5. PurgeTCUandJacket:a. Openvalves.Ensureinletandoutletvalvesonthejacketare
open.b. Turnonthemainpowerusingthetoggleswitchatthefrontof
theTCU.c. Onstartup,theTCUwillself-testandbegintopurgetheair
outoflinesandjacket.d. Duringthepurgeprocess,thereservoirlevelwilldropinorder
tofillthejacketoftheS.U.B.NOTE:VerifybypassvalveisnotrecirculatingfluidinTCU
e. Whenthelevelgetstoolow,theTCUwillsetoffanalarm.Repeatstep4andfillthereservoirtomid-levelandthealarmwillturnoffandthemachinewillcontinuetopurgeandfillthejacket.
f. Thisprocessmayneedtoberepeatedseveraltimesuntilthejacketisfullandmostoftheairisremovedfromthesystem.
g. Placeatrayorbucketunderneaththejacketventvalve.h. Keeptheventvalveopenuntilcoolingfluidcomingoutof
thevalveisfreeofair(TCUneedstoberunningduringthisprocess).Thismaytake1-2minutes.Thenclosethevalve.
i. DoafinalcheckonthefluidleveloftheTCUandensurethefluidlevelisatmid-level.
j. Ifnecessary,adjustdefaulttemperature.
CAUTION: Donotfillreservoirtotopoffilllevel,thecoolantmayneedroomforexpansion.
6. WhentheTCUisinitsfinallocation,rollthecontroltowerintoplaceovertheTCUanduptothestoplocatedinsidethetower.Notethatanaccessdoorislocatedonthefrontofthetowertoviewormanipulatethecontrols.

2.1 Orientation and Location
Section 2 Hardware Installation
TheSingle-UseBioreactorhardwareisshippeddirectlyfromthemanufacturerandwillarrivecrated.Forunpackinginstructionsanddetailedcontentsofthecrate,pleaserefertotheuncratinginstructionsthatcamewiththeshipment.Besuretofollowtheunpackinginstructionsprovidedandretainallpackagingmaterials.
Tools Needed:• Allenwrenchprovidedinshippingdocumentationpacket• Adjustablewrenchneededforlevelingfeet(notsupplied)
a. Positionreactorandtowerindesiredlocationandlockwheelsensuringavailabilityofneededutilities(power,gas,etc.).Rightorlefthandorientationwasdeterminedattimeoforder.Ensurethattowerislocatedonthesamesideasthecableandtubingorganizer.
b. Beforerunningcables,notethatthecableandtubingorganizer
hasanadjustmentfeaturetokeepthecableslinedupwiththerearofthecontroltower.Thearmhasaslidethatcanextendandretractmanuallyusingtheclampingknob.
WARNING:Anyproceduresthatrequirethecontrolboxtobeopenshouldbeperformedwiththemainelectricaldisconnectinthelockedoutpositionandallpowersourcesremovedfromthecontrolbox.Foroperatorsafety,securethelocationoftheS.U.B.outersupportcontainerbydisablingtheswivelcastersbeforeservicing.

ConnectthecorrespondingfacilitygassourcestothedesignatedconnectorsontheTower.Theconnectorsarefractional(inches),soifthefacilityusesmetriclines,simplyusetheadaptersprovidedintheplasticbaginsidetheTower(6mm.to1/4”adapters).

Beforeconnectingpower,verify,again,allgaslineswerecheckedandnotleaking.
• Connectmainpowercord(nocolorcode)tocorrectutilitysource.• ItisstronglyrecommendedthataUPS(UninterruptedPower
Supply)(● ● ●)beused.Thepurposeofthisconnectionistomaintainpowertothecontrolsduringapoweroutage.
• IfUPSpowerisnotavailableornotused,thentheUPSJumperconnectormustbeputinplaceoftheUPScable.

unitislevelbeforeoperation.• UsingtheAllenwrenchprovidedwithS.U.B.unitshipping
documents,removetheloadcellshippingplatesandsecuretheminthebracketattherearofthecontroltower.Therearetwo¼-20socketheadcapscrewsthatneedtoberemovedfromeachloadcellprotectionplate.Movethetowertodesiredpositionandlockinplaceandlevelusingthelevelingfeet.

• RefertoUser’sGuidefordetailedinstructionsoncalibrationsteps(loadcells,sensors,etc).
• Avirtualkeyboardiconislocatedatthebottomofthewindowdisplayfornotes,values,etc.
• StartbyturningontheMAINDISCONNECTswitchlocatedatthebackofthecontroltower.
• EnsuretheEmergencyStop(locatedatthefrontofthetower)isnotactivated(shouldbeintheoutposition).Ifitisengaged,thentwisttodisengage.NotethattheE-stoplightwillremainred,eventhoughitisdisengaged,untilthenextstephasbeencompleted.
• TurnpoweronbypressingthegreenStart/Re-setbuttonlocatedatthefrontofthetowerbehindtheoperatorscreen.E-stoplightwillthenturnoff.
• Notethatagreenoperationlight,locatedatthebackofthetower,willalsoturnon.

Section 3 BPC Installation
1. CarefullyunpacktheS.U.B.BioProcessContainer(BPC)fromitstwoprotectivepolybags.
2. Uncoilandremoveprotectivepackagingfromthesparger,drain,andauxiliarylinesonthebottomoftheBPC.Don’tremovetheprotectivepackagingontopoftheBPCatthistime.
3. CarefullylowerthebottomendoftheBPCintotheS.U.B.withanytubingsetsonthetopsidelayingoverthesideuntilthebearingassemblyislevelwiththedriveshaftmotor.
4. Removetheprotectiveblackcapfromthebearingassemblyandslideintotheretentionmechanismontheagitationmotorandlockitintoplace.

5. Guidebottomtubinglinesetthroughtheirproperchannels:• Insertfritandopenpipespargerlinesthroughthespargeplate
(round,whiteplasticwithslots)first,beforelocatingitinthebaseofunit.
• Insertdrainlinethroughbottomfrontfacedirectlybelowagitationmotor.Hangthedrainlinecoilonthearmprovided.Alsoroutethesamplelinethroughthesnapinslotsonthedraincoilarm.
• AttachBPCpositioningtabs(bottomofchamber)ontothealignmentpinslocatedatthebaseoftheS.U.B.tank.Notethatthe50LBPCdoesnotrequirethisstepandisnotrecommended,buthastabholdersavailable,ifdesired.
• Insertsensors,KleenPaks,andauxiliarylinesetsthroughthe“probebeltopeningonthetank”.
6. Uncoverventfilters,wrapheatingblankets(ifused)aroundthefilters,attach/adjustventbracket,andmountventfiltersinbracket.

7. Routetheoverlayspargeset(locatedatthetopoftheBPCmarked“OverlaySparge”)throughthecableandtubingorganizerarmtothetopofthemainbeamofthecableandtubingorganizerandconnecttothefittingmarkedOVERLAYSPARGE.Thiswillbethelinesetthatwillbeusedtopre-filltheBPCwithair,soensurethattheclamponthislinesetisreleased.
8. RoutetheMicroSpargeLine(labeled),locatedatthebottomoftheBPC,upthroughthemainbeamofthecableandtubingorganizerusingtheblacksnapinfeatures.ConnecttothecorrectgasfittinglabeledMICROSPARGE.Unclampthislinesetwhenreadytospargethroughfrit.
9. RouteMacroSpargeline(labeled),locatedatthebottomoftheBPC,runningparalleltotheMicroSpargeLine,usingthesamesnapinfeatures.ConnecttothecorrectgasfittinglabeledMACROSPARGE.Ensuretheclampisreleasedonthisline,aswellbeforeoperation.NOTE:Toreducetheriskoffiltermembranewetting,positioneachspargefilterabovethelowpointofthespargeline.
10.ConnectthepressuresensorcableontothesensorlocatedatthetopoftheBPC.

11.EnsurethecablehasbeenroutedthroughthecableandtubingorganizerandconnecttothefittinglocatedattherearofthecontrolcabinetlabeledBAGPRESS(● ● ●).
12. IMPORTANTDisengageanyclampsonthelinesetfortheexhaustfilter(s).Iftheclampsareleftengaged,theBPCcouldburstwhenfillingitwithairorliquid.
13.Positionandcloseabarclamp(PartNo.SV20664.01)onthebottomdrainlineascloseaspossibletotheBPCport(Figure14).Alsoplacebarclampsonprobeportsintendedforconventionalprobes(Figure15).
14.InsertRTDorselectedtemperaturesensorintothermo-well(Figure16,Figure17).• Placesmallamountofglycerol(0.5mL)inwelltoaidinheat
transfer.• Sensorshouldbeinserteduntilbaseofprobemeetsthemouth
