    Nestled in the heart of a serene and scenic campus, the Plant Tissue CultureLaboratory stands out as an exemplary temple of learning and research.The layout Plant Tissue Culture Laboratory Unit encompasses science,art and aesthetics- which quite aptly reflect the activities which go on insidethe laboratory. The first thing that anyone who visits the Plant TissueCulture Laboratory at GKVK, is the beautiful landscape of the laboratory.Situated right across a century old Heritage Banyan tree, the set upmanages to imply that both nature and technology can coexist in tandem with

    the thoughtful usage of technology, the world will be able to cope up with thehuge advances in science which can be mind boggling at times.

    In an expansive campus of about 2000 acre, this laboratory is preceded by itsrepute for sporting one of the well created Theme Gardens. The themeof this garden is rightfully associated with the Concept of Tissue Cultureand Biotechnology and brought forth by some of the structures and thelayout of the entire garden.

    One of the most eye catching sculptures is the sculpture of Dr. GottliebHaberlandt who for his findings in plant tissue culture and for thedevelopment of the concept of totipotency has rightly been called as theFather of Plant Tissue Culture. This sculpture also happens to be thesecond sculpture of Dr. Haberlandt in the whole world. The first being theone in his native, Austria.

    The other sculpture which adds life to the theme of the garden is the sculpturewhich gives deeper insight into plant tissue culture and the concept oftotipotency. In this sculpture, one will be able to see stubble of the felledtree being able to sprout new shoots indicating that any cell in a plant hasthe ability to regenerate into a whole plant. The belief that life finds its owncourse and way of survival is best depicted by this masterpiece by a mastersculptor, Mr. Jaganath Jakkepalli

    The other works of art which one can see is that of Murashige and Skoog.The endeavor of this teacher-student duo has helped in the creation of thehighly popular nutrient culture medium - MS media. As MS media is therequisite for most works in plant tissue culture, their works are honorablyacknowledged through this dedicated piece of art. The subtle implication ofthe teacher pupil relationship is depicted, as the two are seen having a deepintellectual conversation over a cup of tea. The sight upholds the attentionof all the viewers, conveying that great thoughts sprout only when theProfessor is deeply and sincerely committed to his profession and passion ofpursuit, while sharing a healthy relationship with the student.

    The roof of the Plant Tissue Culture Laboratory, both literally andpractically, is supported by a DNA double Helix structure which was firstdiscovered by Watson and Crick. The sculptures of these two remarkablygreat visionaries of science, which are quite smaller than the structure of thedouble Helix and seen looking at their discovery with an appreciative eye,marveling at the fact that their discovery has attained such an iconic status inthe history of life sciences.

    The Plant Tissue Culture Laboratory unlike any other laboratory yearns tosubtly bring out the messages about education, science, ethics, aesthetics,and art.

    About Plant Tissue Culture Laboratory

    Unlike the other branches of agriculture, horticulture is a branch which dependson the planting material in greater magnitude. The perennial nature and theexpensive cost of cultivation make it very essential to have quality plantingmaterial which is true to type. For increased vigor and protection for viraldiseases, the planting material for most of the high value horticulture crops areseldom taken from the field but are produced in vitro with the help of planttissue culture.The main function of our plant tissue culture laboratory is to produce qualityplanting material and also involve ourselves in the research aspects of planttissue culture. The production and research happen simultaneously and iscarried out by dedicated scientist, teachers, and lab assistants as well as by the

This is also the only laboratory in the country which is associatedwith a University which has the production capability in such large scales.Even though in Bangalore, there are numerous private companies involved inthe production of planting material for horticulture crops, there is still ashortage of quality planting materials. One particular indigenous and fastdisappearing variety of banana- Nanjangud rasbale is one thing that manyfarmers are not able to get in the outside market. Our plant tissue culturelaboratory is one of the only places where this variety is available. The mainobjective of this farmer centric laboratory is to make high quality tissue culturedplanting material available and affordable to the farmers and meet the existingdemand for the planting materials. The farmers also have a sense of trust withthe University and hence prefer to get their saplings from the Plant tissueculture laboratory of the university.The other aspect on which our plant tissue culture laboratory is working on iscost efficiency. With a dedicated team of scientist and students working on themeans to reduce cost of production of these tissue-cultured planting materials,the University laboratory aims to set bench mark prices and make the use oftissue cultured materials economically feasible. Our plant tissue culturelaboratory is trying to achieve the status of being an interface between thefarmers community and the corporate companies involved in the production ofplanting material.One of the other significant functions that our plant tissue culture laboratorytries to achieve is to make Universities such as ours self sustainable. Some of them are: Banana ( Grand Naine, Red Banana, ElakkiBanana and Nanjangud Rasbale), Pomelo, Carambola, Strawberry, Coleus,Patchouli, Brahmi, Stevia, Anthurium and Orchids.

    Plant Tissue Culture Laboratory Commercial Plant ProductionThanks to the Govt. of India initiative, our laboratory has received substantialfunds to strengthen the laboratory in order to use it for commercial plantproduction along with post graduate research. We are happy that we are ableto develop our lab which has a capacity to produce a minimum of one millionplants per year. Though we are not quite satisfied with our progress, with manyteething problems, which goes with running a production unit under agovernment set up we are yet to achieve a target of 1 lakh plants. We havealready sold about 50,000 banana plants earning around Rs 5 lakh as on today.One of the major constraints we have faced is the constant erratic powersupply. Though we do have a generator, the maintenance and running of thegenerator itself is a challenging task. Never the less, we take pride in letting youknow that ours is one of those promising university laboratories in the countrytoday. We are a highly determined lot here in the laboratory committed toserving the farming kind as well as entrepreneurs and hence a considerableprogress is being achieved in spite of many setbacks in managing the laboratory.Its highly contenting that many farmers who have grown our plants are comingback to us for some more as they have reaped the benefits of our qualityplanting material. The comments of our visitors are a constant tonic in boostingus to deliver more and more. Some of the pictures which have been loaded onthis website are a testimony to this.

    FacilitiesThe research and production of the tissue cultured planting materials isenabled by the state of the art laboratory. This well structured and well keptlaboratory makes working here a truly enjoyable experience with a floor spaceof 1600 sq ft. , separated in to 2 divisions- the R&D unit and the Productionunit.Production Unit:

    The media preparation and sterilization room is well equipped withincubators, centrifuges, autoclaves, Quartz distillation units and BODincubatorsThe culture transfer room uses advanced and cutting edge florescencemicroscopes and laminar air flows to transfer explants in to the sterilizedmedia.Bottles containing media with explants are then transferred to the growthroom which can hold 30,240 culture bottles with an estimated productioncapacity of 5 lakh plants/year.Tunnel house for primary hardening of the regenerated plants.Green house where the saplings after primary hardening are transferredto for secondary hardening.

    Achievements

    With a highly qualified set of staff, dedicated researchers and InspireScholarship holding students working in the plant tissue culture laboratory, it isnot surprising to see that along with the work done; even the knowledge of thepeople in the laboratory is gradually growing. There is some new finding ordeeper understanding in the field of plant tissue culture almost every day.People working and learning in this laboratory are evolving by the day.Some of the noteworthy findings of this laboratory are:

    National award winning break-through in cost reduction of tissuecultured Banana plantlets by utilization of alternative carbon sources,gelling agents, matrices, water sources and light.Similar achievements in production of Potato and Philodendronplantlets.First report on callus induction in Ashoka (Sara ca indica)Reported occurrence of somatic embryogenesis in sweet type carambola,for the first timeFirst successful attempt in obtaining in vitro shoot proliferation in solidbamboo

    In vitro colchicine production from callus cultures of Gloriosasuperba , animportant medicinal plant was reported for the first time.

    The plant tissue culture laboratory has also developed several protocols whichcan be used by the production unit of the laboratory and also production unitsof other corporate companies. Some of the protocols are:

    In Jackfruit elite cultivar Singapore jack, a protocol formicropropagation was developed.In Nanjangud Rasabale, a premier variety of Banana in Karnataka, anattempt is on to increase the frequency of plant production throughmicropropagation, a preliminary work to develop protocol forsomaclonal variation is under progress.A protocol for mass multiplication of Banana Cv. Dwarf Cavendish,Robusta and Grand Naine was developed earlier. An estimation ofyield potentialities of these tissue-cultured plants has also been done.In Strawberry, a protocol for mass multiplication of cultivars Sujathaand Labella was developed.A good protocol has been developed for direct regeneration of plantsfrom nodal segments of C itrus latifolia .

    In Gloriosa, a highly valued medicinal as well as an ornamental crop, aprotocol for micropropagation was developed.In Coleus, a protocol for micropropagation, callusing, somaticembryogenesis was developed.     In Catha edulis (Kat), an exciting medicinal crop of Yemen, work ontissue culture and a detailed study on varietal distribution and variancethrough genetic markers is under progress.In Potato, a protocol for In vitro tuberisation and andmicropropagation was developed. A noteworthy achievement inpotato has been the use of Tapioca as a gelling agent instead of agar.Tapioca source is found to be a cheapest source, thus facilitating asubstantial decrease in the cost of tissue culture production of plants.In Cucumber, a tissue culture a protocol was developed keeping inview of its use in aiding breeding of cucumber for seedlessness.In Curry leaf a protocol was developed for micropropagationIn Chilli, exhaustive work was done to study callusing. The work has abearing in future research towards varietal development in vitro. Thevarieties used for the study include famous Byadagi dabba and Arkaseries.In Anthurium Cv. Ceylon Crinkle Red, a regeneration protocol wasdeveloped and a protocol for seed germination was also developed.

    In Carnation, in an exhaustive study, various methods of direct andindirect regeneration protocol were developed. A varietal evaluation inregard to response to callusing was studied in detail.In Dendrobium Orchid, Cv. Jo Mutant, a protocol for in vitroproliferation was developed. A high frequency proliferation has beenachieved. This work has a commercial bearing.In Gerbera, a protocol for micropropagation of some elite cultivarswas established.In Solid Bamboo (Oxytenenthera stocksii) for the first time, in vitroshoot proliferation has been reported.Induction of callus from matured leaves and petiole segments in jasmineIn sweet type Star fruit (Carambola), for the first time a protocol fordirect and indirect somatic embryogenesis has been developed.

    period of 7 months, it can be assured that laboratory will be successful infulfilling the goal.The Research and Development Unit:The world of plant tissue culture and biotechnology is a relatively new andunexplored one. The findings in this field seem to crop up almost every otherday. Research on plant biotechnology and plant tissue culture happens inthe plant tissue culture laboratory almost all through the year. Thelaboratory is given various projects to work on and the research findingsgenerated by this lab every year reveals the quality of the work done here.

    Presently there are numerous projects and research topics which arebeing worked upon, which include:

    Attempts taken up to standardize efficient protocol for rapid clonalmultiplication and To breed variants with high stevioside contentHardening of Stevia has been a problem with over 80% mortality.Experiments done have given plants which are stocky (3-4 times thicker ingirth). These plants should survive better under conditions of ex vitrohardeningStandardization of protocol for direct and indirect regeneration andsomatic embryogenesis, Inducing variation using physical and chemical
