Robust amplification even with high GC amplicons

Amplification of two human genomic amplicons of mid to high GC content.All reactions were conducted using 30 cycles of amplification and visualizedby microfluidic LabChip® analysis. All polymerases were cycled according tomanufacturer’s recommendations. For the 78% GC amplicon, GC Buffers orenhancers were used when supplied with the polymerase.

OLIGONUCLEOTIDE PROBES

XGen Lockdown Probes are individually synthesized 60ñ120 nu-

cleotide 5í-biotinylated capture oligonucleotides designed to enable

cost-effective, high-depth sequencing, and analysis of speci� c re-

gions of the genome. xGen Lockdown Probes are ideal for target

enrichment and custom panel development, and capture sets can

be easily expanded. Available at three different scales (mini ñ 2

pmole, standard ñ 20 pmole, and XL ñ 200 pmole), users can design

their initial capture panels using the mini scale and extend them by

progressively supplementing with more probes. Whether used with

custom capture panels, or for supplementing existing panels, xGen

Lockdown Probes provide approximately 5,000-fold enrichment with

high coverage over targeted regions, and minimal GC bias. Further-

more, the probes can be added to array-derived RNA baits to en-

hance coverage of GC-rich targets, such as � rst exons. Users can

therefore improve capture performance by rescuing drop-out regions

of array-derived exome capture, improve the uniformity of coverage,

discover additional genetic mutations, or improve coverage of spe-

ci� c regions of interest without redesigning a new panel.

AUTOMATED CELL COUNTER

The TC20 automated cell counter is the next generation of the TC10

automated cell counter which provides accurate and reliable counts

of live mammalian cells in 30 seconds. The TC20 systemís improved

lens and cell counting algorithm make it compatible with a broader

range of cell sizes and types. This includes cells smaller than 10 �m

and complex samples such as stem cells and primary cells derived

from tissue or blood. Many of the advanced features of the TC10

system, such as the auto-focus technology that provides highly re-

producible cell counts with reduced user-to-user variability, remain

in the TC20 system. The TC20 also has the ability to analyze cells

across multiple focal planes, which eliminates live cell undercount-

ing associated with single focal plane analysis. And unlike electrical

impedance-based counters, researchers can view cells in real time

to immediately con� rm their total and live/dead cell counts.

CELL ADHESION ASSAYS

A new suite of cell adhesion assays have been designed to evaluate

optimal cell attachment to extracellular matrices and assess factors

that in� uence cell-matrix interactions. CultreCoat Cell Adhesion As-

says are precoated and ready-to-use in a black stripwell microplate

format that minimizes background, allows greater sensitivity, and

affords � exibility in the number of samples assessed. The stripwell

feature also allows multiple experiments to be conducted simulta-

neously using the same kit. Calcein labeling allows direct compari-

sons between the number of cells that are loaded and the number

that adhere. Controls are provided for determining background and

nonspeci� c binding. The new adhesion protein array kit enables

researchers, on a single microplate, to assess adhesion on

Basement Membrane Extract (equivalent to Matrigel), Laminin,

Collagen I, Collagen IV, Fibronectin, and Vitronectin.

SPE SAMPLE PREPARATION SYSTEM The Microlute solid-phase extraction (SPE) sample preparation sys-

tem provides a faster, trouble-free alternative to cartridges for high

throughput sample cleanup. Handling samples as small as 150 uL,

the Microlute SPE sample preparation system helps increase assay

sensitivity by providing preinjection cleanup and concentration. As a

complete package, including � lter plate, collection plate, waste tray

and manifold, the Microlute SPE sample preparation system offers

all the advantages of automated and high throughput SPE sample

preparation in a convenient microplate format capable of rapidly

processing 96 samples in one go repeatedly and precisely. Using a

MULTI-ION-BEAM TOOL The ORION NanoFab is the � rst multi-ion-beam tool based on Gas Field Ion Source (GFIS) technology. As a major enhancement to the existing helium ion microscope, ORION NanoFab also

utilizes neon ions. The system is therefore capable of providing a complete sub-10 nanometer

nanofabrication and sub-nanometer imaging solution for industry, government, and academic

research laboratories. An optional gallium focused ion beam (FIB) column can also be integrated.

The neon ion beam offers precise machining and nanofabrication capabilities due to higher sputter yields in ion beam milling and faster resist exposure in ion beam lithography. The helium ion
