Step into the world of Cryopreservation
4
Cryopreservation refers to the storage of a living organism, cell or tissue atultra-low temperatures such that it can be restored to the same viable stateas before it was frozen. Storage for an indefinite amount of time requiressamples to be maintained below the glass transition temperature ofaqueous solutions, approximately -130ºC, the temperature at which frozenwater no longer sublimes and recrystallizes. Therefore -150°C mechanical freezers or liquid nitrogen storage tanks are requiredfor long-term preservation.
In the late 19th century Sir James Dewar, a Scottish chemist and physicist,designed a vacuum jacketed vessel capable of effectively storing liquidgases, such as nitrogen, for long periods of time. This type of vessel is stillbeing used today. For years cryopreservation in liquid nitrogen at -196°Chas been the standard for long term preservation.
Over the years it became clear that cross contamination was a real risk withthis kind of preservation. Not only can samples be contaminated by thenitrogen itself, but also by other samples. Evidence of cross contaminationhas been documented in the past, including a well known example ofHepatitis B transmission.
For this reason, storage in vapour phase became more popular. Valuablesamples were no longer stored in liquid phase nitrogen to eliminate the riskof cross contamination.
There are also disadvantages of storage in vapour phase though:1. Loss of storage capacity; The bottom third of the vessel cannot
be used as there is liquid phase nitrogen there.2. The temperature provided by vapour phase storage is highly dependent
on the level of liquid nitrogen in the bottom of the vessel.3. Large vertical temperature gradients can put valuable
samples at greater risk during long term storage.
Dry storage systems like -150ºC mechanical freezers and patentedIsothermal storage systems have been developed to offer the highest levelsof protection for valuable samples, without the risks of cross contaminationand vertical temperature gradients.

Due to the extended operation of compressors within ultra-lowtemperature freezers, lubricant oil is essential to reduce wear,prevent abrasions and seizure and therefore maintain the highestlevels of freezer performance. However, when lubricant oil circulateswithin the refrigeration circuit, it can cause piping to become clogged,resulting in compressor damage. Incorporating a high-efficiency oilseparator, Panasonic -150ºC freezers effectively separate lubricant oilfrom refrigerant, increasing the durability of the compressors andoffering a reliable ultra-low temperature environment.
Highly efficient compressors have been specially developed andincorporated our -150ºC freezers. Microprocessor control with LED digital displayallows accurate temperature setting and a stable -150ºC environment.Accurate temperature setting and a stableperformance are provided by a microprocessorcontroller with an easy to use LED digital display andflat key data entry. The MDF-1156 maintains an ultra-low temperature of -152ºC, far lower than therecrystallization point for pure water (-130ºC). This low temperature provides the ideal preservationenvironment for long-term storage.
Alarm and safety devices ensure high levels ofprotection for valuable samples.A high temperature alarm alerts users when thetemperature of the freezer deviates 10ºC or 15ºCabove the set temperature and a power failure alarmis activated if power to the freezer is lost. As well aslamp and buzzer functions, a remote alarm contactcan be set up to alert users to these abnormalitiesoutside of working hours. ATN models are also equipped with an auxiliary liquidnitrogen back-up system for added protection.

Isothermal LN2 freezersNo liquid nitrogen contactLiquid nitrogen is, like all liquids, a possible transporter of contaminants. Contaminationbetween samples, leakage of liquid nitrogen into sample vials during storage and contact ofliquid nitrogen with skin are just some of the risks that can be eliminated with an Isothermalliquid nitrogen dry storage freezer from Custom BioGenic Systems (CBS).
CBS Isothermal freezers feature a patented liquidnitrogen jacket to provide uniform storage temperaturesin the -190ºC range, free from liquid nitrogen contact.
In the year 2000 Custom Biogenic Systems patentedthe first -190ºC dry storage system. This new storagesystem uses the same vacuum insulated vessel asconventional LN2 freezers, but instead of the liquidnitrogen being in the sample storage space of thefreezer, it is in a patented liquid nitrogen jacket in thewall of the freezer. This liquid nitrogen jacket producesa unique vapour circulation in the freezer. This vapourcirculation, in combination with the convection fromthe freezer walls, produces a very stable temperaturein the -190ºC range without the large verticaltemperature gradients found in traditional vapourphase LN2 storage tanks. The minimal temperaturegradient and lack of liquid nitrogen in the bottom ofthe vessel means that the full internal volume of eachfreezer is available for storing samples at liquidnitrogen temperatures, making the most efficient useof capacity.
The innovative design of CBS Isothermal freezerseliminates a number of major risks of traditional LN2storage, including:l Cross contamination: Studies have shown that viral,bacterial and fungal pathogens can survive aftersuspension in liquid nitrogen. Infected samples cancross contaminate other samples in the same liquidnitrogen tank.l Loss of samples: Storage directly in liquid nitrogencan make vials shrink. This may cause liquid nitrogento seep into the vials, which on re warming, expand andsubsequently explode as nitrogen vapour izes insidethe vials. l Health and safety: Traditional storage in liquidnitrogen exposes users to direct contact with LN2which can lead to cold skin burns.

The Isothermal conceptThe sample storage area iscooled by a liquid nitrogen jacketsurrounding the stainless steelinterior, and by nitrogen vapourentering the freezer from thejacket via directional vents. Thispatented technology providesexceptional temperatureuniformity in the -190°C range,allowing the full freezer capacityto be used with confidence. Thecirculation of vapour within thefreezer also results in less coldair loss during lid opening andimproved visibility. This allowsfull-width lids to be usedproviding quick, unrestrictedaccess to sample racks.
No liquid nitrogen contactWith no liquid nitrogen in thestorage area, samples can bestored safely in the -190°C rangewithout the risk of cross-contamination through liquidnitrogen. The Isothermal designalso provides added user safetywith no splashing or contact ofliquid nitrogen when removingracks and samples from thefreezer.

Automatic operationIsothermal freezers feature the series 2301 auto-fill and monitorsystem, which controls the automatic filling of the liquid nitrogenjacket and provides the user with an easy to read overview of thefreezer temperature and status.
Sample securityA comprehensive alarm system with remote alarm contactconstantly monitors all aspects of the freezer’s operation. Samplesare also protected by lid and control panel locks. The freezer canbe monitored by a central BMS or monitoring system.

Isothermal carouselEasy access to samples from the front of the freezer
The Isothermal carousel liquid nitrogen vapour storage system combines the innovative -190°C Isothermal design with a small opening and an interior rotating carousel. The smallopening provides low liquid nitrogen consumption, a lightweight, user-friendly lid and aconsistently low temperature in the freezer. The interior carousel is rotated by a uniqueratcheting handle on top of the freezer that will position samples to the front of the freezer,even with the lid on.
Each Isothermal carousel system includes theadvanced 2301 auto- fill/monitoring controller toprovide security and ease of operation. There areseveral Isothermal carousel models available andan extensive selection of inventory racks to storeany size tube, vial, box, microplate, etc. to completethe system. Custom configurations can also bedesigned to meet any requirement.

Five liquid nitrogen freezers are available ranging insizes from 90 litres to 720 litres, with sample storagecapacities up to 40,000 2 ml vials or over 3,500 bloodbags. Each unit includes a liquid level auto-fill and alarmsystem to provide security, ease of operation, plusflexibility to operate as vapour storage, immersionstorage or a combination of both to store any type or sizeof sample.
