0. Safety

• Check the rotor, bucket, and tube are free of scratch and crack before operation.• Check the chemical resistance chart(Refer to Appendix3&4) and do not use any samples inapplicable to the tubes and accessories.• Do not use any samples that may generate volatile or explosive vapor.• Always use the same type of tubes. (weight, material,density and volume.)• Do not exceed the maximum rated speed of the rotors, buckets and tubes in use.• The maximum speed of the rotor may vary depending on the device or tube or Adapter.• Make sure that there is no foreign material in the chamber.• Be careful not to load the rotor with too much force.• Firmly tighten the rotor onto the motorshaft.• Remove the rotor from the chamber after centrifugation.• After using the rotor, remove the lid from the rotor body. Keep the rotor upside down.• Do not put the rotor in the boiling water for sterilize. • There is a rotor ID. magnetic in the rotor drive hole which is located on the bottom of the rotor. Make sure not to damage the rotor ID. magnetic when handling the rotor. • Only use rotors, parts, and accessories provided by GYROZEN. GYROZEN. is not responsible for damages of the device and accidents caused by using parts and accessories not recommended.• Inspect the rotor and accessories each time after using them. If you find any abnormality, stop using them and contact GYROZEN.• It is your responsibility to sterilize and/or decontaminate the centrifuge and accessories before requesting service by GYROZEN.• Maximum speeds are based on a solution density of 1.2 g/mL in all rotors

Rotors and accessories are may damaged in repeated use or careless handling. And damage may be worsened under centrifugal forces. Therefore even a small defect on the rotor or accessories may cause personal injury and instrument damage. Carefully read and understand this manual.

1. Rotors

Fixed angle rotor holds tubes at an angle to the axis of rotation. It is designed of the aluminum material, so fixed-angle rotor can withstand much higher gravitational forces than swing-out rotor. It is useful for pelleting subcellular particles and remove particles from a suspension and discard the debris, and isopycnic banding of DNA.

Swing-out holds tubes in the buckets(or Adapters) which swing horizontally.It is useful when samples are to be resolved in density gradients. The longer path length permits better separation of individual particle types from a mixture.

5.3 Tube/Bottle Sterilization

- It is generally recommended to fill more than 80% of the the tube or bottle. However, there is an exception that must be filled to the end of the container to prevent damage. Thus the minimum and maximum filling volumes should be different in accordance with the manufacturer's guidelines.

- There is a risk of tube breakage when filling volume of sample is too less compared to the tube capacity. In this case, use a tube and adapter that meet the sample volume.

(*1) After run, make sure that the sample is not leaked to the rotor hole. If the rotor has been contaminated by aggressive chemicals, immediately clean it by means of a mild cleaning agent.

Check the rotor as follows after or before every use.

1. Make sure that the rotor thread is free of wear .

3. Make sure that there is no cracks or other signs of damage on the rotor body.

2. Make sure that there is no foreign objects in the rotor holes.(*1)

4. Make sure that there is no foreign objects on the drive hole.

5. Make sure that there is no foreign objects on the motor shaft.

Rotors and accessories are may damaged in repeated use or careless handling. And damage may be worsened under centrifugal forces.

Clean the rotor immediately if salts or other corrosive materials are leaked.

When foreign objects adheres to the rotor

1. Wash the rotor with a mild detergent(pH 5-9), using a soft brush.2. Rinse thoroughly with distilled water. 3. Wipe off the moisture with a soft cloth and dry the rotor upside down

Soak the rotor in warm water for a while, then gently remove foreign objects with a soft brush.

[Angle Rotor]After drying the rotor, spread the grease lightly on the rotor threads.[Swing-out Rotor]After drying the rotor, spread the grease lightly on the rotor pivot.

7.1 Rotor

Problem Solution

Lid can not be tightened - Check that the rotor threads are greased.- Check the rotor threads and motor shaft for foreign matters.

Rotor is discolored - When cleaning the rotor, it may appear discolored if not rinsed thoroughly. Rinse the rotor thoroughly with water.- Anodizing may be worn. Contact GYROZEN.

Rotor ID. is not identified Check the drive hole for foreign matters.

Noise and vibration duringrunning

Please re-check the coupling status of the following three matches tominimize the noise.1. the balanced way of coupling of the rotor into the motor shaft.2. the completeness of fixing of the rotor locking nut on the rotor.3. the matching status of rotor lid with the rotor.

Sample leaks out

- Close the tube cap tightly.- Tube volume exceeds maximum filling volume. Refer to the tube manufacturer’s guideline for tube filling volumes.

Tube is cracked

- Tubes may crack if they are used below their lower temperature limit or higher g-force. Refer to the tube manufacturer’s guideline for temperature limit and Max. g-force.- Repeated use of tube may cause cracks. Dispose of cracked tubes.

Tube is broken

- Sample volume is too low to support tube wall. Refer to the tube manufacturer’s guideline for tube volumes.- Tubes run above their rated speed. Refer to the tube manufacturer’s guideline for maximum speed of tubes.

Centrifugation Separations

The most common centrifugation method is pelleting.Pelleting separates particles of different sedimentation coefficients.During centrifugation, the largest particles in the sample traveling to the bottom of the tube first.Differential centrifugation is the successive pelleting of particles from previous supernatant of decreasing sedimentation velocities, using increasingly higher forces and/or long run times.Through centrifugation two fractions are gained. A pellet and a supernatant of unsedimented material.The supernatant can be re-centrifuged at higher speeds to get a new pellet and supernatant.

Density gradient Centrifugation

Samples are layered on top of the density gradient. Under centrifugal force, particles sediment through the gradient column in zones. Each zone consisting of size, sedimentation rate of particles.When applying the sample to the density gradient, insert it carefully so that it does not mix with the density gradient.The centrifugation should be stopped before any of the separated zones reach the bottom of the tube.

Isopycnic gradient separates particles on the basis of density rather than size, independent of time.Each particle sediments in the tube where the gradient density is equal to the particle density.Unlike Rate-zonal centrifugation method, density gradient and the sample are mixed in the tube.

Density gradient Centrifugation permits for the complete separation of several or all components in a sample. Tube is partially filled with a solution having a decreasing density from the bottom to the top of the tube. This method is mainly suited to the viruses.There are two methods in which density gradients may be used. : Rate-zonal and isopycnic.
