AIMThe aim of this study is to investigate the applicability of the molecular diagnostic tool to analyze and to evaluate the “detection of katG S315T mutation in INH (isoniazid) monoresistant M. tuberculosis clinical isolates by performing standard molecular techniques”. OBJECTIVES OF STUDY•To isolate and identify M. tuberculosis from the sputum sample.•To extract genomic DNA from the grown M. tuberculosis culture.•To perform PCR for the amplification of IS6110 for species confirmation.•To perform multiple allele specific-PCR for the amplification of katG gene and detection by PCR RFLP.•To confirm and analyze the mutation in katG gene by using automated DNA sequencing. •To scrutinize the mutations and correlate the result with wild strain H37RV using bioinformatics tools.
