Drugmakers Look To Use PurduePharma’s Bankruptcy To Settle

Opioid Suits -WSJEndo International Plc, Johnson & Johnson and

other drugmakers that face litigation over the opi-

oid crisis are exploring a way to settle the cases by

participating in Purdue Pharma LP’s bankruptcy,

the Wall Street Journal reported, citing internal

documents and a person familiar with the matter.

Five drugmakers battling the cases – Endo, J&J,

Teva Pharmaceutical Industries Ltd , Allergan Plc

and Mallinckrodt Plc – are looking to enact a glob-

al settlement of the litigation that would be imple-

mented through Purdue’s Chapter 11 case, the

WSJ reported citing a person familiar with the mat-

ter. The mechanism, if successful, would allow the

companies to contribute money into a trust set up through the

bankruptcy in exchange for a complete release from liability,

according to the report.

ally. The available cancer treatment

options include chemotherapy, radio-

therapy, and surgery depending on the

location and stage of malignant behav-

iour of cancer. The early diagnosis of

cancer has often been seen as a boon

for the treatment of patients suffering

from cancer, which requires biopsy; a

method which requires collection of tis-

sue sample from tumor followed by their

analysis. The method of biopsy is highly

invasive as it involves surgery, whereas

the emerging of use of extracellular vesi-

cles (EVs) such as exosomes and

microvesicles (MVs), has led to develop-

ment of non-invasive liquid biopsy.EVs

are nanovesicle which are released from

most of the cells, and biofluids including

blood, cerebrospinal fluid, urine, etc.

Their role has been found in metastasis

of various cancer including glioma; a

deadly brain cancer. Despite tremen-

dous research on EVs, there has been a

lack of clear consensus about the char-

acterization of exosomes and MVs.

Biomedical Sciences) andProf.Chi-Man

Lawrence Wu (Department of Material Science and Engineering) atCity University of Hong Kong, dis-

covered that the two important EVs derived from cancer cells react differently towards the gold

nanoislands (AuNIs)-localised surface plasmon resonance (LSPR) biosensor. Their research team

include Dr. Abhimanyu Thakur, Dr.GuangyueQiu, Dr. Siu-Pang NG, Jintao Guan, and Dr.Jianbo Yue.

Their discovery suggests that exosomes and MVsposses different biophysical characteristics, which

can be useful for liquid biopsy in cancer diagnosis as well as prognosis. Their work has been pub-

lished in a top journal related to biosensing (Thakur A et al, Biosensors and Bioelectronics 2017, 94:

400-407; DOI: 10.1016/j.bios.2017.03.036). Dr. Abhimanyu Thakur said, “Cancer derived exosomes

and microvesicleshave distinct biophysical characters, which can be used as a diagnostic marker.

AstraZeneca’s Combo Lung Disease Therapy Fails To Win FDAApproval

The U.S. Food and Drug Administration declined to approve AstraZeneca Plc’s combination therapy to

treat smoker’s lung, the drugmaker said on Tuesday. AstraZeneca said that it will work closely with the

FDA regarding next steps,

including submitting results from

an additional late-stage study,

which was not completed at the

time the marketing application

was submitted to the health

regulator. The therapy, PT010,

was approved in Japan in June

as a triple-combination therapy

to relieve symptoms of chronic

obstructive pulmonary disease

(COPD), AstraZeneca said.

Colloquially known as smoker’s

lung, COPD is a progressively

worsening and potentially

deadly condition that affects

more than 380 million people

worldwide, primarily caused by

smoking but also by occupa-

tional hazards such as air pol-

lution or chemical fumes. COPD

is predicted to be the third lead-

ing cause of death by 2020,

according to the British drug-

maker. The setback comes as

AstraZeneca tries to catch up

with domestic rival

GlaxoSmithKline’s Trelegy,

which is delivered through

inhaler Ellipta. That treatment

pulled in 156 million pounds in

sales last year in its use against

COPD.

Fruit Flies Live Longer With Combination Drug TreatmentA triple drug combination has been used to extend the lifespan of fruit flies by 48% in a new study led by UCL and the Max Planck Institute for Biology of Ageing.

The three drugs are all already in use as medical treatments: lithium as a mood stabiliser, trametinib as a cancer treatment and rapamycin as an immune sys-

tem regulator. The findings, published in Proceedings of the National Academy of Sciences (PNAS), suggest that a combination drug treatment may one day be

helpful at preventing age-related diseases in people. "As life expectancies increase, we are also seeing an increase of age-related diseases so there is an urgent

need to find ways to improve health in old age," said the study's co-lead author, Dr Jorge Castillo-Quan, who began the research at the UCL Institute of Healthy

Ageing before moving to Joslin Diabetes Center, Harvard Medical School. "Here, by studying fruit flies which age much more rapidly than people, we have found

that a combination drug treatment targeting different cellular processes may be an effective way to slow down the ageing process." The researchers were build-

ing on previous studies finding that lithium, trametinib and rapamycin can each extend lifespan in fruit flies (Drosophila), which is supported by other preliminary

evidence in mice, worms, and cells, and observational findings in people. The three drugs all act on different cellular signalling pathways that together form the

nutrient sensing network, which is conserved across evolution from worms and flies all the way to humans. This network adjusts what the body is doing in

response to changes in nutrient levels. The three drugs in question act on different proteins of this network to slow the ageing process and delay the onset of

age-related death. For the latest study, the researchers gave fruit flies doses of lithium, trametinib and rapamycin, separately and in combination. Each drug indi-

vidually extended lifespan by an average of 11%, while pairing two drugs extended lifespan by roughly 30%. When the three drugs were combined, the fruit flies

lived 48% longer than flies in a control group that were not given the treatment. "Previous studies in fruit flies have achieved lifespan extensions of about 5-20%,

so we found it was quite remarkable that this drug combination enabled them to live 48%

longer," Dr Castillo-Quan said. The researchers found that in addition to acting on sepa-

rate signalling pathways within the nutrient sensing network, the drugs also appear to

complement each other to reduce side effects. Rapamycin has undesirable effects on fat

metabolism, which can be similar to insulin resistance in people, but lithium appeared to

cancel out this effect when the two drugs were given together. The researchers will con-

tinue their research to better understand exactly how the drugs work in combination with

each other, and hope to progress to experiments in more complex animals, such as mice,

to gauge the effects on the entire body before eventually progressing to human trials.

Principal investigator, Professor Linda Partridge (UCL Institute of Health Ageing and Max

Planck Institute for Biology of Ageing), said: "There is a growing body of evidence that

polypills -- pills that combine low doses of multiple pharmaceutical products -- could be

effective as a medication to prevent age-related diseases, given the complex nature of the

ageing process. This may be possible by combining the drugs we're investigating with

other promising drugs, but there is a long way to go before we will be able to roll out effec-

tive treatments." "My research groups are working to understand the mechanism of the

ageing process in order to find ways to help people stay healthy for longer. We are not

trying to cheat death, but help people be healthy and disease-free in their final years," she

The total addressable market size for these three products in Mainland China is about US$

Nucala Is The First Biologic ApprovedIn The US For Six To 11-year-Old

Children With Severe EosinophilicAsthma

approved Nucala (mepolizumab)

for use in children as young as six

years old who are living with severe eosinophilic asthma.

Nucala is the only targeted biologic to be approved for the con-

dition in the six to 11-year age group in the US. Dr

The 52-week long-term phase of the study showed that the

safety profile in paediatric patients aged six to 11 years was

similar to the known safety profile in patients aged 12 years

in children is a complicated condition that can be extremely

challenging to treat. Nucala has made a difference for many

patients”. Nucala (100mg dose subcutaneous injection) was

first approved in 2015 as an add-on maintenance treatment for

patients with severe eosinophilic asthma aged 12 years and

older. This approval (40mg dose subcutaneous injection)

extends the current indication in the US for Nucala to patients

aged six to 11 years. Nucala has been approved for use as an

add-on treatment for severe eosinophilic asthma in patients

aged six years and older in the EU since August 2018.

Stable Metal Organic Frameworks Are Prized For TheirAbility To Capture Carbon Dioxide Or Harvest

Atmospheric Water, But U-M Researchers HaveDeveloped A Use For Unstable Metal OrganicFrameworks: As A System For Drug Delivery.

Ninety percent of drugs in development don't dissolve well or at all in the body, according

to a 2012 study. Metal organic frameworks, or MOFs, are rigid, porous structures com-

posed of metal linked by organic ligands. While researching unstable MOFs, U-M chemist Adam

Matzger realized they may work as a delivery system for these kinds of drugs. "It struck us that

while everyone was shooting for stable MOFs, we could use what we had learned about instability

of MOFs to get a degradable substance that would rapidly release drugs. It was a really unexpect-

ed finding," said Matzger, the Charles G. Overberger Collegiate Professor of Chemistry and a pro-

fessor of macromolecular science and engineering. The MOF delivery system takes advantage of

one strategy of delivering insoluble drugs, Matzger says. The strategy is to deliver them in an amor-

phous form, meaning that the drugs are kept from crystallizing. Once the drug compounds crystal-

lize, they become less soluble -- and therefore less bioavailable, which means less of the drug

crosses into the bloodstream. MOFs are able to keep these drugs in an amorphous state because

of their porous structure. MOF-5 in particular looks like a set of cubes stacked on top of each other:

picture an endlessly repeating 3D grid. When the

researchers load a drug into this grid, the pores in

the MOF compartmentalize the drug molecules,

holding the compound in the desired amorphous

state. Researchers have previously used polymers

to hold drug compounds in an amorphous state, but

drug molecules could still migrate and crystallize in

the polymer, which affects solubility and therefore

how much of a drug is bioavailable. Because MOF

rigidly holds drug molecules apart but quickly

decomposes, drug dosage is easily controlled,

Matzger says. The compound itself crosses into the

bloodstream, while the MOF decomposes in the

body. The researchers, who include postdoctoral

fellow Kuthuru Suresh, decided to use MOF-5 for a

few reasons. First, its metal component is zinc, a metal with low toxicity used in many supplements,

and the organic component is an acid called terephthalic acid. Second, MOF-5 is relatively unsta-

ble, which the researchers demonstrated in separate work that examined the stability of different

MOFs. "What we do here is we increase the solubility while guaranteeing dosage stability," Matzger

said. "This approach is relatively universal. We show it for three drugs, but the fact that the pores

are too small to allow a crystal to form is going to be true for all drugs."
