RESEARCH INTERESTS

– Retinopathy of Prematurity

– Age-related Macular Degeneration

– Outcomes from cataract surgery

– The complement system in human disease

– Links between events in the intra-uterine environment with Retinopathy of Prematurity

– The role of circulating proteins in eye diseases

RESEARCH ACTIVITIES, SUPPORT AND GRANTS

Active Research Projects, Support and Grants:

1. Postnatal growth and retinopathy of prematurity (G-ROP)

PI: Gil Binenbaum MD

Role: Co-Investigator

Agency: National Eye Institute (NEI)

Grant number: 5 R01 EY021137 03

Page 12: CURRICULUM VITAE Anne M. Lynch, MB, BCh, BAO, … · 1983 Diploma in Obstetrics (DObst), ... 2010 Assistant Professor, Preventive Medicine and Biometrics, Department of ... Seminar,

Anne Lynch, MD, MSPH Last revised October 11, 2017 Page 12 of 36

Period: 09/30/12 - 08/31/17

Direct costs: $10,000 (approx.) per year to the Dept. Ophthalmology

Summary of specific aims of grant: To develop a prognostic model using postnatal

weight gain for severe ROP.

Grants Under Review:

1. Aptamer-based Models for Predicting Retinopathy of Prematurity

Role: Principal Investigator

Agency: National Eye Institute (NEI)

Grant number: R01EY028122

Period: 04/1/2018 - 03/31/2023

Direct costs: $2,689,408 Indirect costs: $567,056 Total Costs: $3,256,464

Summary of specific aims of grant: To address the role of ROP signaling pathways

through aptamer-based proteomics. Firstly, we will study the relationship of high signal

proteins in plasma samples taken early in the postnatal period with severe ROP.

Secondly, we will develop a statistical model to improve severe ROP risk assessment

based on panels of high signal proteins and established ROP risk factors. Thirdly, we will

study trends in concentrations of proteins in infants who develop severe, low-grade and

no ROP. Finally, we will determine if levels of high signal proteins and other proteins

measured by the technology around the time of the first ROP examination are related to

the development or severity of ROP.

Completed Research Projects, Support and Grants:

1. Complement Activation and Hypertensive Diseases of Pregnancy

Role: Principal Investigator

Agency: Eunice Kennedy Shriver NICHD

Grant number: 1 R21 HD068961-01A1

Period: 12/01/2011 - 11/30/2013 (in extension period 2014)

Direct costs: $420,750

Percent effort: 20%

Summary of specific aims of grant: To determine the association between complement

activation fragments measured at multiple time points in gestation with subsequent

development of hypertensive diseases of pregnancy in a longitudinal study design.

2. Exploring the Fuel-Mediated Programming of Neonatal Growth

PI: Dana Dabelea MD, PhD

Role: Co-Investigator

Agency: National Institute of Diabetes, Digestive & Kidney Diseases (NIDDK)

Grant number: 1R01DK076648

Period: 9/1/09 - 7/31/2014

Direct costs: $496,000 per year

Percent effort: 2.5%

Summary of specific aims of grant: This project will explore a timely public health

problem by testing the hypothesis that maternal obesity programs neonatal growth,

fatness and metabolism, and by identifying specific mediators of these effects that can be

targeted by future interventions. We will enroll 1,920 pregnant women before 15 weeks

Page 13: CURRICULUM VITAE Anne M. Lynch, MB, BCh, BAO, … · 1983 Diploma in Obstetrics (DObst), ... 2010 Assistant Professor, Preventive Medicine and Biometrics, Department of ... Seminar,

of gestation and follow them prospectively through delivery in order to explore the

relationships between maternal body size and behaviors during pregnancy, intra-partum

fuels, markers of inflammation and insulin resistance, and infant body size and fatness.

3. To Support the Data Management Core of the Colorado “Baby Blanket” Perinatal

Summary of specific aims of grant: The National Institute of Child Health and

Development recently released a request for proposals inviting sites across the United

States to be part of the National Children’s Study (NCS). Here in Colorado, the Denver

Metro area and Douglas County have been chosen as potential sites for the study. The

aim of the study is to recruit 250 pregnant women per year for 4 years at each site. The

women will be recruited from the community and monitored closely throughout

pregnancy by a team of investigators. The children will be followed for 21 years. During

pregnancy and after birth information will be collected on many exposures and outcomes.

9. Antenatal Dietary Supplementation is a Risk Factor for Infant Atopy Through

Epigenetics

PI: Pia Hauk MD, National Jewish Health

Role: Co-Investigator

Agency: National Heart, Lung, and Blood Institute (NHLBI)

Period: 8/1/10 - 9/1/2011

Type: Challenge grant

Direct costs: $33,434

Percent effort: 4%

Summary of specific aims of grant: There is widespread use of antenatal dietary

supplementation with the goal of preventing in utero and neonatal abnormalities, but

there has been little investigation about the potential risks of such interventions in

expectant mothers. Folic acid, vitamin B12, and choline are methyl donors that, taken

during pregnancy, have the potential through hypermethylation to alter the expression of

genes that regulate development of allergy, increasing the risk in babies for developing an

allergic disease. Our research team proposes to determine if such concerns about methyl

donor supplementation are justified and pose a real threat in expectant mothers already at

risk for transmitting an atopic genetic predisposition to their infants.

Grant number: K23 HD049684

Type: K23

Period: 9/12/2006 - 8/31/2010

Direct costs: $275,000

Percent effort: 75%

Summary of specific aims of grant: Pregnancy loss is the most common complication of

pregnancy. Recent studies in experimental murine models suggest a significant

contribution of complement activation to immunologically-related pregnancy loss. The

aim of this proposal is to translate these observations to a human study in the setting of a

Mentored Patient-Oriented Research Career Development Award. The primary aim of

this study is to determine whether elevated levels of split products generated by the

alternative or classical pathway of complement activation are associated with pregnancy

loss.

Summary of specific aims of grant: The specific aims of this project are to determine

between 10 and 20 weeks gestation: 1) If levels of the complement activation fragments

C4a, C3a, SC5b-9 predict the development of preeclampsia later in pregnancy, 2) If

levels of proangiogenic PlGF and the antiangiogenic factors sFlt-1 and sEng predict the

development of preeclampsia later in pregnancy, 3) If there is an association between

complement activation fragments and angiogenic factors.

early pregnancy: 1) If there is an association between the complement activation

fragment Bb and antiangiogenic factors, 2) If antiangiogenic factors are risk factors for

preeclampsia 3) If elevated levels of antiangiogenic factors and the complement

activation product Bb together are better predictors of preeclampsia than individual

measures of these markers.
