Ribotypes of Staphylococcus aureus clinical strains. A total of 53 banding patterns were obtained by Southern blot hybridiza- tion of Hindlll-digested genomic DNA with a 23S ribosomal DNA probes2 (pOH4). S. aureus strains from 4 sources were analyzed. The number of strains belonging to each ribotype is given under each ribotype. Methicillin-sensitive S. aureus (MSSA) strains isolated in 1959 in Tokyo are the isolates from community-acquired infection. MSSA and methicillin-resistant S. aureus (MRSA) strains isolated from 1982 to 1983 were obtained from 19 hospitals in Japan. W-MRSA strains are isolates from 20 countries and described previously32; the strains are listed in Fig. 4. KL-MRSA strains are nosocomial MRSA strains obtained consecutively within 1987 to 1989 from Malaya University Hospital, and have been fully described. 29 The underlines of coagulase type signify the discrepancy between coagulase typing and ribotyping (see text for explanation). The underlining in the row of W-MRSA strains indicates 3 clusters of ribotypes (designated cluster A, C, and B from left). MWM, molecular weight marker; Hindlll-digested lambda DNA. 297

Fig. 4. The methicillin-resistant Staphylococcus aureus (MRSA) genotyping and inference of clonotype. Clonotype is described by a Roman numeral (corresponds to the type of mec DNA) and a capital letter, which shows the group of ribotypes to which the strains belong. Capital Roman numeral was used to indicate that the clonotype has been inferred with mec DNA type determined by dot-blot hybridization, and a small numeral was used to indicate the inference is made on mec right extremity polymorphism (MREP) typing of mec DNA. Ribotype number refers to that of Fig. 3. Subtyping is based on the mecl-gene polymorphism. Different filling pattern of meclgene represent different type of mecl mutation. 32 Asterisk shows the operator mutation of mecA gene which presumably has an equivalent outcome with mecl gene inactivation. 32 Open rectangle of mecl gene indicates intact mecl gene, and the strain name with bold font means preMRSA strain to which subtype '0' is assigned. ND, not determined; NT, nontypable (MREP-PCR did not amplify the product; the right junction point might have been spontaneously deleted from the strain).
