Principle of ELISA technology

•The antibody is attached to the surface of the microtitre plate

•Sample and conjugate (Enzyme labelled antigen) are added simultaneously

•These compete for free binding sites on the capture antibody

•The quantity of enzyme labeled antigen bound will determine the resulting colour produced

•Optical density measures the amount of light absorbed within the solution to give a concentration of residue

Preparation of

AM IA simple dilution is completed using provided aqueous buffers such as diluted wash buffer

1.Weigh out 1g of honey

2.Add 19mls of diluted wash buffer

3.Place on a roller for 10 minutes until dissolved

• Conjugate – Known Analyte labelled with Horse Radish Peroxidase (HRP) Enzyme

• HRP – Reacts with Luminol and Peroxide to give off a light reaction

• Light signal inversely proportional to antigen concentration
