MAJOR RESEARCH DIRECTIONS

The Microfabricated Tissue Models Lab (mtm.uic.edu) focuses on engineering the microenvironmental cues around human cells towards stabilizing their long-term phenotype in vitro for key applications in drug development (e.g. drug metabolism/toxicity screening and disease modeling for novel drug discovery) and regenerative medicine (i.e. cell-based therapies). We develop and utilize microfabrication tools, such as protein micropatterning, high-throughput extracellular matrix (ECM) microarrays, and microfluidic platforms to precisely control the microenvironment of both primary and iPSC-derived human cells in 2D and 3D culture formats depending on the hypotheses being posed. We utilize our model systems to mimic key aspects of diseases in vitro and elucidate underlying molecular mechanisms of disease progression as a function of cell-cell, cell-ECM, and cell-soluble factor interactions. Our organ of interest has been primarily the liver (e.g. drug-induced liver injury, non-alcoholic fatty liver disease and hepatitis B virus infection) and we have several publications in that space that are actively being cited in the field. More recently, we have expanded our repertoire to cardiomyocytes to study the underlying genetic determinants of atrial fibrillation in close collaboration with leading cardiologists and are also pursuing an intestinal

culture platform to interface it with the liver for addressing key disease questions where both organs interact in the body. We are well funded, mostly through the National Institutes of Health (NIH) and National Science Foundation (NSF), to pursue the research directions above. Our long-term goals are to a) disseminate our engineered platforms to the wider academic and pharmaceutical communities for use in drug screening and mechanistic investigations, b) make fundamental discoveries into how underlying genetics and cell-microenvironment interactions dictate worsening of non-alcoholic fatty liver disease, hepatitis B virus infection, and atrial fibrillation outcomes in different patient populations to varying degrees, c) discover new molecular targets for the aforementioned diseases against which new drugs can be developed, and d) apply our cell culture expertise in vitro to implantable human tissues (i.e. biomanufacturing) for patients suffering from end-stage diseases and organ failure.
