TO: Hamlet Police Department Attention: Detective Bill Shakespeare 1 Banquo Avenue Hamlet, XX 99999
REPORT DATE: 10/14/11 GFS CASE NUMBER: 11-0101 AGENCY NUMBER: Hamlet PD #2011M1101 SUBJECT(S): King, Duncan (victim) Queen, Macy Beth (suspect)
ITEMS SUBMITTED
On 010/22/11, the following item was submitted by Malcolm Macduff of the Hamlet Police Department by way of Federal Express Tracking Number 797268585956: ITEM NUMBER(S) DESCRIPTION 1 One brown paper bag marked “Seyton Place” containing one black dagger with a brown
leather handle On 10/05/11, the following item was submitted by Malcolm Macduff of the Hamlet Police Department by way of Federal Express Tracking Number 796595858627: ITEM NUMBER(S) DESCRIPTION 2 One manila envelope marked “Duncan King” containing one white FTA card bearing a
reference blood sample from Duncan King 3 One manila envelope marked “Macy Beth Queen” containing one white FTA card bearing a
reference blood sample from Macy Beth Queen
EXAMINATION AND SEROLOGICAL ANALYSIS
Item 1 – Dagger The dagger was examined for the presence of either peroxidase activity (an enzymatic activity characteristic of, but not unique to, blood) and hemoglobin (a chemical unique to blood) or epithelial cells (a type of cell highly characteristic of, but not unique to, skin). The following samples were collected, assigned exhibit numbers, and tested:

CELLS PEROXIDASE
ACTIVITY HEMOGLOBIN EX01-1 Sample of red stains from the blade of the dagger (not tested) POSITIVE POSITIVE EX01-2 Swabbing of the handle of the dagger POSITIVE (not tested) (not tested) INTERPRETATION: The tested stain on the blade of the dagger (EX01-1) is blood, or is a mixture that contains detectable levels of blood.
INTERPRETATION: The tested swabbing of the handle of the dagger (EX01-2) bears skin tissue, or is a mixture that contains detectable levels of skin tissue.
Item 2 – Reference blood sample of Duncan King The following sample was collected and assigned an exhibit number: EXHIBIT NUMBER DESCRIPTION EX02-1 Cutting of the reference blood sample of Duncan King

Item 3 – Reference blood sample of Macy Beth Queen The following sample was collected and assigned an exhibit number: EXHIBIT NUMBER DESCRIPTION EX03-1 Cutting of the reference blood sample of Macy Beth Queen
ANALYSIS
The question samples (EX01-1 and EX01-2) were compared against the reference samples (EX02-1 and EX03-1) by deoxyribonucleic acid (DNA) analysis of short tandem repeats (STRs) using polymerase chain reaction (PCR) technology. The amplification of DNA was performed using Promega’s PowerPlex 16 HS kit, which includes the thirteen core CODIS loci as specified by the FBI (D3S1358, TH01, D21S11, D18S51, D5S818, D13S317, D7S820, D16S539, CSF1PO, vWA, D8S1179, TPOX, and FGA), as well as Penta E, Penta D, and Amelogenin.
The analysis was performed on an ABI Prism 310 Genetic Analyzer. The raw analytical data is available upon request.

The sample identified as EX03-1 (reference blood sample of Macy Beth Queen) produced a DNA profile.
Item 1 – Dagger EX01-1 (blood stain from blade of dagger)
Summary: DNA profile consistent with Duncan King The sample identified as EX01-1 produced a DNA profile at sixteen of the sixteen loci analyzed. This DNA profile is consistent with a single contributor. The DNA profile is consistent with that of EX02-1. Hence, Duncan King cannot be excluded as a source of this sample. Statistical Interpretation: Using the genotypic frequencies published in Applied Biosystems’ AmpFlSTR Identifiler User’s Manual, the statistical probability of another unrelated individual with the same alleles as EX01-1 is:
U.S. Caucasians: 1 in over 7,000,000,000,000 (trillion) U.S. Caucasians U.S. Africans: 1 in over 7,000,000,000,000 (trillion) U.S. Africans U.S. Hispanics: 1 in over 7,000,000,000,000 (trillion) U.S. Hispanics Native Americans: 1 in over 7,000,000,000,000 (trillion) Native Americans
The DNA profile is inconsistent with that of EX03-1. Hence, Macy Beth Queen can be excluded as a source of this sample.
EX01-2 (possible skin cells from handle of dagger) Summary: DNA profile consistent with Macy Beth Queen The sample identified as EX01-2 produced a DNA profile at sixteen of the sixteen loci analyzed. This DNA profile is consistent with a single contributor. The DNA profile is consistent with that of EX03-1. Hence, Macy Beth Queen cannot be excluded as a source of this sample.

CLOSING REMARKS
The submitted items and associated exhibits will be retained until further disposition. Sufficient quantities of reference and question samples remain for reanalysis, depending upon the type of DNA analysis technology used. This analysis was performed under chain of custody. The chain of custody documentation is on file at Guardian Forensic Sciences. Positive and negative controls in each stage of analysis of this case performed as expected.
