- 단백질은 생명체에서 가장 다양한 고분자 물질이며, 생명현상에 있어 아
주 중요한 기능을 수행한다

= 단백질은 아미노산의 선형중합체 임.

아미노산 (Amino acids) : 단백질의 기본단위
- amino acids: a-carbon, amino group, carboxylic group, hydrogen atom,
and a distinctive R-group.
- Side chain: R-group of amino acid
- L- and D-amino acid

2.1 단백질의구성성분: 20 가지아미노산
1) L-amino acids가 단백질에서 발견됨
- 왜 L-amino acids만 발견되는지 모름
- 중성 pH에서 쌍극성 이온 (Zwitterion (dipolar ions))
= pKa of carboxylic acid group: 2.0
= pKa of amino group: 9.0
2) Amino Acids
- 아미노산은 side chains의 성질에 따라 분류함.
= size, shape, charge, hydrogen bonding capacity, hydrophobic character,
chemical reactivity
2-1) 소수성 amino acids
- Simple amino acids: Glycine (Gly, G) and Alanine (Ala, A)
- 탄화수소성 amino acids: Valine, Leucine, Isoleucine, Methionine, and Proline
= Hydrophobic side chain: 단백질 구조에서 소수성 효과를 냄
= Val, Leu, Ile : branch chained amino acids
= Met: thioether group
= Proline: side chain connects

- 방향족 amino acids: Phenylalanine, Tyrosine, and Tryptophan
= Aromatic ring: benzene ring structure
= Phenylalanine: hydrophobic
= Tyrosine: hydroxyl group

= Tryptophane: indole group (two ring structure with amine group)
2-2) 극성 amino acids: Serine, Threonine, Asparagine, Glutamine, and
Cysteine. (Hydrophilic)
= Serine and Threonine: hydroxyl group.
= Asparagine and Glutamine: carboxamide group.
= Cysteine: sulfhydryl (thiol) group.
2-3) 극성 전하를 띈 amino acids: Lysine, Arginine, Histidine, Glutamic acid,
Aspartic acid (Hydrophilic)
= Acidic amino acid: Glutamic acid, Aspartic acid.
= Basic amino acid: Lysine (primary amine group), Arginine (Guanidium
group), Histidine (imidazol group).

3) 왜 단백질에는 20개의 amino acid만 존재할까?
- Diversity: 단백질의 다양한 기능을 수행할 작용기로 작동함.
- Availability: 생명체가 탄생하기 이전에 존재하고 있었음.
- Low reactivity: 반응성이 높은 amino acid는 다른 화합물로 전환되었음.
- Proteins: linear polymers formed by linking the a-carboxyl group of AA
and a-amino group of other AA. (Peptide bond)
- Peptide bond: Dehydro-condensation reaction
- 펩티드 결합의 형성은 에너지를 요구하는 과정임.
2.2 일차구조: 아미노산은펩티드결합에의해 polypeptide 사슬을형성
1) Polypeptide: 펩티드 결합에 의해 연결된 아미노산 중합체
- 잔기 (Residues): Each amino acid in polypeptide
- Polypeptide는 방향성을 가짐 (same as nucleic acids)
= N (amino)-terminal: Beginning of a polypeptide.
= C (carboxyl)-terminal: End of a polypeptide
= 주사슬 (Main chain; backbone): peptide bond and a-Carbon chain
= 곁사슬 (Side chain): 아미노산의 곁사슬 (R-group)
= 펩티드결합은 수소결합을 할 수 있는 능력이 있음: hydrogen acceptors
(carbonyl carbon)과 hydrogen donor (N-H 결합)
수소결합은 단백질의 구조를 안정화 시킴.
2) 단백질의 분자량
- 일반적인 단백질은 50에서 2000개의 아미노산으로 구성됨
- Largest protein: titin (27,000 AAs)
- 아미노산의 평균 분자량: 110 g/mol
= Polypeptide molecular weight: 5500 – 220,000 g/mol
- Unit of molecular weight: Dalton (Da)
= 1 달톤은 수소 원자량과 같음
3) 이황화결합 (Disulfide bond):
- Bond between thiol (sulfhydryl)
group of cystein
- Cystine: two linked cystein
residues by disulfide bond.
1. 단백질의 amino acid 순서는 유전자에 의해 결정된다.
- Insulin: 최초로 아미노산 순서가 결정된 단백질임 (Fredrick Sanger)
- 단백질 1차 구조 (Primary sequence of proteins): 단백질의 amino acid 서열
= 유전정보에 의해 결정됨

- 단백질의 1차 구조의 정보가 필요한 이유?
• 각 단백질의 작용기전을 규명
• 각 단백질의 3차 구조를 밝힘: 단백질의 생리학적 기능에 중요함
• 질병의 분자병리학적 원인 규명: 돌연변이를 규명
• 진화역사를 분자수준에서 연구할 수 있음
2. Polypeptide chain은 유연성이 있지만 입체형태는 제한됨.
- peptide bond의 중요한 특성:
= peptide bond은 평면구조이다.
peptide bond에 연결된 6개 원자는 한 평면 위에 존재한다.
Why??
= 펩타이드 결합은 2중 결합의 특성을 가짐
Length of C-N of peptide bond: 1.32 Å
cf) C-N: 1.49 Å , C=N; 1.27 Å
= peptide bond은 전하를 갖지 않음: 따라서 아미노산 중합체가 구형단백질을
형성할 수 있도록 함.
- Peptide 결합에서 작용기의 공간배치
= Peptide bond는 평면구조이며, 부분 이중결합임
= 분자의 작용기는 두 가지 공간배치가 가능함

cis:
= 단배질의 대부분의 펩타이드 결합은 trans 형임.
cis 형 공간배치는 구조적 방해 (Steric hindrance)가 일어남.
= Proline은 cis 형을 만들 수 있음
