The lab is conducting research in the interdisciplinary area of chemical biology, applying concepts from organic chemistry to develop new chemical tools for the investigation of biological problems. Research focuses on the targeted chemical synthesis of new artificial biomolecules (amino acids, proteins, nucleotides, oligonucleotides) tailored to investigate and manipulate complex cellular processes in in vitro and in vivo biological systems. The PhD project will be positioned at the intersection of Organic Chemistry and Chemical Biology and is part of the DFG funded priority programme SPP1623 (Chemoselective reactions for the Synthesis and Application of Functional Proteins). It will deal with the development of new light-induced protein-protein and protein- DNA crosslinking reactions. In particular the PhD student will synthesize unnatural amino acids with reactive moieties and specific crosslinkers. The unnatural amino acids will be incorporated site-specifically into proteins in bacteria and mammalian cells via genetic code expansion (Lang et al. Nature Chem. 2012, 5, 298, Lang et al. JACS 2012, 134, 10317, Lang and Chin, Chem. Rev. 2014, 114, 4764). Thus modified proteins will be used to elucidate and study protein-protein and protein-DNA interactions with high spatio-temporal resolution in live mammalian cells. During their PhD, candidates are expected to develop a proof knowledge and practical skills in organic chemistry, chemical biology and standard molecular biology. Candidates should hold a Master’s degree in Organic Chemistry or Chemical Biology with a strong background and solid hands-on experience in synthetic organic chemistry. Experience in protein biochemistry and standard molecular biology techniques will be beneficial. A strong interest in chemical biology oriented research is also essential.
