A 550 bp sequence from lambda DNA was amplified (25 cycles) using primers and Taq DNA polymerase supplied in the GeneAmp® Kit (Roche Molecular Systems). PCR products and controls were electro phoresed on a 4% NuSieve® 3:1 Agarose gel in TAE Buffer at 5 V/cm for 3 hours. Lane 1, Msp I digest of pBR322 DNA (1.5 µg); lane 2, Hae III digest of øX174 DNA (1.5 µg); lane 3, no DNA control; lanes 4 – 9, PCR products resulting from different reaction conditions (7 µl of 100 µl reaction mixture); and lane 10, a positive control where kit template was added.

8% Polyacrylamide vertical format 8 bp Msp I 4 bp Ladder Digest Ladder

DNA ladders with 4 bp or 8 bp step sizes were prepared by ligation of Bgl II linkers. Aliquots of 0.8 µg of the ladders were separated on a 3.5% MetaPhor® Agarose gel in a horizontal format and compared to an 8% polyacrylamide gel run in a vertical format in TBE Buffer. The horizontal gel (15 cm × 20 cm and 3.0 mm thick) was run at 6.7 V/cm for 4 hours at 15°C. The vertical gel (10 cm × 20 cm and 1.0 mm thick) was run at 8 V/cm for 2 hours.

Low melt and Genetic Technology GradeTM Agarose Agarose is one of the most important factors in DNA recovery. Lonza offers low melting temperature agarose specifically engineered for a range of recovery methods.

For recovery without purification, use an agarose designed and tested for in-gel reactions (nucleic acid reactions in the presence of re-melted agarose, such as in-gel cloning, in-gel PCR and in-gel sequencing).

Genetic Technology GradeTM (GTG®) Agarose is manufactured, tested and certified for performance in downstream enzymatic applications. The additional GTG® testing provides a more reliable index of product quality, and ensures the agarose will yield biologically active DNA. In-Gel Recovered

Two lanes of Bst E II digest of lambda DNA were electrophoresed in a 1% SeaPlaque® GTG® Agarose gel prepared in 1X TAE. The 2.3 kb band was excised from each lane. One sample was ligated directly to pUC 19 in the presence of remelted agarose (In-gel) and the other sample was purified using a chaotrope-based recovery protocol (Recovered). Transformation into DH5αTM Cells was done following standard protocols.
