Summary of specific aims of grant: To address the role of ROP signaling pathways
through aptamer-based proteomics. Firstly, we will study the relationship of high signal
proteins in plasma samples taken early in the postnatal period with severe ROP.
Secondly, we will develop a statistical model to improve severe ROP risk assessment
based on panels of high signal proteins and established ROP risk factors. Thirdly, we will
study trends in concentrations of proteins in infants who develop severe, low-grade and
no ROP. Finally, we will determine if levels of high signal proteins and other proteins
measured by the technology around the time of the first ROP examination are related to
the development or severity of ROP.

Summary of specific aims of grant: The National Institute of Child Health and
Development recently released a request for proposals inviting sites across the United
States to be part of the National Children’s Study (NCS). Here in Colorado, the Denver
Metro area and Douglas County have been chosen as potential sites for the study. The
aim of the study is to recruit 250 pregnant women per year for 4 years at each site. The
women will be recruited from the community and monitored closely throughout
pregnancy by a team of investigators. The children will be followed for 21 years. During
pregnancy and after birth information will be collected on many exposures and outcomes.

Summary of specific aims of grant: There is widespread use of antenatal dietary
supplementation with the goal of preventing in utero and neonatal abnormalities, but
there has been little investigation about the potential risks of such interventions in
expectant mothers. Folic acid, vitamin B12, and choline are methyl donors that, taken
during pregnancy, have the potential through hypermethylation to alter the expression of
genes that regulate development of allergy, increasing the risk in babies for developing an
allergic disease. Our research team proposes to determine if such concerns about methyl
donor supplementation are justified and pose a real threat in expectant mothers already at
risk for transmitting an atopic genetic predisposition to their infants.
