METODE BAKU HISTOLOGI PEMBUATAN SEDIAAN PENELITIAN

A. Cara pembuatan sediaan bahan penelitian
1. Mengambil jaringan yang dibutuhkan dengan segera mungkin setelah mencit
dimatikan (kurang 2 jam) dengan ukuran 1x1x1 cm3.
2. Memasukkan ke dalam larutan fiksasi dengan urutan sebagai berikut :
a. Fiksasi dengan formalin 10%
b. Dehidrasi dengan alkohol 30 % selama 20 menit I, 20 menit II dan 20 menit
III.
Lalu dilanjutkan dengan alkohol 40 % 1 jam
alkohol 50 % 1 jam
alkohol 60 % 1 jam
alkohol 70 % 1 jam
alkohol 80 % 1 jam
alkohol 90 % 1 jam
(alcohol 70-80 % dapat ditunda sampai keesokan harinya)
c. Larutan xylol alcohol 1:1 dengan waktu kurang lebih 24 jam
d. Clearing dengan larutan xylol 1, 2, 3 dengan waktu masing-masing 20
menit sehingga jaringan terlihat tembus pandang.
e. Xylol paraffin 1:1 selama 20 menit/24 jam dengan dipanaskan dalam oven
60oC.
f. Embeding dan Blocking : Parafin 1, 2, 3 selama 20 menit lalu jaringan
dcetak blok paraffin, kemudian didinginkan kurang lebih 24 jam sehingga
cetakan dapat dibuka
g. Trimming : memotong blok-blok paraffin dengan mikrotom
B. Cara pemotongan blok
1. Menyiapkan kaca objek bersih
2. Kaca objek diberi albumin di tengahnya, direkatkan
3. Blok yang sudah disiapkan dipotong dengan ketebalan 5 mikro, lalu
dimasukkan air panas 60oC. Setelah jaringan mengembang, jaringan diambil
menggunakan kaca objek yang sudah diberi albumin. Kemudian keringkan.
4. Parafin yang ada pada kaca objek atau jaringan dihilangkan dengan
dipanaskan dalam oven 60oC atau dalam tungku.
C. Pewarnaan
Slide jaringan dimasukkan ke dalam :
1. Xylol 1,2,3 masing-masing 10 menit
2. Rehidrasi dengan alkohol xylol 5 menit
3. Bilas alkohol 96%-30% masing-masing 30 menit
4. Bilas aquades 1x 10 menit
5. Rendam dengan hematoksilin 10 menit
6. Bilas dengan air mengalir sampai bersih
7. Bilas aquades lalu acid alkohol (alkohol + NaCl 0.9%)
8. Bilas alkohol 50%-96%
9. Eosin 2-5 menit
10. Bilas alcohol 96% 2x, bilas alkoho lxylol
11. Keringkan dengan kertas saring, langsung dibersihkan kotoran-kotoran
yang ada di sekitar jaringan.
12. Xylol 1 (5 menit), xylol 2 (5 menit), tetesi basa Canada, langsung ditutup
dengan kaca penutup

DOSIS KONVERSI FLUORIDA DALAM PASTA GIGI
Penetuan dosis fluoride dalam pasta gigi yang akan diencerkan.
Faktor konversi dosis manusia ke dosis mencit = 0,0026
Panjang sikat gigi :
a. Sikat gigi anak : 2,2 cm = 22 mm
Kandungan fluorida dalam pasta gigi ;
a. Pasta gigi dewasa : 1,12 % Sodium monofluorophospat
b. Pasta gigi anak : 0,4 % Sodium mono fluorophospat
1 gram pasta gigi setara dengan 1 mg F, setara dengan panjang pasta gigi 12mm.20
Perlakuan yang akan diberikan :
a. Kelompok I : Kelompok kontrol
b. Kelompok II : Perlakuan I
Pasta gigi dewasa sebesar biji jagung = 0,25 g. 52
Konversi ke mencit : 0,25 gram x 0,0026 = 0,00065 gram = 0,65 mg pasta
0,65 mg ~ 0,7 mg pasta
Kandungan fluoride : 1,12 % x 0,65 mg = 0,0073 mg F
c. Kelompok III : Perlakuan II
Pasta gigi anak sepanjang sikat gigi anak : 22 mm , 22/12 = 1,83 gram pasta gigi
Konversi ke mencit : 1,83 gram x 0,0026 = 0,004758 gram = 4,758 mg pasta
4,758 mg ~ 4,8 mg pasta
Kandungan fluorida dalam dosis mencit : 0,4 % x 4,8 mg = 0,019 mg F
d. Kelompok IV : Perlakuan III
Pasta gigi dewasa sepanjang sikat gigi anak : 22mm, 22/12 = 1,83 gram pasta gigi.
Konversi ke mencit : 1,83 gram x 0,0026 = 0,004758 gram = 4,758 mg pasta
4,758 mg ~ 4,8 mg pasta
Kandungan fluoride : 1,12 % x 4,8 mg = 0,054 mg F

CARA PENGENCERAN PASTA GIGI BERFLUORIDA
Pasta gigi ditimbang sesuai dengan dosis yang diharapkan lalu diencerkan
dengan air hingga 0,5 ml (sesuai kapasitas lambung mencit) kemudian dimasukkan
peroral ke mencit dengan bantuan sonde lambung.
Pengenceran pasta gigi dilakukan di Laboratorium Fakultas MIPA Universitas
Negeri Semarang.
