Dr. Sheraz Gul is Head of Assay Development & Screening at Fraunhofer-IME SP, Hamburg. He has worked in Big Pharma and academia and has experience in developing biochemical and cellular assays for High Throughput Screening and his research interests are directed towards maximising the impact of HTS for drug discovery.E-mail: [email protected]

Oliver Keminer is a drug discovery scientist with 15 years of experience whilst working at Eli Lilly (Lilly Forschungs GmbH, Hamburg), Evotec and European ScreeningPort GmbH. WORKSHOP OVERVIEW:

Drug Target Review workshops are limited to 20 attendees to ensure every participant receives the training they require. We continually update our programme to include the very latest screening technologies, ensuring attendees receive the most up-to-date hands-on training and information.

Taking place over 3 days, the workshop offers a series of practical sessions and lectures to examine the design and application of cell-based assays for small molecule screening campaigns in drug discovery. In the practical sessions, participants will have the chance to develop screening-compatible cell-based assays, take part in primary screening using a small molecule library, and profile compounds in dose-response experiments.

Attendees gain insight into the key parameters to be considered when developing cell-based assays and performing small molecule screening campaigns, associated data analysis, validation of Hits and their annotation using a variety of cell health/toxicity/liability assays.

The evening dinner on the first day will offer the opportunity for the participants to network and establish relationships that would be mutually beneficial.

FURTHER DETAILS:

A typical small molecule drug discovery project aims to identify chemical starting points that modify the functions of genes, cells, or biochemical pathways ultimately leading to new therapeutics [1]. Screening using miniaturised microtiter plate formats remains the most widely utilised methodology for identifying novel chemical starting points that are capable of modulating target function in a meaningful, biologically relevant manner [2]. The initial steps in small molecule drug discovery include the selection of a target, development of an assay to monitor its activity, and its screening against a compound library. Subsequently, the Primary Hits are optimised using multiple criteria including structure activity relationships, selectivity, physicochemical properties and liability [3,4] that could result in a Lead compound series.

We are now witnessing a resurgence of cell-based assays including phenotypic assays where the underlying target/s are unknown [5] and advances in cell-based assays are also being made, for example using human induced pluripotent stem (iPS) cell-derived cells that better recapitulate normal human biology compared to transformed cell lines and non-human primary cells [6].

THE EQUIPMENT

During this year’s workshop, the Thermo Scientific Multidrop™ Combi reagent dispenser and Varioskan™ LUX multimode reader will be used to help prepare and analyse samples in the practical sessions of the Cell-Based Assays for Screening Workshop.
