The International Cooperation for Convergence of Technical Requirements for the Assessment of 9

Feed Ingredients (ICCF) was launched in 2017 and aims to develop and establish common guidance 10

documents to provide technical recommendations for the assessment of feed ingredients, including new 11

uses of existing feed ingredients. 12

This guidance document has been developed by the appropriate ICCF Experts Working Group 13

and was subject to consultation by the Parties, in accordance with the ICCF Process. 14

1. INTRODUCTION 51

1.1. Objective of the Guidance 52

This document provides guidance regarding the requirements to be included in application 53

for pre-market approval or authorization for feed ingredients with regards to their 54

manufacturing process and specifications. 55

The required information is an important part of assessing safety during a feed ingredient’s 56

pre-market approval or authorization process. It also supports the identification of the feed 57

ingredient. This guidance document has been developed with an international team of experts 58

and considers the best practices for the provision of meaningful data and information. 59

While the guidance document supports the acceptability of the types of data and 60

information to be provided, applicants are advised to consult the appropriate regulatory 61

authorities and their guidelines during the development phase of new feed ingredients or a new 62

use of an authorized ingredient. This will help to determine whether the information provided 63

is acceptable or needed for a specific pre-market approval or authorization. 64

1.2. Definitions 65

The following definitions apply solely in the context of this guidance document: 66

Active substance1: Any substance in a feed ingredient that contributes to its intended 67

effect. 68

Batch: An identified quantity of a feed ingredient having uniform characteristics, with 69

specified limits and is produced from the same cycle of manufacturing production. 70

Microbial Biomass: The result of a fermentative process. It may include the total product 71

of the fermentative step or the by-product of fermentative steps, where the active substance 72

has been removed for further steps or to produce the ingredient market formulation(s). 73

Carrier: A feed ingredient or water used to facilitate the handling of the feed ingredient 74

under assessment and its incorporation into ingredient market formulations, premixtures, feeds 75

or water. The use of a carrier does not alter the feed ingredient’s intended effect and purpose. 76

Contaminant2: Any substance not intentionally added to feed , which is present in such 77

feed as a result of the production, manufacture, processing, preparation, treatment, packing, 78

packaging, transport or holding of such feed, or as a result of environmental contamination. 79

Feed (Feedingstuff)3: Any single or multiple materials, whether processed, semi-processed 80

or raw, which is intended to be fed directly to animals. 81

Feed ingredient3: A component part or constituent of any combination or mixture making 82

up a feed, whether or not it has nutritional value in the animal’s diet. Ingredients are of plant, 83

animal, microbial or aquatic origin, or other organic or inorganic substances. 84

Hazard Analysis and Critical Control Points (HACCP) Programme3: A system which 85

identifies, evaluates, and controls hazards which are significant for feed safety. 86

Ingredient market formulation: The feed ingredient under assessment formulated with 87

carrier(s) and/or other feed ingredient(s). It is the commercial product used to incorporate the 88

feed ingredient under assessment into premixtures, feeds or water. 89

Materials4: Substances, including raw materials and other inputs (excluding processing 90

aids and carriers) used for the manufacturing process. (see flow chart). 91

Purity: Concentration or other quantitative measurement of the active substance in the 92

feed ingredient. 93

Processing Aid5: Any substance, not including apparatus or utensils, and not consumed as 94

a feed ingredient by itself, intentionally used in the processing of materials, feed or feed 95

ingredient, to fulfil a certain technological purpose during treatment or processing and which 96

may result in the non-intentional but unavoidable presence of residues or derivatives in the feed 97

ingredient or its ingredient market formulation, provided that these residues and derivatives do 98

not have an adverse effect on animal health, human health or the environment6. 99

Specification3: A list of tests results, referenced to analytical procedures, and appropriate 100

acceptance criteria that are numerical limits, ranges, or other criteria for the test described. It 101

establishes the set of criteria to which a feed ingredient and materials must conform to be 102

considered acceptable for its intended use. 103

1.3. Scope of the Guidance 104

The types of feed ingredients covered by this guidance document are determined by the 105

relevant regulations and statutes of each regulatory jurisdiction, where the feed ingredient is to 106

be marketed. 107

This guidance document also provides support for the development of the specifications 108

of the feed ingredient. 109

Note that the aspects related to the actual genetic modification of an organism that 110

produces/is the feed ingredient are out of the scope of this document. 111

2. GENERAL PRINCIPLES 112

There can be significant variability in manufacturing processes given the broad range of 113

feed ingredients used. This guidance document captures this variation. The information to be 114

provided in an application may depend on the process and/or the feed ingredient under 115

assessment. However, in the development of the application, it is necessary for the applicant to 116

consider all data required for a complete assessment of the feed ingredient and its specification, 117

in relation to its manufacturing process. 118

The description of the manufacturing process of the feed ingredient is an important 119

complement to its identification and characterization. The manufacturing process should be 120

completely described and include all process steps from the materials to the packaging of the 121

feed ingredient and/or its ingredient market formulation. The establishment of specification 122

linked to the manufacturing process is also an essential part of the identification and 123

characterization of the feed ingredient. 124

One of the main principles of this guidance document should be the verifiable justification 125

that the process is sufficiently controlled to produce a safe feed ingredient of consistent quality 126

(purity criteria, physico-chemical characteristics, contaminants, and other undesirable 127

substances). 128

When the manufacturing process is based on a series of different steps (e.g., a 129

fermentation step followed by a chemical step), each of the different steps and their sequence 130

should be clearly described. 131

The flow chart in ANNEX I illustrates the different steps of the manufacturing process for 132

a feed ingredient and its ingredient market formulation. It may be used as a template for the 133

description of the feed ingredients’ manufacturing processes. 134

While the information regarding the manufacturing process of feed ingredients may be 135

protected by Intellectual Property Rights, its disclosure during the pre-market assessment may 136

be covered by confidentiality rules. It is advisable to contact the authority of the country/region, 137

where the documentation is to be submitted, to be informed on the confidentiality rules existing 138

in that country/region. 139

3. DESCRIPTION OF THE MANUFACTURING PROCESS 140

The description of the manufacturing process provides important information for the 141

safety assessment of the feed ingredient. It allows for the verification that potential 142

contamination of the feed ingredient or its ingredient market formulation, as appropriate, is 143

sufficiently controlled by the applicant to produce a safe feed ingredient in a consistent manner. 144

The description of the critical control points of the process provides confidence that the 145

presence of contaminants and the introduction of potential hazards are monitored and 146

controlled. 147

Feed ingredients can be manufactured using various processes. It is neither realistic nor 148

achievable to provide detailed guidance for all possible processes. Rather, this guidance 149

document can help applicants to identify what relevant information should be provided in their 150

application. For example, the following information could be included: 151

• The materials and/or processing aids used in the manufacturing process, 152

• The manufacturing process steps, 153

• The ingredient market formulation process, including carrier(s) and/or other feed 154

ingredient(s) used, 155

• The packaging step, including description of the packaging used. 156

3.1. Materials 157

All the materials used in the manufacturing process of the feed ingredient under 158

assessment should be listed and identified using international naming standards, when 159

available. Their quality and safety should be documented by the compilation of safety and/or 160

product datasheets. A quantitative approach may be required when the safety profile of the 161

feed ingredient or one of its contaminants justifies it. 162

3.1.1. CHEMICAL MATERIALS 163

3.1.2. BIOLOGICAL MATERIALS 172

3.1.2.1. MATERIALS OF PLANT ORIGIN 173

The following information may be provided for material of plant origin, as applicable: 174

• Identification and description of the plant derived material, 175

• Name, source, and taxonomic classification of the plant from which the material 176

is derived, 177

• Part of the plants used (e.g., seeds, meal, grain, leaves and other parts/co-178

products), 179

• Specification 180

• Consideration of potential contaminants and toxins, 181

• Representative analysis to verify its identity and specification. 182

3.1.2.2. MATERIALS OF MICROBIOLOGICAL ORIGIN 183

For microorganisms marketed as such, used as a source of a feed ingredient (production 184

strains), or used to produce a feed ingredient (e.g., microbial biomass) the following information 185

may be provided, as applicable: 186

• Identification and description of the microbiologically derived material, 187

• Taxonomic identification of the microorganism according to the most recent 188

taxonomic nomenclature. The information should include genus, species, and/or 189

strain (when applicable), 190

• The microorganism’s name and its code, identifier, or deposition number (when 191

applicable), 192

• Specification 193

• Consideration of potential contaminants and toxins, 194

• Representative analysis to verify its identity and specification. 195

3.1.2.3. MATERIALS OF ANIMAL ORIGIN 196

• Identification and description of the animal derived material, 198

• Name, source (including country of origin of the material) and taxonomic 199

classification of the animal, 200

• Part of the animals used (e.g., blood, tissue, gland, bone, feather), 201

• Specification, 202

• Consideration of potential contaminants, including disease vectors, 203

• Representative analysis to verify its identity and specification. 204

3.1.3. MINERAL MATERIALS 205

The following information may be provided for mineral materials, as applicable: 206

• Identification and description of the mineral derived material, 207

• Name and source of the material (including country of origin), 208

• Specification, 209

• Consideration of potential contaminants, 210

• Representative analysis to verify its identity and specification. 211

3.2. Manufacturing Process steps 212

A feed ingredient manufacturing process is usually composed of multiple successive steps. 213

The data required for the assessment of the feed ingredient shall cover the whole manufacturing 214

process and each step should be described as below. 215

The description of the manufacturing process steps will help in assessing the safety and 216

quality of the feed ingredient. With regards to safety, the applicant should provide information 217

according to the principles of Hazard Analysis and Critical Control Points (HACCP) or an 218

equivalent system. With regards to quality, the applicant should provide evidence that the 219

manufacturing process ensures the production of a feed ingredient with consistent quality to 220

meet the established specification. 221

The sequence of addition of the materials and/or processing aids during each of the 222

manufacturing process steps should be described to allow the evaluation of the potential carry-223

over of unreacted materials and/or their by-products remaining in the feed ingredient or 224

ingredient market formulation. 225

The description of manufacturing process steps should be provided in the form of a 226

narrative along with a flowchart, describing the critical control points and process conditions. 227

Different types of manufacturing process steps can be envisaged, such as: 228

• Chemical steps, involving one or more chemical reactions, 229

• Fermentation/biological steps, involving the use of microorganisms or enzymes, 230

• Mechanical steps, involving the use of machinery (e.g., grinding), 231

• Thermo-physical steps, such as those involving the use of high temperature or 232

pressure. 233

Specific details for each of these types of manufacturing process steps are given below. 234

3.2.1. CHEMICAL STEPS 235

The following information may be required for the description of chemical steps used in 236

the manufacturing process of a feed ingredient: 237

• Measures applied to monitor chemical step performance within appropriate 238

limits, 239

• List of material(s) and/or processing aid(s), (see Section 3.1.1), 240

• Key elements of the manufacturing process step such as: 241

o The sequence of addition of the different material(s) and/or processing 242

aid(s), 243

o The subsequent chemical reactions, preferably with the chemical equation, 244

o Indication of typical process conditions (e.g., times, temperatures, pH 245

conditions, etc.), 246

• Controls considered for the identification of potential reaction by-products, carry 247

over of unreacted materials, residual solvents, and contaminants (Note that 248

contaminants may be carried over from the material(s) and/or processing aid(s) 249

or may be generated during the chemical step). 250

Based on this description, the applicant should identify the potential reaction by-products, 251

carry-over of unreacted materials, and processing aids. This will support the identification of 252

potential contaminants, likely to be found in the resulting material or feed ingredient. 253

3.2.2. FERMENTATION/BIOLOGICAL STEPS 254

The following information may be required for the description of fermentation/biological 255

steps used in the manufacturing process of a feed ingredient: 256

• Measures applied to monitor fermentation/biological step performance within 257

appropriate limits: 258

o The microorganism maintenance from batch to batch (e.g., master cell, 259

working cell bank), 260

o The sterilization and cleaning techniques applied, 261

o The methods used to monitor the fermentative conditions, 262

o The sequence of addition of the different material(s) and/or processing aid(s), 265

o The use of antimicrobials, 266

o The indication of typical fermentation or growth conditions, for example 267

temperature, 268

• Controls considered for the identification of potential contaminants. 269

Based on this description, the applicant should identify any potential substances of 270

concern likely to be found in the resulting material or feed ingredient at the end of the 271

fermentation/biological step, (such as viable cells, presence of fermentation by-products, carry-272

over of substrate, and microbiological contaminants). 273

3.2.3. MECHANICAL STEPS 274

The following elements may be required for the description of a mechanical step used in 275

the manufacturing process of a feed ingredient: 276

• Measures applied to monitor mechanical step performance within appropriate 277

limits and hygienic conditions of the equipment, 278

• List of material(s) and/or processing aid(s) (See Sections 3.1.2 and 3.1.3), 279

• Key elements of the manufacturing process step such as: 280

o The type of mechanical step used (e.g., grinding, mixing), 281

o The indication of typical mechanical step (e.g., duration, pressure) 282

• Controls considered for the identification of potential contaminants. 283

Based on this description, the applicant should identify the potential carry-over of 284

material(s) and the contaminants, linked to the manufacturing process step, relevant to the feed 285

ingredient. 286

3.2.4. THERMO-PHYSICAL STEPS 287

The following elements may be required for the description of a thermo-physical step used 288

in the manufacturing process of a feed ingredient: 289

• Measures applied to monitor the thermo-physical step performance within 290

appropriate limits and hygienic conditions of the equipment, 291

• List of material(s) and/or processing aid(s) (See Sections 3.1.1, 3.1.2, and 3.1.3), 292

• Key elements of the manufacturing process step such as: 293

o The type of thermo-physical steps used, 294

• Controls considered for the identification of potential contaminants. 300

Based on this description, the applicant should identify the potential carry-over of 301

material(s) and the contaminants linked to the production method for the feed ingredient. 302

3.3. Ingredient market formulation process 303

Where an ingredient market formulation is required for the feed ingredient, the following 304

information may be required: 305

• Description of the carrier(s) and/or other feed ingredient(s) included to produce 306

the ingredient market formulation, 307

• Concentration of the feed ingredient under assessment or its active substance(s) 308

in the ingredient market formulation, 309

• Typical composition of the ingredient market formulation, 310

• Process steps to obtain the ingredient market formulation. 311

3.4. Additional information 312

In addition to the description of the manufacturing process step(s), it may be required to 313

provide additional information related to the packaging and the further use of the feed 314

ingredient, such as: 315

• Description of the packaging material, 316

• Recommended storage conditions (e.g., temperature, specific handling), 317

• Shelf-life, 318

• Intended product labelling, including directions for use. 319

4. FEED INGREDIENT SPECIFICATIONS 320

This section provides guidance on how to describe the feed ingredient regarding its purity 321

criteria and other relevant quality parameters. Contaminants likely to be inherent to the 322

materials and/or processing aids used or introduced during the manufacturing process steps 323

should be included in the specification. In the case of an ingredient market formulation, 324

specification should consider the carrier(s) and/or other feed ingredient(s) used to produce it. 325

The purity of the feed ingredient and/or its active substance should be established based 326

on the analysis of multiple batches (typically 3 to 5) of production 7 , using validated or 327

internationally recognized methods of analysis. It should be consistent with label indications. 328

The specification, necessary for the control of the compliance of the feed ingredient, 332

should be proposed by the applicant, based on proper justification. They should reflect the 333

identity, safety, quality (including the purity) and intended effect of the feed ingredient. 334

The description of the specification should also include the acceptable limits, units of 335

measurement, and analytical methods used. 336

5. SPECIAL CONSIDERATIONS 337

There are cases where one or more of the manufacturing process step(s) of a specific feed 338

ingredient may not fall under one of the process steps types described above (Sections 3.2.1, 339

3.2.2, 3.2.3, and 3.2.4). In these cases, the manufacturing process step(s) should be described in 340

such a way that the relevant information is provided with the same level of detail as described 341

in this guidance document for other types of manufacturing process steps. 342
