Effecten van interventies op in-vitro gekweekte
Intestinal Porcine Epithelial Cells (IPECJ2)
• Neonataal; interventie in kuikens met Octaciline = Amoxiciline;
Octacillin getest in IPECJ2; (vergelijking ook met ZnO-VDI-5; biggen/IPECJ2)
• Maternaal; interventie in drachtige zeugen – effect op biggen van Amoxiciline;
Paracilin getest in IPECJ2
(veel overeenkomsten Paracilin en Octacilin; Octacillin wordt besproken !)
• Neonataal; interventie in biggen met Lange (Lc) en Korte (Sc) keten Fructo-
oligosaccharides
In-vivo; 10% Lc-FOS and 90% Sc-FOS / In-vitro IPECJ2; apart Lc and Sc-FOS
(Frutafit® TEX [long chain FOS] / Frutalose® OFP [short chain FOS] SENSUS).

HIF1A effector genes/ proteins
ZnO/Octaciline induced oxidative stress (ROS)
IPECJ2 enterocytes react by secretion of
HIF1A-effectors like HMOX1, SERPINE1 and
EDN1 to widen veins to supply and stimulate
uptake of oxygen and glucose from the blood.
To survive from oxidative stress IPECJ2 cells
promote anaerobic energy metabolism by
regulating genes involved glucose, pyruvate,
and fructose transport (HK, SLC2A1, PDK1,
PFKFB3 and PGK1).
The interplay between IL6 and MT’s may be a
crucial mechanism to cope with ROS and in
protecting IPECJ2 cells from a ROS overload.

• Genen/processen geïdentificeerd bruikbaar als/voor...
o meetlat/uitleesparameter voor darmfunctie
o de ontwikkeling van alternatieve stoffen voor bestaande interventies
o verschaffen inzicht in functie van de darm
• In vitro “bioassay” ontwikkeld / gevalideerd
-Bruikbaar voor het pre-screenen van “voer additieven” in IPECJ2 cellen (goedkoop t.o.v. in-vivo).
-Deels toepasbaar voor kippen /en rund ?
• Interessante resultaten in-vitro
> Overlap respons Amoxicilline / ZnO (alternatieven voor antibiotica/ZnO)
> IPECJ2 test ook te gebruiken om effect van fermentatie producten
microbiota te meten ?
