Abstract
Catalytic enantioselective formation of covalent bonds is a central theme in modern
organic chemistry. Among the many methods for the synthesis of enantiomerically enriched
compounds, transition metal-catalyzed asymmetric allylic substitution stands out due to its
power and versatility. Iridium-catalyzed allylic substitution in particular has recently received
much attention due to its branched regioselectivity, allowing access to the chiral scaffolds that
are difficult to prepare using the well-developed palladium catalysts. This thesis describes the
development of a number of novel enantioselective carbon−carbon bond forming reactions as
well as spiroketalization, catalyzed by a chiral iridium−(phosphoramidite,olefin) complex.
Branched racemic allylic alcohols underwent enantioselective allylic substitutions with
potassium alkenyl- and alkynyltrifluoroborates, allylsilanes, and unactivated alkenes (Scheme
I). The corresponding 1,4-dienes (I),1,4-enynes (II), and 1,5-dienes (III and IV) were
obtained with excellent enantioselectivities (up to >99% ee). Judicious choice of acid
promoter and careful optimization of other reaction parameters were crucial for achieving high
yields and selectivities. Allylic vinylation using potassium alkenyltrifluoroborates was
achieved by employing aqueous hydrofluoric acid and tetrabutylammonium hydrogen sulfate,
which serves the dual role of phase transfer catalyst and acid promoter. Asymmetric allylic
substitution using potassium alkynyltrifluoroborates was accomplished using an improved set
of conditions, involving in situ generation of hydrofluoric acid, which minimizes the handling
of the hazardous mineral acid. In addition, allylsilanes efficiently underwent enantioselective
coupling with the catalytically generated allyliridium intermediate using Sc(OTf)3. The
application of unactivated alkenes as nucleophiles necessitated rational design and use of
diarylsulfonimide (P1) and 4Å molecular sieves for high efficiency. Each of the developed
methods was utilized as the key step in asymmetric syntheses of biologically active natural
products ((−)-nyasol and (−)-hinokiresinol), drug candidates (AMG 837 and JNJ-40418677),
and an agrochemical ((−)-protrifenbute).

Subsequently, asymmetric allylic substitution of allylic carbonates with non-stabilized
alkyl nucleophiles bearing functional groups was developed (Scheme II). Functionalized
alkylzinc bromide reagents were employed to deliver the coupled products V incorporating
convenient synthetic handles with excellent enantioselectivities. The synthetic utility of the
developed methodology was demonstrated through the synthesis of (−)-preclamol, a drug
candidate for the treatment of schizophrenia.

The last chapter of this thesis documents the development of a method for iridium-
catalyzed enantio- and diastereoselective spiroketalization. In the presence of the chiral
iridium−(P,olefin) complex, various ketols VI cyclized to give spiroketals VII as exemplified
in Scheme III. By exploiting the ketol-hemiketal equilibrium and the configurational
preference of the spiroketal center due to anomeric stabilization, a single chiral catalyst
effectively controlled the configurations of two stereogenic centers.

Zusammenfassung
Die katalytische, enantioselektive Bildung kovalenter Bindungen ist ein zentales Thema
der modernen organischen Chemie. Zwischen der Vielzahl bekannter Methoden für die
Synthese chiraler Verbindungen steht die Iridium-katalysierte, asymmetrische, allylische
Substitution dank ihrer Potenz und Vielfältigkeit heraus. Ihr wird besonders viel
Aufmerksamkeit geschenkt, da sie hohe Selektivität für verzweigte Produkte aufweist und
somit die Darstellung chiraler molekularer Grundgerüste erlaubt, welche mit den weiter
verbreiteten Palladiumkatalysatoren nicht zugänglich sind. Diese Arbeit beschreibt die
Entwicklung mehrerer, neuer Methoden für die enantioselektive Bildung von C-C Bindungen
ebenso wie eine Spiroketalisierung, allesamt katalysiert durch einen Iridium-
(Phosphoramidit,Olefin) Komplex.
Verzweigte, racemische Allylalkohole wurden in enantioselektiven, allylischen
Substitutionen mit Kalium Alkenyl- und Alkinyltrifluoroboraten, Allylsilanen und einfachen
Olefinen umgesetzt. Die entsprechenden 1,4-Diene (I),1,4-Enine (II), und 1,5-Diene (III und
IV) wurden mit exzellentem Enantiomerenüberschuss erhalten (bis zu >99% ee). Eine
wohlüberlegte Wahl der Säurepromoter und sorgfältige Optimierung der
Reaktionsbedingungen waren ausschlaggebend für das Erreichen guter Ausbeuten und
Selektivitäten. Allylische Vinylierung mittels Kaliumalkenyltrifluoroboraten konnte durch den
Einsatz von wässriger Flusssäure und Tetrabutylammoniumhydrogensulfat realisiert werden,
wobei letzteres sowohl als Phasentransferkatalysator als auch als Säurepromoter dient.
Asymmetrische, allylische Substitution mit Kaliumalkinyltrifluoroboraten wurde durch
verbesserte Reaktionsbedingungen bewerkstelligt, mit in situ Erzeugung von Flusssäure
wodurch die Handhabung der gefährlichen Mineralsäure minimiert werden konnte.
Allylsilane wurden mit dem katalytisch, unter Verwendung von Sc(OTf)3 generierten
Allyliridiumintermediat effizient gekoppelt. Das Einsetzen von einfachen Olefinen als
Nukleophil erforderte rationales Design und den Einsatz von Diarylsulfonimid (P1) und 4Å
Molekularsieben, um hohe Effizienz zu erreichen. Jede der entwickelten Methoden wurde in
einem Schlüsselschritt für die asymmetrische Darstellung der biologisch aktiven
Naturprodukte ((−)-Nyasol und (−)-Hinokiresinol), potentiellen Arzneimittel (AMG 837 und
JNJ-40418677) und von der Agrochemikalie ((−)-Protrifenbute) eingesetzt.

Anschliessend wurde die asymmetrische, allylische Substitution von Allylcarbonaten mit
nicht-stabilisierten Alkylnukleophilen entwickelt (Schema II). Funktionalisierte
Alkylzinkbromidreagenzien wurden für die Darstellung asymmetrischer Kopplungsprodukte
V eingesetzt und vereinigen so praktische Synthesebausteine mit exzellenter
Enantioselektivität. Der synthetische Nutzen der Methode konnte durch die Darstellung von
(−)-Preclamol demonstriert werden, ein Arztneimittelkandidaten für die Behandlung von
Schizophrenie.

Das letzten Kapitel dieser Arbeit beschreibt die Entwicklung einer Iridium-katalysierter
Methode für eine enantio- und diastereoselektive Spiroketalisierung. In der Gegenwart des
chiralen Iridium−(P,olefin)-Komplex‘ wurden diverse Ketole VI zu Spiroketalen VII
zyklisiert (Schema III). Das Gleichgewicht zwischen Ketol und Hemiketal sowie die durch
den anomeren Effekt beinflusste, Konfigurationstendenz des Spiroketalzentrums wurden
genutzt, um mit einem einzigen, chiralen Katalysator effizient die Konfiguration zweier
Stereozentren zu setzen.
