Sherlock Q-FAMETM Sample Preparation Method

Q-FAME is the newest rapid identification method for the Sherlock®

Microbial Identification System (MIS), which has been optimized forclinical laboratories needing STAT processing of cultures. Q-FAMEtakes less than 25 minutes to identify aerobic bacteria from pureculture, often directly from the primary isolation plate.

The Sherlock® Microbial Identification System (MIS)

Over 750 clinical and environmental laboratories worldwide have usedthe Sherlock® MIS since 1991. The system identifies over 1,500bacterial and yeast species by analyzing the unique fatty acidfingerprints inherent within each species. Fatty acids are separatedfrom each microbe, converted to fatty acid methyl esters (FAMEs)and analyzed by Gas Chromatography (GC).

Sherlock automates all hardware operations and uses sophisticatedpattern recognition software to compare each unknown FAME profileto known FAME profiles for an identification. Additional software toolsallow for sample clustering, strain tracking, custom library generation,21CFR Part 11 compliance and DNA sequencing analyses.

Sherlock is fully automated with no GC experience required.

Benefits of Q-FAME vs. Other Aerobic Bacterial ID Systems

Rapid sample preparation and ID- The Q-FAME identification procedure takes less then 25 minutes to identify aerobic

bacteria from pure culture, at least 300% faster than other culture-based ID methods.

Reduced growing time required- The Q-FAME procedure often requires only 24 hours of growth from the primary isolation

plate, compared to 48 hours of growth required of other culture-based ID methods.-

Fewer bacterial cells required- The Q-FAME procedure uses ~7.0mg of cells, at least 40% fewer cells than other

culture-based ID methods.

Lower cost per sample- The Q-FAME procedure costs as low as $6.00 USD per sample, at least 60% lower cost

for consumables than other culture-based ID methods.

Standardized procedure for all aerobic bacteria- All isolates are prepared using the same procedure, so no upfront biochemical tests,

Gram stains or biochemical cards are required.

Benefits of Q-FAME vs. the Classical MIDI Extraction Procedure

Reduced turnaround time (TAT)- The TAT for one sample is reduced from 120 minutes to 24 minutes, thus making

Q-FAME more amenable to clinical STAT processing of samples.

Less corrosive reagents- Q-FAME reagents are less corrosive and MTBE, a ground water and soil contaminant, is

no longer a reagent used within the extraction procedure.

No more water baths or laboratory shakers- Q-FAME does not require any of these pieces of equipment.

Standard media and growth temperatures used- Clinical Library: Bacteria typically grown on TSA Agar with 5% defibrinated

sheep blood at 35C.- Environmental Library: Bacteria grown on TSA Agar at 30C.

Improved Entries with updated taxonomy- Q-FAME library entries were developed with up-to-date taxonomy.

Reduced wear on the GC Liner- The Q-FAME extract is cleaner and GC liners last twice as long as before.
