TemperatureMeasurement)andThermocouplesinaLyostartodevelopa suitable lyocycle in the development scale to support formulationdevelopment. After formulation development phase TEMPRIS® wasadditionally applied in the Lyostar to perform lyophilization cyclerobustness testings in the development scale to increase processunderstanding and to cover unexpected and unpredictable cycledeviationsduringmanufacturing.Theproductwasalsostressedabovethe known critical temperature to understand the impact to be ableto predict possible difficulties in the production scale. Furthermorea prediction of minimal cycle time in a commercial freeze dryer wasperformed.Thedevelopmentteambasedonthisenhancedproductandprocessunderstandingtogetherwiththeexpertteamoftheproductionscalefreezedryerdiscussedandmutuallyagreedontheparametersofthefirstcommercialscalelyophilizationrunwherethescalewastakenfrom approx. 430 vials on 3 shelves to approx. 45.000 vials on 18shelves.TEMPRIS®wasusedtomigitatetheriskofscaleupandhadbeen established at the production scale freeze dryer before the firstscaleuprunwasundertaken.Experiencesandresultsofthiseffortwillbepresented.
