An open-label, randomized, controlled, multi-center, Phase I/II trial investigating 2 EMD 525797 doses in combination with cetuximab+irinotecan versus cetuximab+irinotecan alone, as second-line
treatment for subjects with k-ras wild type (WT) metastatic colorectal cancer (mCRC). Subjects with k-ras WT mCRC will be eligible for enrollment if they are refractory to or progressive after
first-line chemotherapy with an oxaliplatin-containing therapeutic regimen.

• Notwendigkeit von mindestens 2 Paracentesen innerhalb der letzten 4 Wochen vor Einschluss in diese Studie
• kein hämorrhagischer Aszites (Hämatokrit im Aszites > 2 %)• keine parallele Behandlung des Aszites bis auf
Paracentesen, Diurese oder dem Studienmedikament• keine parallele Antitumor-Behandlung mit anderen
Substanzen als die Studienmedikation (Chemotherapie, die mindestens 4 Wo vor Start der Studie begonnen wurde darf fortgeführt werden.

• Documented progressive disease based on investigator assessment according toResponse Evaluation Criteria in Solid Tumours (RECIST) following receipt ofcisplatin and/or carboplatin (minimum doses described below*) administeredfor recurrent and/or metastatic disease independent of whether patientprogressed during or after platinum based therapy.Cisplatin, minimum dose: at least two cycles of cisplatin, ≥60 mg/m2/cycle ora total cumulative dose of ≥120 mg/m2 during 8 weeks Carboplatin, minimum dose: at least two cycles of carboplatin area under theconcentration-time curve (AUC) ≥4/cycle or a total cumulative dose of AUC ≥8during eight weeks.

Adenokarzinom der Prostata, hormonresistent und resistent gegen Docetaxel-Vorbehandlung, Krankheitsprogreß nach vorhergehender Hormon – oder Docetaxel-Behandlung

Einschlusskriterien• Metastatic or unresectable locally advanced/recurrent breast cancer HER2-positive
disease documented as ISH-positive and/or 3+ by IHC on previously collected tumor tissue and prospectively confirmed central laboratory prior to study enrollment. Tumorgewebe muss vorhanden sein.
• Disease progression on the last regimen received as defined by the investigator.• Prior treatment with an anthracycline, trastuzumab, a taxane, lapatinib, and capecitabine
in the neoadjuvant, adjuvant, locally advanced, or recurrent/metastatic setting and documented disease progression (by investigator assessment) after at least two regimens of HER2-directed therapy in the metastatic or unresectable locally advanced/recurrent setting.
• Patients must have had at least 6 weeks of prior exposure in the metastatic (or unresectable locally advanced/recurrent) setting to lapatinib and capecitabine (given together or separately) unless they were intolerant of lapatinib and/or capecitabine. Patients who were found to be intolerant to lapatinib can be considered eligible if they experienced disease progression during a single trastuzumab-based regimen in the metastatic (or unresectable locally advanced/recurrent) setting.
Ausschlusskriterien • Brain metastases that are untreated or symptomatic, or require any radiation, surgery, or
steroid therapy to control symptoms from brain metastases within 2 months of randomization
