13 Chapter 6  Combined Proteolytic Pre-treatment and HIER Introduction Some antigens are more efciently retrieved by a combination o heating and enzyme digestion, e.g. some cytokeratins and immunoglobulin light chains. The protocol below describes a method o frst treating with Proteinase K and then HIER by either water bath or microwave method. Materials Required Silanized Slides (Code S3003) or slides coated with other suitable adhesives Target Retrieval Solution, pH 6 (Code S1700, S1699, K8005 or K8015)* Tris-Buered Saline (Code S3001) Tris-buered NaCl Solution with Tween 20 (TBST), pH 7.6 (Code S3006) *Other target retrieval solutions will work with a similar protocol optimized accord ing to individual laboratory requirements. Protocol 1. Deparafnize and rehydrate tissue sections. 2. Cover tissue sectio ns with Prote inase K and incubate or 5-10 minutes. 3. Rinse with distilled water and place in T ris-Buered Saline. 4. Proceed to antigen retrieval using either PT Link, another water bath or microwave method below. HIER - Water Bath 5. Fill container with enough retrieval solution (200 mL) to cover slides and equilibrate to 95-99 °C in water bath. Place the incubation conta iner into the water bath and incubate or 20-40 minutes. 6. Remove the container rom the water bath and cool the contents with the lid removed or 20 minutes at room temperature. 7. Rinse with T ris-Buered Saline (TBS) or distilled water at room temperature. 8. T ranser slides t o T ris-Buered NaCl Solut ion with Tween 20 (TBST), pH 7.6 Wash Buer. 9. Proceed with IHC staining.

Protocol

3. Rinse with distilled water and place in Tris-Buered Saline.

method below.

HIER - Water Bath

5. Fill container with enough retrieval solution (200 mL) to cover slides and equilibrate to

95-99 °C in water bath. Place the incubation container into the water bath and incubate

or 20-40 minutes.

6. Remove the container rom the water bath and cool the contents with the lid

removed or 20 minutes at room temperature.

7. Rinse with Tris-Buered Saline (TBS) or distilled water at room temperature.

8. Transer slides to Tris-Buered NaCl Solution with Tween 20 (TBST), pH 7.6 WashBuer.

9. Proceed with IHC staining.

HIER - Microwave

5. Fill incubation container with enough retrieval solution (200 mL) to cover slides

and insert slide holder. Insert slides in holder and cover.

6. Place the incubation container into microwave oven and incubate or 2 x 5 minutes.

7. In between steps 4 and 5, fll up the container with enough distilled water (50 mL)

to cover slides.

8. Ater the second treatment, leave the sections in the retrieval solution at room

temperature to cool or 15-20 minutes.

9. Rinse with distilled water.

10. Proceed with IHC staining.
