Adding a new process
1. Click ADD PROCESS
2. Click Select
3. Choose the process from the drop down
menu. (The following slides will explain each
process in more detail.)
4. Once a process has been adjusted, click SAVE
to save changes to the program
5. Click the forward arrow to run the program
6. Click the back arrow to adjust settings in a
previously added process

Incubation
Edit incubating temperature, heating
period and other parameters
Temperature range is 20-60ºC
• The IntelliPlex 1000 does not contain a cooling
system, so the low end of the temperature range
will be dependent upon room temperature
(RT+5˚C)

Incubation
Close lid while heating (suggested)
• Closes heating lid to cover the microplate
Shake before reaching temp (not
suggested)
• The microplate will start to shake before the
desired temperature is reached
• Timer starts once the microplate shakes, not
once the desired temperature is reached

Washing
Edit the Wash Cycle Number (3 cycles is
suggested)
Washing Volume o Volume of wash buffer dispensed into wells
during each wash cycle (ELISA = 200μL)
Last Dispense Volume o Dispenses 150 μL of wash buffer into wells after
the last wash cycle to ensure appropriate buffer volume for analysis
o Do not select this option if nest step is SA-PE labeling as it dilutes the SA-PE solution.

Washing
Magnetic
• Activates magnetic function during wash
cycles
• Must be clicked ON ■ for πCode assays.
Clean tip
• Tips will be cleaned after each row before
proceeding to next row (only required for the 1st
washing process for most assays)

SA-PE Reaction
Includes all steps for fluorescent labeling:
SA-PE dispense (50 µ L/well), incubation, 4 wash
cycles, and 10 sec. shaking to spread πCode
evenly. (All parameters except incubation period are
fixed.)
Heating period
• Adjustable from 1s to ~24hr (Suggested
heating period is 10 min)

Pause
Allows for a pause between processes
It may be needed to add additional solution to a
sample manually. To do so, take 96 well plate out
of the IntelliPlex 1000.
Before continuing with the program, ensure that
96 well plate has been positioned correctly in
the IntelliPlex 1000
Continue with the next process in the program
by clicking “OK”

Disperse
Shake and pipet the components in wells.
The tips will conduct self-clean among
rows.
• This is the last step of a program. It spreads
πCode MicroDiscs evenly before analysis.
• Not an adjustable parameter.

Preparing to run program
Check and save the program before proceeding
to next step
Click to edit the program or rows if needed
Ensure there is sufficient wash buffer and
ddH2O for running the assay
Make sure the air vents of buffer bottles are
loose
Remember to add SAPE in V tray. See next page
for SAPE usage calculation.
Click START to run the program.

SA-PE usage calculation Dead volume: 500μL
Minimum dispense: 400μL for one row
Example:
For 3 rows reaction… 400 µ L x 3 rows + 500 µ L = 1.7mL (at least)
* Add 300 µ L more if running more than 6
rows
