various materials in the hydrogen peroxide solution.

ApparatusBeaker, test tubes, water bath of 95C and Bunsen burner.

MaterialsFresh liver, potato cubes, manganese dioxide, hydrogen peroxide, wood splints and parafilms.

Procedure1. Six fresh empty test tubes were labelled 1, 2, 3, 4, 5, 6 and were stood in a rack.2. A razor blade was used, the provided liver was cut into several pieces of roughly 0.8 cm x 0.8 cm x 0.5 cm.3. One piece of liver was placed into tube 1.4. The second piece of liver was placed into the bottom of tube 2. A wooden splint was used to gently spread the liver, without mashing it, over as wide an area as possible of the bottom of the test tube. Tube 2 was placed in the water bath of 95C for about five minutes.5. A weighing balance was used, two 0.5g portions of manganese dioxide powder were measured out and each portion was pour into tube 5 and 6.6. Tube 6 was put in the water bath for five minutes.7. Tube 2 and 6 were let cool after five minutes.8. The third piece of liver was put on a white tile. With a mortar or pestle, it was gently mashed into pulp. The pulp was scoop into tube 3.9. The potato cubes was cut into roughly 0.8 cm x 0.8 cm x 0.5 cm. One of the cube was placed into tube 4.10. Another six fresh empty test tubes were prepared and were stood in rack. 5 cm3 of hydrogen peroxide was put into each of the tube.11. Next, hydrogen peroxide was quickly added into test tube 1, 2, 3, 4, 5 and 6. 12. The parafilm provided was quickly stretched and seal the mouth of the test tubes by stretching the film over it. Step 11 and 12 should be done quickly.13. The test tubes were left for 20 minutes.14. After 20 minutes, a glowing splint was inserted into each tube one at a time by penetrating the parafilm with it.15. All the observations were recorded in Table 1. The observations on each test tube were immediately recorded after the reaction has started.
