• Cell culture and cryo-storage - Generation of human induced pluripotent stem cells (hiPSC) and bonemarrowmesenchymalstemcells.Dailymaintenancethroughtheobservationofcellhealth(visualizationofcells with light microscope and culture media assessment) and then culture of cell lines (feeding,division/expansionandcryostorage).

• Characterizationofpluripotentstemcellandmesenchymalstemcells:-AmajorstageinmyPhDstudies

the characterization of the cell lines through quantitative PCR, immunofluorescence staining,immunohistochemistryandflowcytometry.

• Stem cell differentiation studies – After the successful maintenance of induced pluripotent stem cell

cultures,thecoreofmyPhDwastodesigndifferentiationprotocoltoinducechondrogenicdifferentiationin the human induced pluripotent stem cells. This required extensive understanding of embryonicdevelopmentandcellsignallingpathways.

• 3DTissueengineering–Thiswasimportanttoinvestigatetheabilityofthedifferentiatedhumaninduced

pluripotent stem cells chondroprogenitors to generate a 3D cartilage construct suitable for cell-basedtherapies. The Stem Cell Biology group in the University of Bristol, using mesenchymal stem cells,polyglycolicacidscaffoldandrecombinantgrowthfactors,developedthetissueengineeringprotocol.ForthehiPSC,myPhDwastoinvestigatethefactorsthatinduceitschondrogenicdifferentiation.

• ELISA and colorimetric assay use and result processing - This was important in the measurement of

cartilageextracellularmatrixcomponentssuchastypeIandtypeIIcollagensandsulphatedproteoglycans.

• Histology and immunohistochemistry – These techniqueswere used to analyze the cellularmorphologyand the quality of the 3D tissue engineered cartilage using a wide range of stains. During my PhD Iexperiencedinthedevelopmentandoptimizationofimmunohistochemistrystains.

• Dataanalysis–ThroughoutmyPhDdataanalysishasbeen important.DuringmyrecentPhDstudies the

statisticalpackagePRISM(v5.0c)wasusedtoinvestigatewithin/betweengrouptrends.

• Cell signaling experimentation –Daily experimentation at theUniversity of Bristol was the treatment ofhiPSCwithvarious factors toactivatesignalingpathways involved in theself-renewalanddifferentiation.ThisincludedtheTGFβ,BMP,FGF,andActivinsignalingpathways.

• Blood transfusion and blood banking skills - During my work at King FahdMilitaryMedical Complex in

Dhahran, I experienced all routine and specialized blood banking tests that included ABO +RH forwardgrouping, reversegrouping,Antibodyscreeningand identification,directand indirectantihumanglobulintest, Compatibility testing, resolving ABO discrepancies, acid elution, pre-warm technique, salinereplacementat4oC,coldagglutinins,TypespecificindirectantihumanglobulintestTSIAT,Neutralizationbypooledplasmaandantigentyping).

• Donorregistry,selectionandphlebotomy–Iinstalledacomputerizedprogrammeforthedonorregistryat

KFMMC.Along-sideMrJosephRobinson(Bloodbanksupervisor)tookdecisionroleindonorselection.

• Bloodcomponentpreparationandstorage-duringmytimeatKFMMC,bloodcomponentpreparationandstoragewasanimportanttaskinmyjobdescription.

• Generationandmaintenanceoflabrecordsandstatistics.Adheretostatutoryhealthandsafety

requirements.
