CHROMOSOME MAPPING
Linkage : จํานวนยนี 2 ยีน หรือ กลุมของยีนที่อยูบนโครโมโซมเดียวกัน หรือ เชื่อมอยูดวยกันบนสวนของโครโมโซมเดียวกัน ( linked genes )
ปกติยีนที่อยูบนโครโมโซมตางแทงกันเมื่อมีการสรางเซลลสืบพันธุจะดําเนินตามกฎเมนเดลขอที่ 2

จํานวนทั้งหมด
จํานวนทั้งหมด : จํานวน parental types (PT) + recombinant types (RT)
% crossing over = 2 ( % recombination )
ดังนั้น % recombination มีคาสูงสุดเทากับ 50 % x-over = 100
ถา % recombination > 50 หมายความวา ยีน 2 ยีน ที่ทําการทดสอบนั้นไม linked กัน หรืออยูบนโครโมโซมตางกัน

ตัวอยางเชน ถาเกิด cross-over ระหวางตําแหนงของยีน A และ Bของยีโนไทป AB/ ab เทากับ 30 %
ดังนั้นเซลลสืบพันธุ RT คือ Ab และ aB เกิดขึ้น 15 %( Ab = aB = 7.5 % )
PT คือ AB และ ab เกิดขึ้น 85 %( AB = ab = 42.5 % )

ตัวอยาง ในกรณีทํา testcross ระหวาง Ab/ aB กับ ab/ ab
เกิดลูก Ab/ab 40 % aB/ab 40% AB/ab 10%
และ ab/ab 10 %
ยีโนไทป RT คือ AB/ab และ ab/ab = 10+10 = 20 %
% x-over = 2(20%) = 40%

แผนที่ยีนการทําแผนที่ยีนอาศัยหลัก 2 ขอ คือ
1. ตําแหนงของยีนบนโครโมโซมเรียงกันเปนเสนตรงในชวงความยาว หนึ่ง
2. ตําแหนงของยีนหาไดจากระยะทางของยีนขางเคียงทั้ง 2 ขาง
ระยะทางของยีน 1 map unit ( centimorgan) = 1 % recombination
ตัวอยาง ถาระยะทางระหวางยีน X และ Y เทากับ 12 หนวย สิ่งมีชีวิตที่มียีโนไทป XY/ xy เมื่อสรางเซลลสืบพันธุควรมีการสรางเซลลสืบพันธุ RT (Xy และ xY) อยางละ 6 %

การทดสอบระยะทางระหวางยีน 2 ตําแหนง
โดยวิธี Two-point Testcross
ทําโดยหา % Recombination จากลูก RT ที่เกิดขึ้น
ตัวอยางเชน การผสมทดสอบ coupling phase genotype AC / ac
- เกิดลูกที่มีฟโนไทปเดนของยีนทั้งสองตําแหนง 37%
- ฟโนไทปดอยของยีนทั้ง 2 ตําแหนง 37%
- ฟโนไทปเดนที่ตําแหนง A แตดอยที่ C 13% และ
- ฟโนไทปที่ดอยที่ตําแหนง A แตเดนที่ตําแหนง C 13%

เซลลสืบพันธุ RT ไดแก Ac และ aC
RT genotypes คือ Ac / ac และ aC / ac
ซึ่งมีจํานวนเทากับ 13% + 13% = 26 % = % Recombination
ดังนั้นระยะทางระหวาง A-C = 26 หนวย ( map unit )

การทดสอบระยะทางระหวางยีน 2 ตําแหนง
โดยใช Three-point Testcross
ปกติการหาระยะทางระหวางยีนทําโดยการตรวจดูลูกที่เกิดจาก single crossing-over เทานั้น
แตถายีนทั้ง 2 อยูหางกันจนสามารถเกิด double crossover ลูกที่เกิดจาก double x-over จะมีฟโนไทป เชน เดียวกับ
parental type ซึ่งไมสามารถจําแนกได
จึงตองมี marker gene อยูระหวางยีนทั้งสอง จึงเรียกวิธีนี้วา การทดสอบ 3 ตําแหนง ( ถายีน 2 ยีน อยูหางกันไมเกิน 5 หนวย จะไมเกิด double crossover )

ระยะทางระหวางยีน = % SCO + % DCO
ระยะทาง A-B = %SCO(A-B)+% DCO = 18+2 = 20%
= 20 หนวยระยะทาง B-C = %SCO(B-C)+% DCO = 8+2 = 10%
= 10 หนวยระยะทาง A-C = ระยะทาง A-B + ระยะทาง B-C
= 20+10 = 30 หนวย
ในกรณีถาทําเพียง Two-point Testcross ระหวาง A-C ลูก DCO จะเหมือน PT
% Recombination = % SCO A-B+ % SCO B-C
= 18 + 8 = 26%

ลําดับของยีน ( Gene Order )การเรียงลําดับของยีนก็ใชหลักการวา ตําแหนงของยีนเรียงกันเปน
เสนตรงแลวหาโอกาสที่เปนไปได
ตัวอยางเชน ถาระยะทางระหวาง A-B = 12 หนวย B-C= 7 หนวย A-C = 5 หนวย
โอกาสที่ 1 ถาให A อยูกลาง ดังนั้น
B 12 A 5 C
7โอกาสที่ 2 ถาให B อยูกลาง ดังนั้นA 12 B 7 C
5โอกาสที่ 3 ถาให C อยูกลาง ดังนั้น

จากทั้งโอกาส จะพบวาโอกาสที่ 3 เปนลําดับที่ถูกตอง

ลักษณะของ linked genes เปน repulsion phase มียีโนไทป Ab / aB

Chromosome painting for plants?
• Schubert, Fransz, Fuchs and de Jong (2001): no chromosome painting (CISS) in plants!
• Through repeat homogenization no chromosome specific paint by CISS (Heslop-Harrison et al.)
• Alternatively, “BAC-FISH painting” in plants is possible with pooled chromosomal DNA if:
DNA from small genome species (Arabidopsis)Painted to related genomes (Capsella, Arabis)
