426 Free Papers—Oral Presentations Two ACC cell lines, ACC-2 and ACC-M, were chosen to assess the cell proliferation, the cell invasion, the cell cycle and apopto- sis change after BMP-2 treatment, by MTT, Transwell Matrigel invasion assay, flow cytometry (FCM), and Bcl-2 was examined by immunohistochemical staining as well. The level of phosphorylated extracellular signal-regulated kinase 1 and 2 (ERK1/2) was detected by Western bolt analysis. Furthermore, ACC-M cells, mixed with BMP-2 or not in Matrigel, were subcu- taneous injected into mice respectively. Tumour volume measurements began on day 7 (tumour formation) and continued three times a week until the end of this study. Results: BMP-2 was overexpressed in the cytoplasm of ACCs, and the expression ratio of BMP-2 in the solid subtype of ACCs group was higher than that of cribriform-tubular subtype, but the expres- sion ratio of BMP-2 had no correlation with the tumour size, patient gender and age. In vitro study, BMP-2 stimulated prolifer- ation of ACC-2 and ACC-M cells. Though there was no significant change on the ratio of G0/G1 phase, S phase, or G2/M phase, Bcl-2 expression was significantly upregu- lated and invasion activity of the cells was significantly enhanced after BMP-2 treat- ment. Western blot showed the p-ERK1/2 level of ACC-2 and ACC-M cell lines was remarkably upregulated 10 minutes after BMP-2 treatment. In vivo there was sig- nificant increase of the tumour volume at early stage with BMP-2 treatment. Sekine Department of Oral and Maxillofacial Surgery, Shimane University Faculty of Medicine, Izumo, Japan Background and Objectives: An accurate evaluation of cervical lymph node metas- tases is crucial for the correct management of oral cancer. A number of studies have reported the predictive factor for cervical lymph node metastases in oral cancer. This study was undertaken to confirm the feasibility of nuclear morphometric analysis of lymph node metastases of squamous cell carcinoma in the tongue. Methods: Forty nine cases of squamous cell carcinoma (25 cases with metastasised lymph nodes, 24 node-negative cases) were examined. Using computerised image sys- tem, the mean nuclear area, the nuclear perimeter, the circular rate, the largest to the smallest dimension ratio (LS ratio) of the nuclei, and the coefficient of variation of the nuclear area (NACV) were measured in the biopsy specimens. The relationship between these results and the lymph node involved has evaluated retrospectively. Results: The nuclear area and perimeter were significantly higher in cases with metastasised lymph node than in node- negative cases (P < 0.05). On the other hand, the circular rate was slightly smaller in cases with metastasised lymph node than in node-negative cases. The LS ratio and NACV were slightly higher in cases with metastasised lymph node than in node- negative cases. In lymph node positive cases, the level of lymph node metastases was in inverse proportion to circular rate. And significant difference was not found in other parameters. Kim Department of Oral and Maxillofacial Surgery, Dankook Dental Hospital, Dongnam-gu, Cheonan-si, Chungnam, Korea Background and Objectives: Recently, the role of serum tumour marker has been stud- ied for an important issue on diagnosing and treating tumours in the head and neck region because tests using tumour markers need relatively simple procedures and are acceptable to patients, compared with other test methods. Tumour marker tests were performed on patients with squamous cell carcinoma, which were known to have the highest prevalence among tumours in the head and neck region. Association between each tumour marker, and diagnosis and prognosis of tumours was assessed. Methods: Tumour marker tests were car- ried out on 31 patients who visited Oral and Maxillofacial Surgery Department in Dankook University Dental Hospi- tal between January 2003 and August 2008 and who were diagnosed as primary oral squamous cell carcinoma throughout histopathologic diagnosis. Blood samples from these patients were analysed to measure tumour markers using nuclear medicine diagnostic equipment. Measured entries were as follows: PSA (prostate- specific antibody), SCC-Ag (squamous cell carcinoma related antigen), CA 19-9 (cancer antigen 19-9), ferritin, -FP (alpha-fetoprotein), CYFRA 21-1 (cytokeratin-19 fragments), CA125 (can- cer antigen 125) and p53. Results: Analyses on each tumour marker indicated that squamous cell carcinoma in the head and neck region had statis- tically significant correlation with p53, SCC-Ag (TA-4), CYFRA 21-1 and ferritin. p53 demonstrated the highest sensitivity. Especially, 4 cases among 18 cases which Ferritin was measured exhibited metasta- sis. In all those 4 cases, Ferritin values were higher than the standards (15–332 ng/mL). Therefore, ferritin is considered to have a close relation with metastasis of squamous cell carcinoma. Conclusions: This study shows that tumour marker tests are more useful in evaluat- ing progression and prognosis of tumours rather than in diagnosing them.
