Schematic diagram of the amplification process with the GenomiPhi amplification method. Random hexamer primers anneal to the template DNA at multiple sites. Phi29 DNA polymerase initiates replication at multiple sites on the denatured linear DNA simultaneously. As synthesis proceeds, strand displacement of complementary DNA generates new single-stranded DNA. The subsequent priming and strand displacement replication of this DNA results in the formation of double-stranded DNA.

Animal serum origins Serum origin refers to the country in which the raw blood was collected, and is not to be confused with the country in which the finished product was processed. With HyClone serum products, the country or countries of origin are stated in the certificate of analysis that you receive with your product. The supply of FBS, a byproduct of the beef industry, is impacted by complex market conditions. For these reasons, we ensure adequate supply for our customers by not only sourcing FBS from several regions, but also working with a broad base of serum and raw blood suppliers. To ensure traceability and security of supply, raw serum is obtained in New Zealand, Australia, the United States, Canada, Central America, and South America. Moreover, we are traceability certified by the International Serum Industry Association (ISIA) and send samples of characterized and defined FBS to Oritain for independent testing.

Industry leading traceability program gives peace of mind
In addition, we go beyond industry standards of document traceability by performing independent testing by Oritain™ on each batch of characterized and defined FBS from the United States, Australia and New Zealand. Look out for the Oritain logo on your FBS bottle.
Origin of sera is important. As proof that we ensure proper origin, traceability, and truth in labeling, we are traceability certified by the International Serum Industry Association (ISIA).

The Testament to QualityThere can be few companies anywhere in the world can lay claim to such distinguished clients as Whatman in the past 250 years.
In November, 1767, the artistic Thomas Gainsborough wrote; “I beg you to accept my sincerest thanks for the favour you have done me concerning the paper for drawings. I had set my heart upon getting some of it, as it is so completely what I have long been in search of… Upon my honor I would give a guinea a quire for a dozen quires of it. “
Whatman paper became enormously popular with leading artists such as J M W Turner, and the quality and durability of English papers, including those made by Whatman, gave the English watercolour school a 50 year advantage over European artists. At the end of the 18th century, the erratic genius William Blake used Whatman papers for four of his illuminated books, the public being informed that these were printed on “the most beautiful wove paper that could be procured.”
Throughout history, heads of state and world leaders have shown a particular penchant for Whatman paper. Napoleon sat for five hours on the bleak island of St. Helena writing his long and detailed will on Whatman paper only three weeks before his death in 1821. George Washington signed many state documents on Whatman paper. Queen Victoria chose Whatman paper for her personal correspondence.
In the 1930’s Soviet leaders used Whatman paper to published the 5 year plan for the future of the USSR, while the peace treaty with Japan was signed on Whatman paper at the close of World War II.
Today, Whatman filters have gained universal acceptance among the major scientific and industrial concerns of the developed world, from Japan to the United States, the UK to southeast Asia.
A remarkable testimony to Whatman quality – achieved through the pursuit of excellence.*

Protein purification Protein purification methods can be organized as a single or multi-step chromatographic strategies to optimize the required purity and yield. A strategy can comprise capture(C), intermediate purification (iP), and polishing steps (P).
• Capture or affinity chromatography is where the target protein of interest is isolated, concentrated, and stabilized.
• Intermediate chromatography is used to remove contaminants with techniques including IEX, CIEX, and HIC.
• Polishing is utilized to remove the most difficult impurities and aggregates of the target protein to increase purity.
Depending on your downstream analysis technique, a single or multi-step chromatographic process can be followed by sample desalting, buffer exchange, and concentration.

Affinity chromatography – Tagged proteinsAffinity chromatography step enables the isolation of target tagged protein from initial sample (e.g., lysate, cell culture). After this step, the purity level is > 80%.
If a higher purity level is required, then it is recommended to add extra steps, such as IEX and/or SEC. See pages 24 and 25

Affinity chromatography – AntibodiesAffinity chromatography step enables the isolation of target anitbody from initial sample (e.g., lysate, cell culture). The purity obtained after this step is usually high (above 90%). However, to further remove antibody aggregates and/or fragments, it is then recommended to add a SEC step (see page 24-25). The purity level will then be very high (From 95% to 99%).

Western Blotting DetectionECL Western Blotting Detection Reagents: ECL based on horseradish peroxidase (HRP)-conjugated secondary antibodies has become the most commonly used detection method for Western blotting. It is a sensitive detection method, where the light emission is proportional to protein quantity. Minute quantities of proteins can be detected and quantitated.
• Longer shelf life: 18 month shelf life on ECL Select and Prime Products
• Stability: ECL products are stable and stored at room temp
