Institute of Botany Białowieża Geobotanical Station Head: dr. Bogdan Jaroszewicz, [email protected] Scientific activity of the Station focuses on ecology of natural forests, plant ecology, plant invasions and plant-animal interactions. The research is based on long-term studies, with the oldest ones started in 1960s. Your laboratory training will focus on field and lab experiments related to seed dispersal and survival. Field experiments will be carried out in the Białowieża Forest and at Biebrza Marshes (North-East Poland). Techniques and skills. Students will be trained in investigation of ecological phenomena related to plant dispersal: sampling of soil for seed bank studies, conduction of greenhouse experiments, design of field experiments focused on plant dispersal,especially by zoochory, determination of plant species on the basis of seed and seedling morphology.

Institute of Experimental Plant Biology Department of Plant Bioenergetics Head: prof. Anna Rychter, [email protected] Scientific activity: Energy economy of the plant cell; influence of environment and role of mitochondria in maintaining cell redox homeostasis; cyanide-resistant respiration and alternative oxidase (regulation and activity); influence of environmental factors on formation of reactive species and oxidative defense systems. Projects offered for visiting students: ATP and NADH changes as affected by differences in supply of sulphur or nitrogen; alternative oxidase and cyanide-resistant respiration during stress; plant defense against reactive species formation; structure and capacity of plant respiratory chain examined by Blue Native electrophoresis(BN PAGE). You will receive training in (some of) the following techniques: spectrophotometry, oxygraphy, luminometry, HPLC, immunodetection, gel electrophoresis, electron and fluorescence microscopy.

Metabolism of glucose and amino acids under normal and pathological conditions; proteome and lipidome of thylakoid membranes, changes in chloroplast structure under chilling stress. Projects offered for visiting students: analysis of lipid and protein composition of thylakoids of higher plants; investigation of chloroplast structure in situ by confocal laser scanning microscopy; glucocorticoid effects on regulation of gluconeogenesis with the use of renal tubules grown in primary cultures; regulation of AMPK signaling pathways; diabetes-induced changes in mitochondrial morphology and function; effect of PPAR agonists on metabolic and proteomic profiles in kidney of diabetic animals; role of antioxidant status in energy metabolism of renal tubules

Ins�tute  of  Biochemistry      Department  of  Molecular  Biology      Head:  prof.  Krzysztof  Staroń,  [email protected]      Scien�fic  ac�vity.  Inves�ga�on  of  selected  nuclear  proteins:  topoisomerase  I  (topo  I),  enhancer  of  rudimentary  homolog  (ERH)  and  protein  kinase  CK2  (CK2).  Studies  are  aimed  at  finding  protein  partners  of  topo  I,  ERH  and  CKII  and  understanding  the  structural  basis  of  the  interac�ons  as  well  as  their  role  in  subnuclear  localiza�on  of  the  proteins.  Two  of  these  proteins  are  also  targets  of  an�-­‐cancer  drugs.    Projects  offered  for  visi�ng  students:  protein  kinase  ac�vity  of  topo  I;  interac�ons  between  ERH  and  its  protein  partners;  apopto�c  proteins  phosphorylated  by  CKII.    Techniques  used:  bacterial  transforma�on,  heterologous  protein  expression;  DNA  isola�on  and  purifica�on,  PCR,  DNA  recombina�on  in  vitro,  DNA  electrophoresis,  protein  isola�on  and  affinity  purifica�on,  Blue  Na�ve-­‐  and  SDS-­‐PAGE,  western-­‐blo�ng,  protein  interac�on  assays,  siRNA,  eukaryo�c  cell  cultures,  confocal  and  fluorescence  microscopy  of  tagged  proteins.

Ins�tute  of  Microbiology      Department  of  Applied  Microbiology      Head:  prof.  Jacek  Bielecki,  [email protected]    Scien�fic  ac�vity.  Mechanisms  of  microbial  pathogenesis  and  produc�on  of  vaccines,  use  of  live  vectors  (Listeria  monocytogenes)  in  biotechnology  of  new-­‐genera�on  vaccines  (induc�on  of  intracellular  immunity,  use  of  molecular  mechanisms  of  toxin  ac�vity  and  different  determinants  of  pathogenesis  in  vaccine  design),  an�-­‐cancer  vaccines.      Projects  offered  for  visi�ng  students:  inves�ga�on  of  molecular  mechanisms  of  bacterial  toxin  ac�vity  and  different  determinants  of  pathogenesis  for  use  in  vaccine  design;  construc�on  of  bacterial  strains  expressing  toxins  as  poten�al  an�-­‐cancer  vaccines.    Methods  used:  determina�on  of  cytotoxicity  of  chosen  bacterial  pathogenic  factors  in  eukaryo�c  cells,  cloning  and  expression  of  genes  encoding  bacterial  toxins.  Modern  molecular  biology,  microbiology  (cloning,  PCR,  Southern  (DNA)  blo�ng,  Northern  (RNA)  blo�ng,  Western  blo�ng,  DNA  sequencing,  oligo-­‐directed  mutagenesis,  protein  expression)  ,  advanced  microscopy  and  op�cal  imaging  techniques.  Some  experience  in  �ssue  culture  methods  is  desirable.

Ins�tute  of  Microbiology      Department  of  Bacterial  Gene�cs      Head:  prof.  Dariusz  Bartosik,  [email protected]    Research  group  of  prof.  Katarzyna  Jagusztyn-­‐Krynicka  Scien�fic  ac�vity.  Inves�ga�ons  of  pathogenic  bacteria:  func�onal  analysis  of  Campylobacter  and  Helicobacter  unique  genes  whose  products  are  poten�ally  involved  in  pathogenesis,  construc�on  of  avirulent  Salmonella  expressing  Campylobacter  an�gens  and  evalua�on  of  the  efficacy  of  vaccine  prototype  (animal  model  –  chickens).      Projects  offered  for  visi�ng  students:  mechanisms  of  Campylobacter  spp.  and  H.  pylori  Dsb  (disulphide  bond)  pathway  func�oning.  Dsb  family  of  redox  proteins  facilitate  forma�on  of  disulfide  bridges  in  periplasmic  proteins  of  Gram-­‐nega�ve  pathogens.  Understanding  of  the  process  crucial  in  the  pathogenicity  of  these  microorganisms  might  also  facilitate  iden�fica�on  of  new  candidate  proteins  for  vaccine  construc�on.      Techniques  used:  classical  bacterial  gene�cs  and  molecular  microbiology.  Laboratory  is  equipped  for  conduc�ng  work  on  gene  cloning,  evalua�on  of  gene  expression,  protein  purifica�on  and  studying  interac�on  between  pathogen  and  eukaryo�c  cells.

Ins�tute  of  Microbiology      Department  of  Bacterial  Gene�cs      Head:  prof.  Dariusz  Bartosik,  [email protected]    Research  group  of  prof.  Dariusz  Bartosik  Scien�fic  ac�vity.  Biology  of  mobile  gene�c  elements  (MGE)  -­‐  transposable  elements,  integrons,  plasmids  and  bacteriophages,  as  well  as  genomic  islands  and  islets  acquired  by  lateral  transfer  events.  We  analyze  MGEs  in  strains  of  Proteobacteria  isolated  form  various  environments.  The  analyzed  elements  are  a  source  of  gene�c  modules  used  for  construc�on  of  specific  casse�es  useful  in  gene�c  engineering  and  biotechnology.      Projects  offered  for  visi�ng  students:  iden�fica�on  and  characteriza�on  of  novel  bacterial  plasmids  and  transposable  elements;  cloning  of  basal  plasmid  replicons  (by  construc�on  of  shu�le  plasmids),  characteriza�on  of  the  thus  obtained  mini-­‐deriva�ves  (e.g.  determina�on  of  host  range  etc.);  DNA  sequencing  and  bioinforma�c  sequence  analyses.  Transposable  elements  will  be  iden�fied  using  entrapment  vectors  which  enable  capture  of  func�onal  TEs.      Techniques  used:  isola�on  of  plasmid  and  megaplasmid  DNA,  DNA  cloning,  introduc�on  of  foreign  DNA  into  bacterial  cells,  PCR  amplifica�on,  DNA  sequencing,  DNA  hybridiza�on,  bioinforma�c  sequence  analyses.

Ins�tute  of  Zoology      Department  of  Hydrobiology      Head:  prof.  Joanna  Pijanowska,  [email protected]    Scien�fic  ac�vity.  Evolu�onary  and  popula�on  ecology  of  plankton  and  fish.  Phytoplankton-­‐zooplankton  and  zooplankton-­‐fish  interface.  Molecular,  behavioral,  life-­‐history  and  morphological  adapta�ons  to  preda�on  and  food  limita�ons.  Role  of  Heat  Shock  Proteins  (HSP)  in  individual  reac�on  to  stress  from  bio�c  and  abio�c  factors  in  Daphnia  as  a  model.    Projects  offered  for  visi�ng  students:  two  pa�erns  of  prey  selec�on  by  a  plank�vorous  fish  (roach  as  a  predator  harvester,  and  a  predator  hunter)  and  prey  (Daphnia)  demographic,  life-­‐history  and  behavioral  responses  to  the  risk  of  preda�on;  varia�ons  in  life  history,  morphology  and  depth  selec�on  behavior  of  Daphnia  mediated  by  the  presence  of  filamentous  cyanobacteria.    Techniques  used:  standard  methods  in  growing  planktonic  algae,  cyanobacteria,  zooplankton  (flow-­‐through  cultures)  and  fish  in  lab  cultures;  video  techniques  in  fish  behavioural  studies;  “plankton  organ”  as  a  tool  for  studying  depth  selec�on  pa�erns  in  Daphnia.

Institute of Zoology Department of Parasitology, head: prof. Edward Sinski, [email protected] Scien�fic  ac�vity:  Biology  of  parasites  including  immunology  of  nematode  infec�ons  in  

laboratory  models.  Immunomodula�on  and  immunogenesis  of  resistance  and/or  suscep�bility  to  parasite  infec�ons.  Cytokines  and  Th1  and  Th2  responses  to  parasites.  Apoptosis.    

Ecology  and  molecular  biology  of  host-­‐parasite  interac�ons.  Helminthes  in  wild  animal  popula�ons.  Environmental  and  zoono�c  reservoir  of  parasites  for  human  communi�es.  Vectors,  reservoirs,  environmental  condi�ons  and  transmission  of  �ck  borne  diseases  (TBD)  pathogens.    

The  laboratory    training  will    consist  of:    a.  Heligmosomoides  polygyrus:  Apoptosis  and  opioid  pep�des  in  immunoregula�on  during    primary  

infec�on.  An�parasi�c  control    -­‐  changes  in  larvae  glycoproteome  impact  the  parasite  infec�vity.  Using  immunological  and  molecular  methods  we  will  be  looking  for  immune  recogni�on  and  response  to  the  nematode  an�gens  altered  by  plant  saponines.  Training  programme  for  MSc  students.    

b.    Cryptosporidium  and  Giardia    project-­‐  detec�on  of  waterborne  protozoa  of  public  health  interest  in  environmental  samples  (watre,  food  and  fecal  samples).  Training  programme  for  MSc  students.    

c.  The  study  on  the  helminth  communi�es  in  small  rodents  popula�ons.  Diversity,  specificity  and  the  role  of  rodents  as  intermediate  hosts  for  human  parasites,  i.e.  Echinococcus  mul�locularis.    

d.  The  ecology,  epidemiology  and  gene�c  diversity  of  �ck  transmi�ed  pathogens.  Training  programme  for  MSc  students.  

       Students  will  be  trained:  to  improve  effec�veness  in  research  and  knowledge    in  parasi�sm  and  parasi�c  diseases
